    Strategies for Feeding Unweaned Dairy Beef Cattle to Improve Their Health

    In order to answer the question of whether nutritional interventions may help to reduce the incidence of respiratory disease in dairy beef calves at arrival, the present review is divided in three sections. In the first section, the nutrition of calves previous to the arrival from the origin farm to the final rearing farm is reviewed. In the second section, the possible consequences of this previous nutrition on gut health and immune status upon arrival to the rearing farm are described. The main consequences of previous nutrition and management that these unweaned calves suffer at arrival are the negative energy balance, the increased intestinal permeability, the oxidative stress, the anemia, and the recovery feed consumption. Finally, in the third section, some considerations to advance in future nutritional strategies are suggested, which are focused on the prevention of the negative consequences of previous nutrition and the recovery of the gut and immune status. Moreover, additional suggestions are formulated that will be also helpful to reduce the incidence of bovine respiratory disease (BRD) that are not directly linked to nutrition like having a control golden standard in the studies or designing risk categories in order to classify calves as suitable or not to be transported.info:eu-repo/semantics/publishedVersio

    Transformational design for food systems: cultural, social and technological challenges

    Due to climate changes, resources availability and evolving markets, the food system is developing towards an articulated and complex ecology, with fast transformations occurring in food production, preparation, delivery and disposal. In this context, innovation is needed not just to ideate solutions to deal with a fast-changing system but also to accompany the change adopting a systemic long-term approach. We reflect on the transformational potential of design in the food sector enabled by digital technologies, one of the current major drivers of change. We define two levels of changes implying digital technologies, those that radically change the food system and those enabling changes within a given system. These levels are exemplified with case studies documented in literature and with students’ projects showing how transformational design can help grasp the complexity of current problems, and question the current status quo by facilitating a dialogue among stakeholders to stimulate behaviour change without prescribing it. In this article we encourage a paradigm shift of design from craft activity to a holistic approach of systemic thinking where the designer assumes the role of promoter and facilitator of change. Reflections on challenges at cultural, social and technological levels are provided in the conclusion section

    Objetivos: La arteriografía cerebral y colocación de stent carotídeo se ha descrito que pueden ocasionar la aparición de cefalea, sin saber quiénes la desarrollan ni que características presenta. Material y método: Mediante estudio prospectivo observacional, se recogieron los datos de pacientes que fueron sometidos a arteriografía y/o stenting carotídeo ingresados en Hospital Clínico Lozano Blesa. Se estudiaron variables sociodemográficas, factores de riesgo cardiovascular, afectación carotídea y antecedentes de cefalea primaria. Se realizó una entrevista estructurada antes, después del procedimiento, tras 24 horas y al cabo de un mes, para registrar la aparición de cefalea. Resultados: se incluyeron un total de 25 pacientes con una media de edad de 67 años; un 92% (N=23) fueron varones y un 8 %(N=2) mujeres. El 21% (N=5) tenían antecedentes de cefalea, en el 60% (N=12) migraña con aura y 40% (N=8) cefalea tensional. La cefalea apareció en 4 pacientes (16%), siendo en el 75% (N=3) durante el procedimiento y en un caso en las primeras 6 horas. En un caso la duración fue menor de 10 minutos, y en otro entre 10-120 minutos. Las principales localizaciones fueron en igual porcentaje la región fronto-temporal y la facial. Descrita como opresiva en un 75% (N=3), con intensidad leve-moderada (50%) y el 50% precisaron analgesia. Se vió una relación estadísticamente significativa entre la aparición de la cefalea y afectación de la arteria carótida interna izquierda (p=0.059).     Evaluation of potential biomarkers to determine adequate colostrum provision in male dairy-beef calves upon arrival at the rearing facility beyond 14 days of age

    Colostrum consumption is crucial for passive immunization and development of the newborn calf. However, the incidence on failed transfer of passive immunity in male calves destined to dairy-beef production remains high to date. In addition, the lack of an automated procedure to validate the immunization status upon arrival at rearing facilities in calves beyond 14 d of age impedes the identification of failed transfer of passive immunity, and therefore, of those calves at high risk of suffering diseases. For this study, 82 newborn male Holstein calves (43.3 ± 0.86 kg of body weight; mean ± standard error) from a commercial dairy farm were used to investigate potential serum biomarkers of colostrum provision. The potential biomarkers selected were IgG, IgG1, cholesterol, alkaline phosphatase, gamma-glutamyl transferase (GGT), and total protein (TP). Treatments were as follows: high-colostrum (HC; n = 49), in which calves received 4 L of colostrum within the first 2 h after birth and 2 L of colostrum in the next 3 feedings within the first 24 h after birth, for a total of 10 L of colostrum; and low-colostrum (LC; n = 33), in which calves received only 2 L of colostrum within the first 2 h after birth. After colostrum consumption, calves were allocated to individual hutches and fed 2 L of milk replacer twice daily at a concentration of 125 g/L as fed. Starter feed and water were offered ad libitum. At approximately 14 d of age (14.2 ± 0.81 d of age; mean ± standard error) calves were transported 2.5 h to a research unit at IRTA (Torre Marimon, Spain) simulating the arrival to a rearing facility. Blood samples were collected before feeding at birth, 48 h after birth, and at arrival to the rearing facility. Results on the serum concentrations of the potential biomarkers at arrival to the rearing facility showed that IgG, IgG1, GGT, and TP were greater for the HC calves compared with the LC calves. Serum concentrations of cholesterol and alkaline phosphatase did not show differences between treatment groups. Additionally, body weight losses from birth until arrival to the rearing facility were greater for the LC treatment compared with the HC. Because of their low cost, quickness, and ease of measurement, GGT and TP were good indicators of colostrum intake in calves arriving at rearing facilities beyond 14 d of age.info:eu-repo/semantics/publishedVersio

    Drinking water chlorination in dairy beef fattening bulls: water quality, potential hazards, apparent total tract digestibility, and growth performance

    The first study aimed to evaluate the effect of drinking water disinfection (chlorination: NaClO 15%) and conditioning (acidification: H3PO4 diluted 1:5 in water) on water quality, water and feed consumption, apparent total tract digestibility, and its potential hazardous effects on Holstein bulls fed high-concentrate diets. Twenty-four animals (221 ± 20.9 kg of BW, and 184 ± 9.9 days of age) were individually assigned to one of four treatments according to a 2 × 2 factorial arrangement: conditioning (with or without acidification) and disinfection (with or without chlorination). The entire study lasted 210 days. Physicochemical and microbiological water quality, water and feed consumption, haematological and biochemical blood parameters, and apparent total tract digestibility were measured; data were analysed via a mixed-effects model. Chlorination and acidification increased (P = 0.02) free residual chlorine in water, and chlorination reduced (P = 0.01) total coliform and Clostridium perfringens counts in water. Treatment did not affect water consumption, total DM intake, or blood parameters. At the beginning of the study, NDF digestibility decreased (P = 0.04) with acidification, however, this was restored at the end of the study. The second study evaluated the potential benefit of drinking water chlorination and acidification on the performance of crossbred Holstein bulls fed high-concentrate diets under commercial conditions. Ninety-six animals (322 ± 35.0 kg of BW, and 220 ± 14.2 days of age) were allocated into six pens assigned to one of the two treatments: untreated drinking water or drinking water treated with chlorination and acidification for a total of 112 days. Physicochemical and microbiological water quality, water and concentrate consumption, eating behaviour, growth performance, and carcass quality were analysed via a mixed-effects model. Water conditioning and disinfection increased (P = 0.01) free residual chlorine concentration and reduced (P = 0.04) total coliform count in water. Although water consumption and eating behaviour were similar between treatments, water conditioning and disinfection increased average daily weight gain (P = 0.03), BW before slaughter (P = 0.01), and hot carcass weight (P = 0.01). In conclusion, drinking water chlorination and acidification in fattening dairy beef bulls is recommended as it improves growth performance without any detrimental side effects on health or nutrient digestibility.info:eu-repo/semantics/publishedVersio

    Effect of transport and rest stop duration on the welfare of conditioned cattle transported by road

    The effects of providing rest on physiological and behavioural indicators of welfare of cattle being transported by road has not been well studied in North America. New revisions to Canada’s Health of Animals Regulations Part XII: Transportation of Animals indicate un-weaned and weaned calves can be transported a maximum of 12 and 36 h, respectively, before an 8 h rest is required. Therefore, the aim of this study was to assess the effects of rest duration, after 12 and 36 h of transport, on physiological and behavioural indicators of welfare in 7–8 mo-old beef calves. Three hundred and twenty conditioned calves (258 ± 23.9 kg BW) were randomly assigned to a 2 × 4 factorial design where the main factors included transport duration: 12 h (12; n = 160) and 36 h (36; n = 160) and rest stop duration: 0 h (R0; n = 80), 4 h (R4; n = 80), 8 h (R8; n = 80) and 12 h (R12; n = 80). After the resting period, animals were transported for an additional 4 h. Blood and hair samples were taken from 12 animals per treatment prior to and after the first and the 4 h transport; and then 7 h, 2 d and 28 d after the 4 h transport. The concentrations of haptoglobin, creatine kinase, non-esterified fatty acids (NEFA), lactate, and serum and hair cortisol were determined. Standing and lying behaviour was assessed for 14 d after transport, while feeding behaviour of calves in one pen per treatment were assessed for 28 d after transportation using an electronic feed bunk monitoring system. Body weight (BW), average daily gain (ADG) and shrink (%) was assessed for all calves. The data was modeled using generalized linear mixed methods (SAS PROC GLIMMIX), where transport and time (nested in rest) were considered fixed effects and animal and pen were considered random effects. Statistically significant (p < 0.05) effects of transport were observed on BW and shrink, where 36 h-transported calves had lower (p < 0.01) BW and greater (p < 0.01) shrink than 12 h-transported calves. A transport × time (nested in rest) interaction (p < 0.01) was observed for lying percentage where, 36-R8 calves had greater (p < 0.01) lying percentage than 12-R8 calves on d 1 after transportation. The area under the curve (AUC) for NEFA was greater (p < 0.01) for 36-R0 calves than 12-R0, 36-R4, and 36-R8 calves, and greater (p < 0.01) in 36-R12 calves than 12-R12 calves. Haptoglobin AUC was greater (p = 0.05) in 36-R12 than 12-R12 calves. Overall, physiological indicators of reduced welfare were greater in calves transported for 36 than 12 h, while no clear differences were observed between rest stop groups with the exception of NEFA. Based on these results, conditioned calves benefit from shorter transport durations but there was no clear evidence that calves rested 4, 8, and 12 h following transportation experienced reduced transport related stress compared to those that were not rested (0h).info:eu-repo/semantics/publishedVersio

    Synthesis, Characterization and Evaluation of the Cytotoxicity of Ni-Doped Zn(Se,S) Quantum Dots

    Quantum dots (QDs) are semiconductor nanocrystals with desirable optical properties for biological applications, such as bioimaging and drug delivery. However, the potential toxicity of these nanostructures in biological systems limits their application. The present work is focused on the synthesis, characterization, and evaluation of the toxicity of water-stable Ni-doped Zn(Se,S) QDs. Also, the study of nondoped nanostructures was included for comparison purposes. Ni-doped nanostructures were produced from zinc chloride and selenide aqueous solutions in presence of 3-mercaptopropionic acid and Ni molar concentration of 0.001 M. In order to evaluate the potential cytoxicity of these doped nanostructures, human pancreatic carcinoma cells (PANC-1) were used as model. The cell viability was monitored in presence of Ni-doped Zn(Se,S) QDs at concentrations ranging from 0 μg/mL to 500 μg/mL and light excited Ni-doped Zn(Se,S) nanostructures were evaluated at 50 μg/mL. Results suggested that Ni-doped Zn(Se,S) nanostructures were completely safe to PANC-1 when concentrations from 0 μg/mL to 500 μg/mL were used, whereas non-doped nanostructures evidenced toxicity at concentrations higher than 200 μg/mL. Also, Ni-doped Zn(Se,S) QDs under light excitation do not evidence toxicity to PANC-1. These findings suggest strongly that Zn(Se,S) nanostructures doped with nickel could be used in a safe manner in light-driving biological applications and drug delivery

    Effect of rest, post-rest transport duration, and conditioning on performance, behavioural, and physiological welfare indicators of beef calves

    The aim of this study was to assess the effects of conditioning, rest, and post-rest transport duration on welfare indicators of 6–7 mo old beef calves following a 20-h transport. Three hundred and twenty-eight weaned calves (237 ± 29.7 kg of BW) were randomly assigned to a 2 × 2 × 2 nested factorial design: conditioning, conditioned (C) or non-conditioned (N); rest, 0 (R0) or 8 (R8) h, and post-rest transport, 4 (T4) or 15 (T15) h. Calves were sampled before (LO1) and after (UN1) the initial 20-h journey, before (LO2) and after (UN2) the additional 4 or 15-h journey, and at 1, 2, 3, 5, 14, and 28 d after UN2. Data was analyzed using the GLIMMIX procedure of SAS. Fixed effects included conditioning, transport, and time nested within rest period, while random effects included animal and pen. Greater shrink (p < 0.01) was observed in C than N calves after the initial 20-h transport. During the first week after transportation, the mean ADG of N calves was greater than C calves (p < 0.01). From d 14 to d 28, however, the mean ADG of C calves was greater than N calves (p < 0.01). Flight speed, cortisol and L-lactate concentrations were greater (p ≤ 0.05) in C than N calves between LO1 and d 5, while greater (p ≤ 0.02) non-esterified fatty acids, creatine kinase, serum amyloid-A, and haptoglobin concentrations were observed in N than C calves between LO1 and d 3. The R8-T4 calves had greater (p < 0.01) ADG than R8-T15 calves between LO1 and d 5. The R0-T4 calves had greater L-lactate concentrations than R0-T15 and R8-T4 calves (both p = 0.02) on d 1. The R0 calves had greater (p < 0.01) ADG than R8 calves between 14 and 28 d. This study suggests that C calves are better fit for transport than N calves as evidenced by behavioural and physiological parameters.     Characterizing the diagnostic sensitivity and specificity of pain biomarkers in cattle using receiver operating characteristic curves

    Biomarkers are used to assess pain and analgesic drug efficacy in livestock. However, often the diagnostic sensitivity and specificity of these biomarkers for different painful conditions over time have not been described. Receiver operating characteristic (ROC) curves are graphical plots that illustrate the diagnostic ability of a test as its discrimination threshold is varied. The objective of this analysis was to use area under the curve (AUC) values derived from ROC analysis to characterize the predictive value of potential pain biomarkers at specific time points following a painful stimulus. The biomarkers included in the analysis were plasma cortisol, salivary cortisol, hair cortisol, infrared thermography (IRT), mechanical nociceptive threshold (MNT), substance P, kinematic gait analysis, and a visual analog scale for pain. A total of 7,992 biomarker outcomes collected from 7 pain studies involving pain associated with castration, dehorning, lameness, and abdominal surgery were included in the analysis. Each study consisted of 3 treatments: uncontrolled pain (tissue damage), no pain (handled controls), and analgesic use (tissue damage, administered a nonsteroidal anti-inflammatory drug). Results comparing analgesic effects to uncontrolled pain consistently yielded AUC values >0.7 (95% confidence interval: 0.40 to 0.99) for plasma cortisol (time points: 1.5, 2, 3, 4, 6, and 8 h), hair cortisol (time point: 62 d), and IRT (time point: 72 h). Results comparing analgesic effects to uncontrolled pain consistently yielded AUC values <0.7 (95% confidence interval: 0.28 to 0.90) for salivary cortisol (6, 13, 20, 34, 48, and 62 d); MNT (6, 25, and 49 h); substance P (1, 2, 3, 4, 6, 8, 12, 18, 24, 48, 72, 96, 120, 144, 312, 480, 816, 1,152, and 1,488 h); kinematic gait analysis including area (8, 16, 48, 72, 96, and 120 h), force (8, 16, 24, 48, 72, 96, and 120 h), and pressure (8, 16, 24, 48, 72, 96, and 120 h); and a visual analog scale for pain (1, 2, 3, 4, 5, and 6 d). These results indicate that ROC analysis can be used to characterize the predictive value of pain biomarkers and provide new knowledge on the diagnostic accuracy of pain biomarkers within this data set. This analysis, using data from 7 studies, was a preliminary approach to identify biomarkers and collection time points that could inform additional analytical approaches or meta-analyses with larger sample sizes, which are needed to further validate these hypotheses and conclusions.info:eu-repo/semantics/publishedVersio
