    Robust estimation of bacterial cell count from optical density

    Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data

    지금까지 본고에서는 한국과 터키 두 나라에서 각각 구비 전승되어 오던 문학 가운데 “길 위에 선 자”들이 등장하는 설화를 중심으로 성공의 요인과 길에 대한 양국의 생각을 확인하였다. 물론 두 설화의 비교가 두 나라의 가치관이나 민족성을 논의하기에 부족함은 사실이지만, 최소한 두 나라의 문학적 유사성과 그 차이에 대한 기반적 논의를 한 것에 의의가 있을 것이라 생각된다. 이후 두 나라의 설화들을 차례로 비교 분석함으로써 문학적 측면에서 바라본 한국과 터키의 모습을 논의해나가고자 한다

    본 연구는 터키 국립 이스탄불대학교에서 한국어문학을 전공하는 4학년재학생을 대상으로 진행항 “한국문학 번역교육 실습”교육 사례를 중심으로 진행되었다. 이상의 연구를 위해 먼저 이스탄불대학교 한국어문학과에 대해 살펴보았을 뿐 아니라 “한국문학 번역교육 실습”의 교육목표와 내용 그리고 자료 선정 이유 및 해당 과목의 강의계획서를 확인하였다. 나아가 해당 과목의 15주간 교육과정 내용과 실제 교육안을 바탕으로 진행된 교육 사례를 정밀하게 살펴보았다. 마지막으로 “한국문학 번역교육 실습”에서 나타난 결과를 통해 본 교육과정의 의의와 문제를 확인함으로써 새로운 교육 방안 마련을 위한 토대로 삼았을 뿐 아니라 해외에서 앞으로 진행될 한국문학 번역교육의 발전방안에 관해 제언해보고자 하였다. 이에 본 연구는 해외 다른 나라에서 문학번역 교육을 진행하는 교수자에게 하나의 교육 사례를 제시하였다는 점과 문하번역 교육의 발전을 위한 방안을 권고하였다는 점에서 그 의의를 찾을 수 있다

    This study was conducted to learn how marriage immigrant womencan improve their Korean language skills while performing their roleas parents. For this purpose, it was intended to use elementary schoolKorean textbooks as textbooks for Korean education targeting marriedimmigrant women. Of course, there are many Korean textbooks outnow, and ones that can be used by married immigrant women arebeing distributed sufficiently. However, the focus of this paper is notonly to improve the Korean language skills of married immigrantwomen but also to establish a role as a parent and restoreself-esteem. A minor but serious problem in multicultural families liesin ‘difference’. Children experience it from their peers at school, andmarried immigrant women are hurt by their children's disregard fordifferences. Therefore, in order to restore the self-esteem of childrenof multicultural families and married immigrant women, they shouldbe able to actively participate in their children's school problems. Inthis regard, if married immigrant women use Korean languageeducation as a textbook for their children, it is thought that they willbe able to recover not only Korean language skills but also theirauthority as mothers.Journal of Korean Studies in Central Eurasi

    A Study on Korean Language Education Plan for Occupational Purpose, Focused on Writing of Official Documents

    This study aimed to research Korean language education for occupational purpose in other words “Business Korean” by identifying education methods, contents and their results of lesson through the lesson examples and accomplished during the spring semester of 2019 aimed for Korean Language and Literature third grade learners at Istanbul University. As the result, we were able to provide a basic understanding of the information request and delivery and content constitution of the documents, which are important factors in Korean language education for occupational purposes. Furthermore, it improved the sense of repulsion and diffuculty about new lesson and confirmed its importance in that it makes learners to realize how to create necessary contents and ideas for their business practice by themselves. If there is a suggest about Korean language education plan for occupational purposes based on more examples, we can discuss it by dividing it into subjects related to learners, teacher and lesson. When Korean language education is going to put into effect for occupational purposes, the number of learnes participate in should be suitable for learners and the level of Korean language should be advanced. Also, teacher should possess professionalism, and the textbook, period or time and course of study should be determined under the big goal of business dialogues and essays. However, according to the result of researcher’s Korean language education for occupational purposes lesson that progressed for one semester, progressing this lesson for one year and 15 weeks for one semester for one and a half or two hours for 15 to 20 students that aimed for advanced Korean learners, by dividing it business dialogues and essays will produce its fruits with the effort of the teacher

    A Study on the Development plan of Tasks for the Evaluatıon of performance level In Korean Speakıng educatIon

    This study focused on the Korean speaking education conducted for the first-year students of the Department of Korean Language and Literature, Istanbul University, Turkey in the spring semester of 2020. During Korean speaking education, attention was paid specifically to success evaluation, and various tasks were presented to evaluate success to prepare a plan for achievement evaluation.To this end, in Chapter 2, we tried to take a close look at the overall course of the 'Korean Speaking 4' course, which was a part od freshman syllabus of Korean Language and Literature at Istanbul University in spring 2020. First, by looking at the course plan; the goals and contents of education, methods, evaluations, objects, and materials were identified. In addition, by organizing the contents of syllabus for 15 weeks, it was possible to accurately examine the roadmap of the course. In addition, by examining the 4th week’s lecture plan among the 15-week training syllabus, it was possible at a glance to confirm the method of the course. Next, we reviewed 4 tasks in the mid-term evaluation in the 8th week and 4 tasks in the final evaluation in the 15th week. First, the learned grammar and speaking task was used as a task for evaluating competence achievement, and secondly, it was found that the role play and interview techniques were used as a task to evaluate linguistic abilities. Thirdly, discussion and debate exercises were used as a task to evaluate the achievement of communicative competence, and fourth, it was found that presenting personal topics was used as a task to evaluate achievement of strategic competency. Lastly, the scoring table prepared to evaluate the achievement of the intermediate evaluation and final evaluation tasks were checked. The items in the scoring table are commonly composed of 'task performance, accuracy, fluency, speaking attitude, pronunciation and intonation, and remarks'. In addition, in the scoring table of communicative competence ability evaluation, the flowness of dialogues and the content validity evaluations were added in the scoring table of communicative competence evaluation since it was a group task. In the strategic competence evaluation scoring table, it was confirmed that the item of content validity was added as a scale since it was an individual presentation.Based on the contents of chapter 2, In chapter 3, we aimed to suggest a potential plan to evaluate Korean speaking abilities. The first plan was to have a great variety of new challenges rather than classifying the types of tasks to evaluate speaking skills. The second goal was to add fun elements for complex subjects so that the learner can enjoy the tas keven he/she finds it difficult. The third goal was to create an overall evaluation of all subjets together. The study above is meaningful in the form that it provides an understanding of how to evaluate the success of Korean Speaking students. Also, the value of the study is meaningful because it depends on an actual syllabus of an instructor/professor.     본고에서는 다년간의 한국에서의 생활로 일정 수준의 말하기와 듣기가 가능한, 그리고 초등학교에 자녀가 취학할 예정이거나 혹은 이미 취학한 학부모가 된 이주여성을 위한 한국어교육 프로그램을 개발하고자 한다. 교육에 주 자료는 한국의 초등학교 국어교과서이며 차시는 30차시, 각 차시별 4시간씩, 6개월 간 진행할 수 있는 프로그램으로 구성할 것이다. 위의 프로그램은 한국어 교사나 다문화 지도사가 주 선행자가 진행할 것이며, 복지관이나 다문화가정지원센터, 도서관, 학교 등에서도 진행될 수 있을 것이다

    The purpose of this research is to examine the meaning of the story of "The Debts have been Repaid at the Hell", which is known as a story characterized simple witticism or fraud. We focus on the analysis of the loser who paid back money that was lost, and we thoroughly examine why he failed to accept the money regardless of whether he believed in the hollow afterworld. Therefore, the major conflict of the narrative is defined as a defeat of the invisible principle of life's visual reality logic, which is the principle of behavior demonstrated by an individual who is known as a person who eats money. As a result, the person who receives a good money has devastated the environment of his inner life by pursuing a visible reality logic. And the person who fails to pay money, brings a person to his life through 'the story of afterlife'. It is understood that he advises him to look back. In this respect, the story of "the debts have been repaid at the hell" reflect human limitations that are easy to overlook and invisible values ​​that are biased by realistic and reasonable values. We can understand that this story led to a small failure, a small loss, and a way of enriching the back of life, and life as a whole. This narrative goes beyond the meaning of simple trivial competition and demonstrates a precious understanding of the principles of human life. Therefore this research has significance because it reaffirms the story
