    Non-acoustic detection systems can be used in combination with Sonar systems to determine the presence of an underwater threat, such as terrorist divers. The goal of the CAIMAN (Coastal Anti Intruders MAgnetometers Network) joint experiment (Italian Navy, NATO Undersea Research Centre and INGV Marine Geophysics) is the application of High Definition Geophysics Magnetic techniques in a port protection scenario, where conventional measurements of very low magnetic sources, like intruder swimmers, are strongly disturbed by ambient, natural and artificial, background noise and other time-variant magnetic anomalies. Two tri-axial fluxgate magnetometers were deployed on the sea bottom and connected to a shore side measurement station. A team of navy divers, wearing both COTS and EOD equipment, performed some coastal approach runs on each magnetometer alternatively. Magnetic signature data were logged and post processed using MATLAB®. Results demonstrated the effectiveness of high definition time reduction techniques using a self-referred integrated array design

    High-intensity exercise training induces morphological and biochemical changes in skeletal muscle

    Skeletal muscle shows an elevated plasticity and can adapt its metabolic and contractile properties in response to a variety of stimuli such as physical exercise. This implies a series of biochemical and morphological changes in the recruited muscle, in order to produce the more appropriate functional response dependent on the specific stimulation. To determine the effective role of physical exercise in the muscle plasticity, in the present study we investigated the effect of two different exercise protocols on fiber composition and metabolism of two specific muscles of mice: the quadriceps -a fast-twitch muscle- and the gastrocnemius -a typical slow-twitch muscle. Mice were run daily on a motorized treadmill for 8 weeks, at a velocity corresponding to 60% (low-intensity exercise) or 90% (high-intensity exercise) of the maximal running velocity previously determined by an incremental exercise test. We found that at the end of training the body weight was significantly increased in highintensity exercise mice (18.2 ± 1.4 %) compared to low-intensity exercise (8.7 ± 0.6 %) and control (12.7 ± 0.5 %) groups, and it was lesser in low-intensity exercise mice compared to controls. In contrast, the food intake of both exercise training mice was greater compared to control group. Whereas low-intensity exercise mice, despite consumed significantly more food compared to control mice, increased the weight lesser, the weight increase of high-intensity exercise mice, that consumed significantly more food compared to other experimental groups, was significantly greater. These effects were accompanied by a progressive reduction in blood lactate levels at the end of training in both the exercised mice compared with controls; in particular, blood lactate levels after highintensity exercise were significantly lower than those measured in low-intensity exercise mice. Moreover, in the present study we demonstrated that high-intensity exercise training produced a significant increase in the expression of mitochondrial complex enzymes (significant for the enzymes corresponding to the Complex IV, II and I of mitochondrial chain) both in gastrocnemius and quadriceps muscle, compared with controls. These changes were associated with an increase in the amount of slow fibers in both these muscle of high-intensity exercise mice. No changing in the expression of mitochondrial enzymes and in the percentage of slow fibers were found in low-intensity exercise mice

    Intracellular distribution of Tankyrases as detected by multicolor immunofluorescence techniques

    Poly(ADP-ribose) polymerases are a family of enzymes that catalyze the conversion of NAD+ into ADP-ribose. Among them, Tankyrases have been found to bind to centrosome, mitotic spindle and microsome proteins, in the cytoplasm, and to telomeres in the nucleus, where they play a relevant role in telomere metabolism. However, their precise intracellular localization during interphase has not been so far fully elucidated. We investigated this aspect in situ by double immunofluorescence experiments using antibodies recognizing Tankyrases 1–2 or other proteins residing in specific organelles (Golgi apparatus, mitochondria, lysosomes, endoplasmic reticulum). We used HeLa cells as a model system in vitro, before and after treatment with either actinomycin D or etoposide, to also investigate the possible relocation of Tankyrases during apoptosis. We observed that Tankyrases are distributed both in the nucleus and in the cytoplasm; in this latter compartment, they were found to colocate with the Golgi apparatus but never with the mitochondria; a pool of Tankyrases also colocates with the endoplasmic reticulum and lysosomes. Interestingly, in cells with clear signs of apoptosis, Tankyrases were detectable in the cytoplasmic blebs: this suggests that they are not massively cleaved during apoptosis and persist in the largely heterogeneous apoptotic remnants which are known to contain components of cytoplasmic and nuclear origin

    A Preliminary Study on SVM based Analysis of Underwater Magnetic Signals for Port Protection

    People who attend to the problem of underwater port protection usually use sonar based systems. Recently it has been shown that integrating a sonar system with an auxiliary array of magnetic sensors can improve the effectiveness of the intruder detection system. One of the major issues that arise from the integrated magnetic and acoustic system is the interpretation of the magnetic signals coming from the sensors. In this paper a machine learning approach is proposed for the detection of divers or, in general, of underwater magnetic sources. The research proposed here, by means of a windowing of the signals, uses Support Vector Machines for classification, as tool for the detection problem. Empirical results show the effectiveness of the method

    A new approach for time-space wear modeling applied to machining tool wear

    A general methodology is proposed and demonstrated for modeling the progression of crater wear on cutting tools used for steel machining. Dimensional analysis and Finite Element Analysis of metal cutting are the foundations of this new approach. The dimensional analysis resulted in a differential equation which describes wear rate as a function of dimensionless variables. Numerical models of cutting allow one to estimate local variables associated with wear at tool-chip contact. Orthogonal cutting simulations were carried out with commercial code DEFORM - 2D Ver 11.0. AISI 1018 steel was used as the workpiece material for an uncoated carbide cutting tool.     Component-aware Orchestration of Cloud-based Enterprise Applications, from TOSCA to Docker and Kubernetes

    Enterprise IT is currently facing the challenge of coordinating the management of complex, multi-component applications across heterogeneous cloud platforms. Containers and container orchestrators provide a valuable solution to deploy multi-component applications over cloud platforms, by coupling the lifecycle of each application component to that of its hosting container. We hereby propose a solution for going beyond such a coupling, based on the OASIS standard TOSCA and on Docker. We indeed propose a novel approach for deploying multi-component applications on top of existing container orchestrators, which allows to manage each component independently from the container used to run it. We also present prototype tools implementing our approach, and we show how we effectively exploited them to carry out a concrete case study

    Harbour Sea-floor Clearance: “HD” High Definition Magnetic Survey Performance

    Seafloor clearance methods based on acoustic, direct-inspection, and single-sensor magnetic approaches suffer from limitations in controlling the target-sensor distance, and may prove ineffective when the small size or the dangerous nature of targets requires high accuracy in localization. Moreover, random magnetic variations over time bring about spatial decorrelation phenomena, and hinder the application of double-sensor methods in noisy harbour environments. The new High Definition (HD) magnetic survey protocol tackles the measurement-distance problem in two ways: first, by varying the sensor depth dynamically, and secondly by backprojecting the measured field according to seafloor data and vertical incremental factors associated with the bandwidth characteristics of targets. The method to make up for timeinduced loss in spatial localization ability exploits the local behaviour of a coherence function, which correlates local observations to a set of spatially-stabilized reference stations. The consequent normalization of measured magnetic signals allows one to assign the monitored areas with a specific level of confidence in the detection results, ranging from 100% (certainty) to 0% (random events). The principles of HD detection have been fully applied in the seafloor clearance of the firing test site located south of Cape Teulada (Sardinia, Italy), where very weak signal sources such as cartridge cases, mines, and small objects down to 1 Kg mass values (lobster pots) have been successfully localized, even when covered by extensive colonies of Posidonia

    Effect of platelet lysate on human cells involved in different phases of wound healing

    Background Platelets are rich in mediators able to positively affect cell activity in wound healing. Aim of this study was to characterize the effect of different concentrations of human pooled allogeneic platelet lysate on human cells involved in the different phases of wound healing (inflammatory phase, angiogenesis, extracellular matrix secretion and epithelialization). Methodology/Principal Findings Platelet lysate effect was studied on endothelial cells, monocytes, fibroblasts and keratinocytes, in terms of viability and proliferation, migration, angiogenesis, tissue repair pathway activation (ERK1/2) and inflammatory response evaluation (NFÎşB). Results were compared both with basal medium and with a positive control containing serum and growth factors. Platelet lysate induced viability and proliferation at the highest concentrations tested (10% and 20% v/v). Whereas both platelet lysate concentrations increased cell migration, only 20% platelet lysate was able to significantly promote angiogenic activity (p<0.05 vs. control), comparably to the positive control. Both platelet lysate concentrations activated important inflammatory pathways such as ERK1/2 and NFÎşB with the same early kinetics, whereas the effect was different for later time-points. Conclusion/Significance These data suggest the possibility of using allogeneic platelet lysate as both an alternative to growth factors commonly used for cell culture and as a tool for clinical regenerative application for wound healing

    High-intensity exercise training induces morphological and biochemical changes in skeletal muscles

    IN THE PRESENT STUDY WE INVESTIGATED THE EFFECT OF TWO DIFFERENT EXERCISE PROTOCOLS ON FIBRE COMPOSITION AND METABOLISM OF TWO SPECIFIC MUSCLES OF MICE: the quadriceps and the gastrocnemius. Mice were run daily on a motorized treadmill, at a velocity corresponding to 60% or 90% of the maximal running velocity. Blood lactate and body weight were measured during exercise training. We found that at the end of training the body weight significantly increased in high-intensity exercise mice compared to the control group (P=0.0268), whereas it decreased in low-intensity exercise mice compared to controls (P=0.30). In contrast, the food intake was greater in both trained mice compared to controls (P < 0.0001 and P < 0.0001 for low-intensity and high-intensity exercise mice, respectively). These effects were accompanied by a progressive reduction in blood lactate levels at the end of training in both the exercised mice compared with controls (P=0.03 and P < 0.0001 for low-intensity and high-intensity exercise mice, respectively); in particular, blood lactate levels after high-intensity exercise were significantly lower than those measured in low-intensity exercise mice (P=0.0044). Immunoblotting analysis demonstrated that high-intensity exercise training produced a significant increase in the expression of mitochondrial enzymes contained within gastrocnemius and quadriceps muscles. These changes were associated with an increase in the amount of slow fibres in both these muscles of high-intensity exercise mice, as revealed by the counts of slow fibres stained with specific antibodies (P < 0.0001 for the gastrocnemius; P=0.0002 for the quadriceps). Our results demonstrate that high-intensity exercise, in addition to metabolic changes consisting of a decrease in blood lactate and body weight, induces an increase in the mitochondrial enzymes and slow fibres in different skeletal muscles of mice, which indicates an exercise-induced increase in the aerobic metabolism
