    Metallurgical and statistical approaches to the study of cast iron street furniture

    The evolution of microstructure in relation to dating and nationality of origin was investigated in twenty-four cast iron objects of street furniture produced between XIX and XX centuries in United Kingdom, France, and Italy. Chemical composition of the metalworks was evaluated by glow-discharge optical emission spectrometry. Fragments from the cast irons were analyzed by optical microscopy and scanning electron microscopy with energy-dispersive spectroscopy. Form, distribution, and size of graphite were evaluated in the microstructure according to standard EN ISO 945-1:2008. An image analysis software was employed to quantify the area fraction of graphite in the matrix, major axis, and shape factor of graphite lamellae, area fraction of manganese sulfides (ψS), area fraction of steadite, and number of eutectic cells per area unit. All data were grouped and linear discrimination analysis (LDA) was applied to assess the group assignment and the probability of correct classification for each metalwork. The results showed that the microstructural features were compatible with those of cast irons produced in the XIX and XX centuries. Values of ψS also suggested re-melting of cast irons, associated with recycling of cast iron and/or steel scraps. The high values of steadite found in the metalworks are probably due to the excellent castability required for complex shape castings in these centuries.     HHV-6A infection and systemic sclerosis: Clues of a possible association

    Systemic sclerosis (SSc) is an autoimmune disease characterized by vasculopathy, excessive extracellular matrix deposition, and fibrosis of the skin and internal organs. Several infectious agents, including human herpesvirus-6 (HHV-6), have been suggested as possible triggering factors, but a direct association is still missing. We characterized 26 SSc patients for the presence of HHV-6 in tissues and blood, the anti-HHV-6 response, HLA-G plasma levels, and KIR typing. Given the prominent role of endothelial cells (EC) in SSc pathogenesis, along with HHV-6 tropism for EC, we also investigated the expression of pro-fibrosis factors in HHV-6 infected EC. Results showed the presence of HHV-6A in skin biopsies, and an increased virus load was associated with disease severity and poor natural killer (NK) response against the virus, particularly in subjects exhibiting a KIR2 phenotype. HLA-G plasma levels were significantly higher in HHV- 6A/B-KIR2 positive SSc patients and in vitro HHV-6A infection-induced pro-fibrosis factors expression in EC, supporting its role in the development of the fibrosing process. Our data suggest an association between virus infection/reactivation and disease, opening the way to future studies to understand the mechanisms by which HHV-6A might contribute to the multifactorial pathogenesis of SSc

    Modulation of micrornome by human cytomegalovirus and human herpesvirus 6 infection in human dermal fibroblasts: Possible significance in the induction of fibrosis in systemic sclerosis

    Human cytomegalovirus (HCMV) and Human herpesvirus 6 (HHV‐6) have been report-edly suggested as triggers of the onset and/or progression of systemic sclerosis (SSc), a severe autoimmune disorder characterized by multi‐organ fibrosis. The etiology and pathogenesis of SSc are still largely unknown but virological and immunological observations support a role for these beta-herpesviruses, and we recently observed a direct impact of HCMV and HHV‐6 infection on the expression of cell factors associated with fibrosis at the cell level. Since miRNA expression has been found profoundly deregulated at the tissue level, here we aimed to investigate the impact on cell microRNome (miRNome) of HCMV and HHV‐6 infection in in vitro infected primary human dermal fibroblasts, which represent one of the main SSc target cells. The analysis, performed by Taq-man arrays detecting and quantifying 754 microRNAs (miRNAs), showed that both herpesviruses significantly modulated miRNA expression in infected cells, with evident early and late effects and deep modulation (>10 fold) of >40 miRNAs at each time post infection, including those previously recognized for their key function in fibrosis. The correlation between these in vitro results with in vivo observations is strongly suggestive of a role of HCMV and/or HHV‐6 in the multistep patho-genesis of fibrosis in SSc and in the induction of fibrosis‐signaling pathways finally leading to tissue fibrosis. The identification of specific miRNAs may open the way to their use as biomarkers for SSc diagnosis, assessment of disease progression and possible antifibrotic therapies

    Introduction of Probiotic-Based Sanitation in the Emergency Ward of a Children's Hospital During the COVID-19 Pandemic

    Background: Antimicrobial resistance (AMR) represents a major threat to public health, especially in the hospital environment, and the massive use of disinfectants to prevent COVID-19 transmission might intensify this risk, possibly leading to future AMR pandemics. However, the control of microbial contamination is crucial in hospitals, since hospital microbiomes can cause healthcare-associated infections (HAIs), which are particularly frequent and severe in pediatric wards due to children having high susceptibility. Aim: We have previously reported that probiotic-based sanitation (PCHS) could stably decrease pathogens and their AMR in the hospital environment, reduce associated HAIs in adult hospitals, and inactivate enveloped viruses. Here, we aimed to test the effect of PCHS in the emergency room (ER) of a children's hospital during the COVID-19 pandemic. Methods: Conventional chemical disinfection was replaced by PCHS for 2 months during routine ER sanitation; the level of environmental bioburden was characterized before and at 2, 4, and 9 weeks after the introduction of PCHS. Microbial contamination was monitored simultaneously by conventional culture-based CFU count and molecular assays, including 16S rRNA NGS for bacteriome characterization and microarrays for the assessment of the resistome of the contaminating population. The presence of SARS-CoV-2 was also monitored by PCR. Results and conclusions: PCHS usage was associated with a stable 80% decrease in surface pathogens compared to levels detected for chemical disinfection (P < 0.01), accompanied by an up to 2 log decrease in resistance genes (Pc < 0.01). The effects were reversed when reintroducing chemical disinfection, which counteracted the action of the PCHS. SARS-CoV-2 was not detectable in both the pre-PCHS and PCHS periods. As the control of microbial contamination is a major issue, especially during pandemic emergencies, collected data suggest that PCHS may be successfully used to control virus spread without simultaneous worsening of the AMR concern

    CHARACTERIZATION AND CONSERVATION OF EMILIA ROMAGNA MAIZE LANDRACES

    The project aims at the characterization and conservation of regional maize landraces. Genetic material, consisting of 32 accessions, was retrieved both from recent surveys in Emilia Romagna, 11 accessions, and from the germplasm bank of CREA-MAC (Bergamo), 21 accessions. In particular, these last samples derive from the whole Italian collection of maize landraces carried out in the 50\u2019s by Brandolini and Fenaroli. In the first two years of the project, we carried out the landrace census and field characterization of the different materials according to UPOV guidelines and prepared, for each accession, a descriptive sheet. For each landrace, 100 seeds were sown and, from each plant, leaf samples were collected for DNA analysis. Approximately 2,000 plants, around 60 plants per landrace, were sampled. Each landrace is maintained in field by controlled randomintermating. For genetic analyses, 80 SSR markers were tested to identify the 10 most polymorphic to be used for the genetic characterization of all samples. . In parallel to these activities, an agronomic trial has been set up with the 32 landraces on four replicates to investigate production potential and resistance to mycotoxigenic fungi, with a special focus on Fusarium verticillioides. Considering that landraces can be exploited in conditions of low input agriculture, one of the objectives of the project is the identification of the best performing landraces suitable for cultivation nowadays (with a particular interest at mountain and disadvantaged areas). With this purpose, seeds, obtained from the propagation carried out in the first year, were distributed to partner farms and fields have been set-up in different hilly and mountainous areas of the region Emilia Romagna. These fields are presently underway with the double aim to evaluate the landraces in low input conditions and to propagate seeds for the future cultivation.     CHARACTERIZATION AND VALORIZATION OF MAIZE LANDRACES FROM VALLE D'AOSTA

    During 1949-1950 in Italy begun a formal investigation to characterize maize (Zea mays L.) cultivation. In 1954, started a project for the sampling of all Italian maize landraces; this work ended with the collection of 562 different accessions collected in all regions with the exception of Valle d’Aosta, even if historical cultivation of maize in this Region is well documented. In Italy maize landraces have been extensively grown until the mid of the XX century when the cultivation of hybrid took place due to their significant agronomic performances. Despite that, being Valle d’Aosta a mountain region where intensive maize cultivation never started, it was possible to preserve the presence of some landraces. These local materials, which are still cultivated, mainly at domestic level, have high importance from a genetic and historical point of view. Recently, 5 maize landraces from Valle d’Aosta and 2 landraces from the adjacent Canavese (Piedmont) have been collected and subjected to historic, morphologic and genetic characterization. These landraces were named after the sampling location as it follows: Arnad, Arnad-Crest, Chatillon, Entrebin, Perloz, Bianco Canavese, and Rostrato Canavese. Firstly, on these 6 varieties the historic characterization has been carried out. Information and photographs have been searched in local archives and this was crucial to prove their long presence in all the sampling sites under study. From this historic reconstruction, the variety Entrebin resulted as the one that is better historically characterized. To study the variability and differentiation of landraces from Valle d’Aosta, the genetic characterization was performed by the means of 10 SSR markers tested on 20 samples from each landrace. This study highlighted a significant genetic variability among the landraces and, especially, a good level of differentiation between the accessions under investigation. This last result may be explained by the long reproductive isolation experienced by these materials. Complete morphological characterization is actually ongoing. Preliminary morphological observations revealed that these landraces have, generally, flint kernels with the exception of Bianco Canavese (dent) whose color is variable from white (Bianco Canavese) to dark red (Chatillon). Arnad landrace showed 8 kernel rows, probably being an Eight-rowed Flint while the others presented more rows, like many Derived Races. Interestingly, Perloz and Rostrato Canavese showed kernels with an apical beak which was more pronounced in the latter. This suggest that these two landraces belong to the “Rostrata” group, which is common in mountain areas. The present work confirms the importance of mountain areas in conserving biodiversity and increases the rich Italian maize germplasm with materials well adapted to marginal areas. Such new genetic variability may be used to breed new materials for a more resilient agriculture

    Clinical practice: Drug desensitization in children

    Immediate type allergic reactions to medication are potentially life threatening and can hamper drug therapy of several medical conditions. Exact incidence and prevalence data for these reactions in children are lacking. If no alternative drug treatment is available, a desensitization procedure may secure the continuation of necessary therapy. Desensitization is only appropriate in case of a strong suspicion of an IgE-mediated allergic reaction. It should be performed by trained clinicians (allergy specialists) in a hospital setting where treatment of a potential anaphylactic reaction can be done without any delay. In this article, literature describing desensitization procedures for several antibiotics, antineoplastic agents, and vaccines in children is reviewed. In general, desensitization schemes for children differ only in final dose from schemes for adults. Contradictory data were found regarding the protective effects of premedication with antihistamines and glucocorticoids

    Fluoro-edenitic fibres in the sputum of subjects from Biancavilla (Sicily): a pilot study

    BACKGROUND: An excess of mortality for malignant neoplasms of the pleura in Biancavilla, promoted an investigation for pleural mesothelioma, disclosing 17 cases. As the absence of known sources of asbestos exposure, a local stone quarry, located near the inhabited area, used for the extraction of building materials, was investigated. Amphibolic fibres were found in the quarry and identified as fluoro-edenite "new end-member of the edenite / fluoro-edenite series" and recognized as the fluoro-edenite holotype by International Mineralogical Association – Commission on New Minerals and Mineral Names. A pilot study was performed to verify the feasibility of using spontaneous sputum as an exposure indicator for these fibres, in a context in which the use of aerosol-induced sputum technique would not be easily accepted. METHODS: Hypothesizing a behaviour of the new fibre analogous to that of asbestos, the determination of the free fibres and the ferruginous bodies in spontaneous sputum was carried out. Phase Contrast Optical Microscope and an Environmental Scanning Electron Microscope fitted with X-ray energy dispersive analysis system (micro-analysis) were used to examine the samples. The criteria for inclusion in the study were: 1) subjects hospitalized for exacerbation of chronic obstructive pulmonary disease symptoms, 2) age ≥ 45 years, 3) residence in Biancavilla for at least 30 years. RESULTS: The preliminary findings are related to 12 subjects (7 females and 5 males). Uncoated fibres (with length > 5 μm, diameter < 3 μm, aspect ratio 3.1) and ferruginous bodies were searched. Six out of twelve subjects (4 females, 2 males) had at least one of the three samples positive for the presence of fluoro-edenite, confirmed by micro-analysis. The fibre concentration found in the sputum ranged from 0.04 to 10 fibres/g; the length from 20 to 40 μm, the diameter was < 0.5 μm. No ferruginous bodies were found in any of the samples. The four women with a positive sample were housewives. Of the two men with a positive sample, one was a farmer and the other a mason. Therefore, it may be assumed that the exposure to fluoro-edenitic fibres was mainly environmental. CONCLUSION: The occurrence of the pleural mesothelioma cases and the presence of fluoro-edenitic fibres in spontaneous sputum, evidence the need to study the biological activity of fluoro-edenitic fibres and the implementation of epidemiological monitoring systems

    Cancer effects of formaldehyde: a proposal for an indoor air guideline value

    Formaldehyde is a ubiquitous indoor air pollutant that is classified as “Carcinogenic to humans (Group 1)” (IARC, Formaldehyde, 2-butoxyethanol and 1-tert-butoxypropanol-2-ol. IARC monographs on the evaluation of carcinogenic risks to humans, vol 88. World Health Organization, Lyon, pp 39–325, 2006). For nasal cancer in rats, the exposure–response relationship is highly non-linear, supporting a no-observed-adverse-effect level (NOAEL) that allows setting a guideline value. Epidemiological studies reported no increased incidence of nasopharyngeal cancer in humans below a mean level of 1 ppm and peak levels below 4 ppm, consistent with results from rat studies. Rat studies indicate that cytotoxicity-induced cell proliferation (NOAEL at 1 ppm) is a key mechanism in development of nasal cancer. However, the linear unit risk approach that is based on conservative (“worst-case”) considerations is also used for risk characterization of formaldehyde exposures. Lymphohematopoietic malignancies are not observed consistently in animal studies and if caused by formaldehyde in humans, they are high-dose phenomenons with non-linear exposure–response relationships. Apparently, these diseases are not reported in epidemiological studies at peak exposures below 2 ppm and average exposures below 0.5 ppm. At the similar airborne exposure levels in rodents, the nasal cancer effect is much more prominent than lymphohematopoietic malignancies. Thus, prevention of nasal cancer is considered to prevent lymphohematopoietic malignancies. Departing from the rat studies, the guideline value of the WHO (Air quality guidelines for Europe, 2nd edn. World Health Organization, Regional Office for Europe, Copenhagen, pp 87–91, 2000), 0.08 ppm (0.1 mg m−3) formaldehyde, is considered preventive of carcinogenic effects in compliance with epidemiological findings

    Is exposure to formaldehyde in air causally associated with leukemia?—A hypothesis-based weight-of-evidence analysis

    Recent scientific debate has focused on the potential for inhaled formaldehyde to cause lymphohematopoietic cancers, particularly leukemias, in humans. The concern stems from certain epidemiology studies reporting an association, although particulars of endpoints and dosimetry are inconsistent across studies and several other studies show no such effects. Animal studies generally report neither hematotoxicity nor leukemia associated with formaldehyde inhalation, and hematotoxicity studies in humans are inconsistent. Formaldehyde's reactivity has been thought to preclude systemic exposure following inhalation, and its apparent inability to reach and affect the target tissues attacked by known leukemogens has, heretofore, led to skepticism regarding its potential to cause human lymphohematopoietic cancers. Recently, however, potential modes of action for formaldehyde leukemogenesis have been hypothesized, and it has been suggested that formaldehyde be identified as a known human leukemogen. In this article, we apply our hypothesis-based weight-of-evidence (HBWoE) approach to evaluate the large body of evidence regarding formaldehyde and leukemogenesis, attending to how human, animal, and mode-of-action results inform one another. We trace the logic of inference within and across all studies, and articulate how one could account for the suite of available observations under the various proposed hypotheses. Upon comparison of alternative proposals regarding what causal processes may have led to the array of observations as we see them, we conclude that the case fora causal association is weak and strains biological plausibility. Instead, apparent association between formaldehyde inhalation and leukemia in some human studies is better interpreted as due to chance or confounding
