    Foram utilizados seis cordeiros machos da raça Santa InĂȘs, castrados, com idade mĂ©dia de quatro meses, que foram abatidos com peso entre 30-35 kg com o objetivo de avaliar o e feito de trĂȘs tipos de acondicionamento da carne ovina (natural, resfriada e congelada) na qualidade sensorial e microbiolĂłgica da carne. Para a anĂĄlise sensorial foi separado e preparado o mĂșsculo Longissimus dorsi para um painel de provadores que avaliaram a carne quanto ao aroma, sabor, maciez, suculĂȘncia, mastigabilidade e aparĂȘncia geral. O mesmo mĂșsculo foi utilizado para a anĂĄlise microbiolĂłgica onde se determinaram coliformes totais e fecais, bolores e leveduras. A carne in natura foi mais macia e de melhor mastigabilidade (p<0,05) em relação Ă  resfriada e congelada. Observou-se tambĂ©m nas carcaças nĂŁo resfriadas maior incidĂȘncia de bolores e leveduras e presença de coliformes fecais. Apesar da carne in natura ter apresentado maior maciez, o crescimento microbiano foi maior neste tipo de conservação pĂłs abate, o que pode comprometer a saĂșde humana

    Nanostructured 3D Constructs Based on Chitosan and Chondroitin Sulphate Multilayers for Cartilage Tissue Engineering

    Nanostructured three-dimensional constructs combining layer-by-layer technology (LbL) and template leaching were processed and evaluated as possible support structures for cartilage tissue engineering. Multilayered constructs were formed by depositing the polyelectrolytes chitosan (CHT) and chondroitin sulphate (CS) on either bidimensional glass surfaces or 3D packet of paraffin spheres. 2D CHT/CS multi-layered constructs proved to support the attachment and proliferation of bovine chondrocytes (BCH). The technology was transposed to 3D level and CHT/CS multi-layered hierarchical scaffolds were retrieved after paraffin leaching. The obtained nanostructured 3D constructs had a high porosity and water uptake capacity of about 300%. Dynamical mechanical analysis (DMA) showed the viscoelastic nature of the scaffolds. Cellular tests were performed with the culture of BCH and multipotent bone marrow derived stromal cells (hMSCs) up to 21 days in chondrogenic differentiation media. Together with scanning electronic microscopy analysis, viability tests and DNA quantification, our results clearly showed that cells attached, proliferated and were metabolically active over the entire scaffold. Cartilaginous extracellular matrix (ECM) formation was further assessed and results showed that GAG secretion occurred indicating the maintenance of the chondrogenic phenotype and the chondrogenic differentiation of hMSCs

    SCI1 Is a Direct Target of AGAMOUS and WUSCHEL and Is Specifically Expressed in the Floral Meristematic Cells

    The specified floral meristem will develop a pre-established number of floral organs and, thus, terminate the floral meristematic cells. The floral meristematic pool of cells is controlled, among some others, by WUSCHEL (WUS) and AGAMOUS (AG) transcription factors (TFs). Here, we demonstrate that the SCI1 (Stigma/style cell-cycle inhibitor 1) gene, a cell proliferation regulator, starts to be expressed since the floral meristem specification of Nicotiana tabacum and is expressed in all floral meristematic cells. Its expression is higher in the floral meristem and the organs being specified, and then it decreases from outside to inside whorls when the organs are differentiating. SCI1 is co-expressed with N. tabacum WUSCHEL (NtWUS) in the floral meristem and the whorl primordia at very early developmental stages. Later in development, SCI1 is co-expressed with NAG1 (N. tabacum AG) in the floral meristem and specialized tissues of the pistil. In silico analyses identified cis-regulatory elements for these TFs in the SCI1 genomic sequence. Yeast one-hybrid and electrophoresis mobility shift assay demonstrated that both TFs interact with the SCI1 promoter sequence. Additionally, the luciferase activity assay showed that NAG1 clearly activates SCI1 expression, while NtWUS could not do so. Taken together, our results suggest that during floral development, the spatiotemporal regulation of SCI1 by NtWUS and NAG1 may result in the maintenance or termination of proliferative cells in the floral meristem, respectively.Fil: Cruz, Joelma O..     Measurement of the Λ0b→ J/ψΛ angular distribution and the Λ0b polarisation in pp collisions

    This paper presents an analysis of the Λ0b→ J/ψΛ angular distribution and the transverse production polarisation of Λ0b baryons in proton-proton collisions at centre-of-mass energies of 7, 8 and 13 TeV. The measurements are performed using data corresponding to an integrated luminosity of 4.9 fb−1, collected with the LHCb experiment. The polarisation is determined in a fiducial region of Λ0b transverse momentum and pseudorapidity of 1 < pT< 20 GeV/c and 2 < η < 5, respectively. The data are consistent with Λ0b baryons being produced unpolarised in this region. The parity-violating asymmetry parameter of the Λ → pπ− decay is also determined from the data and its value is found to be consistent with a recent measurement by the BES III collaboration

    Analysis of shear forces during mash disk formation

    This report concerns a forming process in which mash is forced from a spreading manifold into moulds on a rotating drum, transported in the moulds underneath a surface held flush with the drum (the shoe), and ejected from the moulds. The quality of the final product is understood to be related to the shear stresses experienced by the mash in the moulds as it is transported under the shoe. We describe and analyse mathematical models of the forming process, focusing on the fluid mechanics of mash in a mould. We treat this as a driven cavity flow and obtain flow profiles, stress profiles, and expressions for the maximum shear stress for different rheological models of the mash (Newtonian fluid, power-law fluid, and Bingham plastic)
