    Exercise-based multimodal programming : A treatment gap for older adults with advanced cancer [Commentary]

    [Extract] Approximately 60% of new cancer diagnoses occur in patients aged 65 years and older.1 Normal aging is associated with a decline in health and physical function.2 A cancer diagnosis and subsequent treatment can accelerate this age-related physical decline, increasing the risk of adverse health events and mortality.1 Aerobic and resistance exercise is an effective therapy to improve physical fitness and quality of life and to reduce cancer-related fatigue in individuals diagnosed with cancer, including advanced cancer.3 The American College of Sports Medicine guidelines recommend maintenance of physical activity during active cancer treatment,3 but guidelines specific to advanced cancer or elderly populations are lacking. Older patients with cancer are vastly underrepresented in clinical trials, including exercise-based trials,4 due in part to the challenges introduced by the observed heterogeneity among older adults with respect to comorbid conditions, functional status, motivation, and safety-related concerns of the treating health care professionals.4 Older adults are more likely to fear physical activity due to potential injury and to lack of guidance regarding how to start exercising.5,

    BACKGROUND: glutathione (GSH) is the most abundant thiol antioxidant in mammalian cells. It directly reacts with reactive oxygen species (ROS), functions as a cofactor of antioxidant enzymes, and maintains thiol redox potential in cells. GSH depletion has been implicated in the pathogenesis of neurological diseases, particularly to Parkinson's disease (PD). The purpose of this study was to investigate the change of cellular antioxidant status and basic cell functions in the relatively early stages of GSH depletion. RESULTS: in this study, GSH was depleted by inhibition of glutamylcysteine synthetase using buthionine sulfoximine (BSO) treatment in Ht22, a neuronal cell line derived from mouse hippocampus. Treatment with BSO produced dose-dependent decreases in total GSH level, Fe3+-reducing ability (FRAP assay), Cu2+-reducing ability (Antioxidant Potential, AOP assay), and ABTS free radical scavenging ability (ABTS assay) of the cells, but the sensitivity of these indicators to dosage varied considerably. Most of the changes were completed during the first 8 hours of treatment. Cell viability was tested by MTT (3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl tetrazolium bromid) assay, and cells at lower density in culture were found to be more sensitive to GSH depletion. The activity of antioxidant enzymes, such as glutathione peroxidase (GPx), glutathione reductase (GR), and copper/zinc superoxide dismutase (Cu/Zn-SOD) were affected by GSH depletion. A cDNA expression array assay of the effects of BSO treatment showed significantly decreased mRNA level for 3 genes, and significantly increased mRNA level for 10 genes, including the antioxidant enzymes Cu/Zn-SOD and thioredoxin peroxidase 2 (TPxII). CONCLUSIONS: the study suggests that there are BSO-sensitive and BSO-resistant pools of GSH in Ht22 cells, and that different categories of antioxidant react differently to GSH depletion. Further, the effect of GSH status on cell viability is cell density dependent. Finally, the alterations in expression or activity of several antioxidant enzymes provide insight into the various cellular responses to GSH depletion

    River export of nutrients and organic matter from the North Slope of Alaska to the Beaufort Sea

The definitive version was published in Water Resources Research 50 (2014): 1823–1839, doi:10.1002/2013WR014722.While river-borne materials are recognized as important resources supporting coastal ecosystems around the world, estimates of river export from the North Slope of Alaska have been limited by a scarcity of water chemistry and river discharge data. This paper quantifies water, nutrient, and organic matter export from the three largest rivers (Sagavanirktok, Kuparuk, and Colville) that drain Alaska's North Slope and discusses the potential importance of river inputs for biological production in coastal waters of the Alaskan Beaufort Sea. Together these rivers export ∼297,000 metric tons of organic carbon and ∼18,000 metric tons of organic nitrogen each year. Annual fluxes of nitrate-N, ammonium-N, and soluble reactive phosphorus are approximately 1750, 200, and 140 metric tons per year, respectively. Constituent export from Alaska's North Slope is dominated by the Colville River. This is in part due to its larger size, but also because constituent yields are greater in the Colville watershed. River-supplied nitrogen may be more important to productivity along the Alaskan Beaufort Sea coast than previously thought. However, given the dominance of organic nitrogen export, the potential role of river-supplied nitrogen in support of primary production depends strongly on remineralization mechanisms. Although rivers draining the North Slope of Alaska make only a small contribution to overall river export from the pan-arctic watershed, comparisons with major arctic rivers reveal unique regional characteristics as well as remarkable similarities among different regions and scales.     Needles in haystacks: using fast-response LA chambers and ICP-TOF-MS to identify asbestos fibres in malignant mesothelioma models

    Malignant mesothelioma is an aggressive cancer associated with exposure to asbestos. Diagnosis of mesothelioma and other related lung diseases remains elusive due to difficulties surrounding identification and quantification of asbestos fibres in lung tissue. This article presents a laser ablation-inductively coupled plasma-mass spectrometry (LA-ICP-MS) method to identify asbestos fibres in cellular models of mesothelioma. Use of a high-speed laser ablation system enabled rapid imaging of the samples with a lateral resolution of 3 μm, whilst use of a prototype time-of-flight ICP-MS provided pseudo-simultaneous detection of the elements between mass 23 (Na) and mass 238 (U). Three forms of asbestos fibre (actinolite, amosite and crocidolite) were distinguished from a non-asbestos control (wollastonite) based on their elemental profile, which demonstrated that LA-ICP-MS could be a viable technique for identification of asbestos fibres in clinical research samples

    Effects of forest management on early-successional avian species in South Carolina

    This study represents the largest known effort to inventory Ruffed Grouse and Golden- winged Warblers in the State of South Carolina. We found low Ruffed Grouse and Golden- winged Warbler occupancy rates across two seasons (spring and summer 2020 and 2021), indicating the need for both robust monitoring protocols and targeted habitat management for the benefit of these species. Our results indicate unique habitat preferences of Ruffed Grouse in the Southern Blue Ridge Ecoregion. Additionally, our results provide insight into multiple parameters that drive early-successional songbird species occupancy. This project provides information that will aid in both habitat management and conservation of high priority early- successional avian species. This project also provides context for efficient monitoring protocols

    Effects of training multiple form classes on acquisition, generalization and maintenance of word retrieval in a single subject

    A study by McNeil et nl. reported no generalization effects in two individuals with aphasia following application of a word finding treatment in which subjects were trained sequentially on lexical items arranged by form class. The present investigation examined, in one of the two subjects from the original study, whether training on lexical items from a variety of form classes concurrently would result in greater response generalization than was observed previously in this subject. Results replicated earlier findings with positive acquisition and maintenance effects and little evidence of generalization to untrained items within or across form classes
