    PinR mediates the generation of reversible population diversity in Streptococcus zooepidemicus

    Opportunistic pathogens must adapt to and survive in a wide range of complex ecosystems. Streptococcus zooepidemicus is an opportunistic pathogen of horses and many other animals, including humans. The assembly of different surface architecture phenotypes from one genotype is likely to be crucial to the successful exploitation of such an opportunistic lifestyle. Construction of a series of mutants revealed that a serine recombinase, PinR, inverts 114 bp of the promoter of SZO_08560, which is bordered by GTAGACTTTA and TAAAGTCTAC inverted repeats. Inversion acts as a switch, controlling the transcription of this sortase-processed protein, which may enhance the attachment of S. zooepidemicus to equine trachea. The genome of a recently sequenced strain of S. zooepidemicus, 2329 (Sz2329), was found to contain a disruptive internal inversion of 7 kb of the FimIV pilus locus, which is bordered by TAGAAA and TTTCTA inverted repeats. This strain lacks pinR and this inversion may have become irreversible following the loss of this recombinase. Active inversion of FimIV was detected in three strains of S. zooepidemicus, 1770 (Sz1770), B260863 (SzB260863) and H050840501 (SzH050840501), all of which encoded pinR. A deletion mutant of Sz1770 that lacked pinR was no longer capable of inverting its internal region of FimIV. The data highlight redundancy in the PinR sequence recognition motif around a short TAGA consensus and suggest that PinR can reversibly influence the wider surface architecture of S. zooepidemicus, providing this organism with a bet-hedging solution to survival in fluctuating environments

    Redox stratification drives enhanced growth in a commercially important deposit-feeding invertebrate: implications for bioremediation and integrated aquaculture.

    Effective and affordable treatment of waste solids is a key sustainability challenge for the aquaculture industry. Here, we investigated the potential for a deposit-feeding sea cucumber, Holothuria scabra, to provide a remediation service whilst concurrently yielding a high-value secondary product in a land-based recirculating aquaculture system (RAS). The effect of sediment depth, particle size and redox regime were examined in relation to changes in the behaviour, growth and biochemical composition of juvenile sea cucumbers cultured for 81 d in manipulated sediment systems, describing either fully oxic or stratified (oxic-anoxic) redox regimes. The redox regime was the principal factor affecting growth, biochemical composition and behaviour, while substrate depth and particle size did not significantly affect growth rate or biomass production. Animals cultured under fully oxic conditions exhibited negative growth and had higher lipid and carbohydrate contents, potentially due to compensatory feeding in response to higher microphytobenthic production. In contrast, animals in the stratified treatments spent more time feeding, generated faster growth and produced significantly higher biomass yields (626.89 ± 35.44 g m-2 versus 449.22 ± 14.24 g m-2; mean ± SE). Further, unlike in oxic treatments, growth in the stratified treatments did not reach maximum biomass carrying capacity, indicating that stratified sediment is more suitable for culturing sea cucumbers. However, the stratified sediments may exhibit reduced bioremediation ability relative to the oxic sediment, signifying a trade-off between remediation efficiency and exploitable biomass yield

    Who's most at risk of poor body image? Identifying subgroups of adolescent social media users over the course of a year

    Types and stability of appearance-related social media use patterns remain under-explored despite established links between social media use and wellbeing. This study aimed to identify subgroups of social media users, and explore whether subgroup membership was stable over time and associated with body image-related outcomes. Adolescents (N = 766; Mage = 12.76, SD = 0.73; 49.40% female) completed four surveys across 1-year, reporting several social media use indices, body dissatisfaction, dietary restraint, and strategies to increase muscle. Latent profile analyses identified two subgroups (moderate and high users), that remained reasonably stable over time. The high subgroup exhibited poorer body image at baseline, though differences seemed to dissipate somewhat over 1-year. Examination of subgroup transition over time showed more rapid increases in poor body image outcomes among social media increasers and more rapid declines for reducers. Prevention programs which aim to reduce high levels of social media use among children, young adolescents, and high-risk individuals (i.e., appearance-focused users) appear warranted

    The telepresence experience can be evoked in a number of ways. A well-known example is a player of videogames who reports about a telepresence experience, a subjective experience of being in one place or environment, even when physically situated in another place. In this paper we set the phenomenon of telepresence into a theoretical framework. As people react subjectively to stimuli from telepresence, empirical studies can give more evidence about the phenomenon. Thus, our contribution is to bridge the theoretical with the empirical. We discuss theories of perception with an emphasis on Heidegger, Merleau-Ponty and Gibson, the role of the senses and the Spinozian belief procedure. The aim is to contribute to our understanding of this phenomenon. A telepresence-study that included the affordance concept is used to empirically study how players report sense-reactions to virtual sightseeing in two cities. We investigate and explore the interplay of the philosophical and the empirical. The findings indicate that it is not only the visual sense that plays a role in this experience, but all senses

    Shared social identity content is the basis for leaders' mobilization of followers

    Objectives There is growing research interest in the social identity approach to leadership in sport. Researchers have examined how leaders’ representation of a shared social identity allows them to motivate group members but has neglected the role that identity content plays in this process. The present research addresses this issue in two experimental studies that examine the effect of sharedness in identity content (i.e., beliefs about what it means to be a member of a group) on leaders’ mobilization of group members. Design A 2 X 2 experimental — between-participant — design, with two shared and two non-shared conditions. Method In Study 1, 160 athletes imagined themselves in one of four sport team scenarios and responded to measures of mobilization (e.g., willingness to invest time on task). In Study 2 (laboratory experiment), we manipulated sharedness and assessed 114 participants’ behavioural mobilization and task performance. Results Study 1 supports the hypothesis that identity content that is shared (rather than non-shared) between leaders and group members increases members’ willingness to invest time on a task. Study 2 replicates these results and also shows that increased effort among group members mediates the relationship between shared identity content and members’ improved task performance. Conclusions The present research is the first to provide evidence that sport leaders’ capacity to mobilize the effort of group members rests upon their ability to build shared identity content

    Block-Diagonalization and f-electron Effects in Tight-Binding Theory

    We extend a tight-binding total energy method to include f-electrons, and apply it to the study of the structural and elastic properties of a range of elements from Be to U. We find that the tight-binding parameters are as accurate and transferable for f-electron systems as they are for d-electron systems. In both cases we have found it essential to take great care in constraining the fitting procedure by using a block-diagonalization procedure, which we describe in detail.Comment: 9 pages, 6 figure

    Electrochemically copper-doped bismuth tellurium selenide thin films

    We report the first results of a study on electrochemically doped copper bismuth tellurium selenide thin films electrodeposited from aqueous nitric acid electrolytes containing up to 2 mM of Cu(NO3)2. The effect of Cu(NO3)2 concentration on the composition, structure and thermoelectric properties of the bismuth tellurium selenide films is investigated by scanning electron microscopy, energy-dispersive X-ray spectroscopy, X-ray diffraction and Seebeck and Hall effect measurements. A Cu(NO3)2 concentration of 1.5 mM is found to offer a Seebeck coefficient of up to −390 μV K−1 at room temperature, which is the highest reported to date for an electrodeposited bismuth tellurium compound
