    Uncertainty in the availability of natural resources: Fossil fuels, critical metals and biomass

    Energy policies are strongly influenced by resource availability and recoverability estimates. Yet these estimates are often highly uncertain, frequently incommensurable, and regularly contested. This paper explores how the uncertainties surrounding estimates of the availability of fossil fuels, biomass and critical metals are conceptualised and communicated. The contention is that a better understanding of the uncertainties surrounding resource estimates for both conventional and renewable energy resources can contribute to more effective policy decision making in the long term. Two complementary approaches for framing uncertainty are considered in detail: a descriptive typology of uncertainties and a framework that conceptualises uncertainty as alternative states of incomplete knowledge. Both have the potential to be useful analytical and communication tools. For the three resource types considered here we find that data limitations, inconsistent definitions and the use of incommensurable methodologies present a pervasive problem that impedes comparison. Many aspects of resource uncertainty are also not commonly captured in the conventional resource classification schemes. This highlights the need for considerable care when developing and comparing aggregate resource estimates and when using these to inform strategic energy policy decisions

    Design criteria and technology requirements for a system of radar reference devices to be fixed to the surfaces of the inner planets are discussed. Offshoot applications include the use of radar corner reflectors as landing beacons on the planetary surfaces and some deep space applications that may yield a greatly enhanced knowledge of the gravitational and electromagnetic structure of the solar system. Passive retroreflectors with dimensions of about 4 meters and weighing about 10 kg are feasible for use with orbiting radar at Venus and Mars. Earth-based observation of passive reflectors, however, would require very large and complex structures to be delivered to the surfaces. For Earth-based measurements, surface transponders offer a distinct advantage in accuracy over passive reflectors. A conceptual design for a high temperature transponder is presented. The design appears feasible for the Venus surface using existing electronics and power components

    Synthesis, structure and conductivity of sulfate and phosphate doped SrCoO3

    In this paper we report the successful incorporation of sulfate and phosphate into SrCoO3 leading to a change from a 2H– to a 3C–perovskite polymorph. Structural characterization by neutron diffraction showed extra weak peaks related to oxygen vacancy ordering, and these could be indexed on an expanded tetragonal cell, containing two inequivalent Co sites, similar to previously reported for Sb doped SrCoO3. Conductivity measurements on the doped systems showed a large enhancement compared to the undoped hexagonal system, consistent with corner–sharing of CoO6 octahedra for the former. Further work on the doped samples shows, however, that they are metastable, transforming back to the hexagonal cell on annealing at intermediate temperatures. The incorporation of Fe was shown, however, to improve the stability at intermediate temperatures, and these co–doped phases also showed high conductivities

    Preliminary comparison of 3.5-cm and 12.6-cm wavelength continuous wave observations of Mars

    Radar observations of Mars at Goldstone in 1990 were conducted by transmitting pure sinusoidal signals at 3.5-cm wavelengths and receiving the Doppler-spread echoes from Mars at Earth. Radar transmissions were circularly polarized and the echoes recorded in two senses: depolarized and polarized. Latitudes of the subradar points are between 3.5 deg and 11.1 deg S; longitude coverage is discontinuous. The observed depolarized and polarized echo total cross-sections and their ratios for two wavelengths were compared and discussed

    Training the Foot Soldiers of Inquiry: Development and Evaluation of a Graduate Teaching Assistant Learning Community

    As part of a Howard Hughes Program for Innovation in Science Education grant at Iowa State University, a series of interdisciplinary graduate teaching assistant learning communities (TALC) were developed. The purpose of these communities was to create an environment to facilitate teaching assistants\u27 pedagogical development and training to enhance the implementation of inquiry experiences in the undergraduate laboratories. The TALC evaluated in this study were held for two consecutive semesters and included teaching assistants who facilitated multi-week course-based research experiences in their respective STEM courses. Topics discussed during the TALC were based on the teaching assistants\u27 concerns related to teaching this type of course. Evaluation consisted of weekly reflection responses, a pre- and post-survey of instructional methods they consider to facilitate inquiry, pre-post definitions of inquiry-based instruction, and end-of-semester evaluations of the learning community experiences.     Inhibition of Îł-secretase induces G2/M arrest and triggers apoptosis in breast cancer cells

    Îł-Secretase activity is vital for the transmembrane cleavage of Notch receptors and the subsequent migration of their intracellular domains to the nucleus. Notch overexpression has been associated with breast, colon, cervical and prostate cancers. We tested the effect of three different Îł-secretase inhibitors (GSIs) in breast cancer cells. One inhibitor (GSI1) was lethal to breast cancer cell lines at concentrations of 2 ΌM and above but had a minimal effect on the non-malignant breast lines. GSI1 was also cytotoxic for a wide variety of cancer cell lines in the NCI60 cell screen. GSI1 treatment resulted in a marked decrease in Îł-secretase activity and downregulation of the Notch signalling pathway with no effects on expression of the Îł-secretase components or ligands. Flow cytometric and western blot analyses indicated that GSI1 induces a G2/M arrest leading to apoptosis, through downregulation of Bcl-2, Bax and Bcl-XL. GSI1 also inhibited proteasome activity. Thus, the Îł-secretase inhibitor GSI1 has a complex mode of action to inhibit breast cancer cell survival and may represent a novel therapy in breast cancer

    Variable-frequency-train stimulation of skeletal muscle after spinal cord injury

    Skeletal muscle, after spinal cord injury (SCI), becomes highly susceptible to fatigue. Variable-frequency trains (VFTs) enhance force in fatigued human skeletal muscle of able-bodied (AB) individuals. VFTs do this by taking advantage of the catch-like property of skeletal muscle. However, mechanisms responsible for fatigue in AB and SCI subjects may not be the same, and the efficacy of VFT stimulation after SCI is unknown. Accordingly, we tested the hypothesis that VFT stimulation would augment torque-time integral in SCI subjects. The quadriceps femoris muscle was stimulated with constant frequency trains (CFTs) (six 200 s square wave pulses separated by 70 ms) or VFTs (a train identical to the CFT, except that the first two pulses were separated by 5 ms) in SCI and AB subjects. After 180 contractions (50% duty cycle), isometric peak torque decreased 44, 56, and 67 percent, in the AB (n = 10), acute SCI (n = 10), and chronic SCI (n = 12) groups, respectively. In fatigued muscle, VFTs enhanced the torque-time integral by 18 percent in AB subjects and 6 percent in chronic SCI patients, and had no effect in acute SCI patients when compared to the corresponding CFT. The much faster rise times in SCI subjects (~80 ms vs. 120 ms in AB subjects) probably contributed to the inability of VFTs to enhance torque-time integrals in SCI patients. The results suggest that the use of VFT stimulation in patients with SCI may not be as efficacious as it is in AB persons

    A combined approach for comparative exoproteome analysis of Corynebacterium pseudotuberculosis

    Background: Bacterial exported proteins represent key components of the host-pathogen interplay. Hence, we sought to implement a combined approach for characterizing the entire exoproteome of the pathogenic bacterium Corynebacterium pseudotuberculosis, the etiological agent of caseous lymphadenitis (CLA) in sheep and goats. Results: An optimized protocol of three-phase partitioning (TPP) was used to obtain the C. pseudotuberculosis exoproteins, and a newly introduced method of data-independent MS acquisition (LC-MSE) was employed for protein identification and label-free quantification. Additionally, the recently developed tool SurfG+ was used for in silico prediction of sub-cellular localization of the identified proteins. In total, 93 different extracellular proteins of C. pseudotuberculosis were identified with high confidence by this strategy; 44 proteins were commonly identified in two different strains, isolated from distinct hosts, then composing a core C. pseudotuberculosis exoproteome. Analysis with the SurfG+ tool showed that more than 75% (70/93) of the identified proteins could be predicted as containing signals for active exportation. Moreover, evidence could be found for probable non-classical export of most of the remaining proteins. Conclusions: Comparative analyses of the exoproteomes of two C. pseudotuberculosis strains, in addition to comparison with other experimentally determined corynebacterial exoproteomes, were helpful to gain novel insights into the contribution of the exported proteins in the virulence of this bacterium. The results presented here compose the most comprehensive coverage of the exoproteome of a corynebacterial species so far
