    THE TYPES OF INDIGENOUS KNOWLEDGE TO BE RETAINED FOR YOUNG NEW ZEALAND BASED SAMOANS: A SAMOAN GRANDPARENTS’ PERSPECTIVE

    As a result of mass migration from Samoa to neighbouring countries during the 1950s and 1960s, there are now more Samoans living outside of their homeland than there are within the country itself. As many Samoan migrants are getting older, there is an urgent need to transfer the wealth of indigenous knowledge that they possess to the younger generations before it is too late. This is to ensure the survival of Samoa’s culture, heritage, customs, traditions and principles. This research uses semi-structured interviews with eight New Zealand based grandparents to understand the phenomena at hand. The aim of this study is to find out what types of indigenous knowledge that older NZ-based Samoans consider desirable for retention by younger NZ-born Samoans. Emergent results include the importance of Fa’aSamoa (Samoan way of life) which lies at the very core of all things Samoan, the church, respect (fa’aaloalo) for elders, the Samoan language and the importance of family

    The psychological impact of slaughterhouse employment: A systematic literature review

    The role of a slaughterhouse worker (SHW) involves the authorized killing of living beings, yet there is limited understanding of the consequences this behavior has on their well-being. The purpose of this systematic review is to collate and evaluate the current literature on the psychological impact of slaughterhouse employment. Fourteen studies met the specific a priori inclusion criteria. The findings from this review were demarcated by the focus of studies: (1) the prevalence of mental health disorders, (2) the types of coping mechanisms used, and (3) the link between slaughterhouse employment and crime perpetration. It was found that SHWs have a higher prevalence rate of mental health issues, in particular depression and anxiety, in addition to violence-supportive attitudes. Furthermore, the workers employ a variety of both adaptive and maladaptive strategies to cope with the workplace environment and associated stressors. Finally, there is some evidence that slaughterhouse work is associated with increased crime levels. The research reviewed has shown a link between slaughterhouse work and antisocial behavior generally and sexual offending specifically. There was no support for such an association with violent crimes, however. Based on existing research, we suggest future directions for research (i.e., applying more methodological rigor) but highlight key findings for practitioners and policymakers that warrant attention

    High Spatial and Temporal Resolution Census Data Reveal Communities at Risk Along the Wildland-Urban Interface (WUI) in California, USA

    We tracked census tract level population change along California\u27s wild land-urban interface (WUI) during the past decade (2010-2019), an ecological sensitive region transitioning from developed land to wilderness. Our results from Mann-Kendall analysis, a method employed for monotonic trend detection showed that about one-third (29.1%) of census tracts in California’s WUI have seen a significant population increase from 2010 to 2019, affecting 12.7% population in California. The population increase along WUI is largely driven by the sixteen counties in the San Francisco Bay Area (10) and Southern California (6). We also found that higher proportion of WUI residents in Bay Area and larger number of WUI residents in Southern California. Bay Area counties in general have a higher proportion of population living in WUI tracts with significant population increase than Southern California counties. However, the lower proportion of residents living in WUI in Southern California counties account for a much larger population. Riverside is the county with the highest number of residents living in WUI tracts that have experienced significant population increase during the past decade. These residents also account for a high proportion (29.2%) of total population in Riverside. Preliminary results showed that the increase of population along WUI is driven by the house affordability and house ownership in 16 counties of Bay Area and Southern California. These factors can still explain a significant amount of the spatial pattern if extended to all counties in California

    The Type I Interferon Receptor Is Not Required for Protection in the Chlamydia Muridarum and HSV-2 Murine Super-Infection Model

    Chlamydia trachomatis/HSV-2 vaginal co-infections are seen clinically, suggesting that these sexually transmitted pathogens may interact. We previously established an intravaginal Chlamydia muridarum/HSV-2 super-infection model and observed that chlamydial pre-infection protects mice from a subsequent lethal HSV-2 challenge. However, the mechanism of protection remains unknown. The type I interferon, IFN-β, binds to the type I interferon receptor (IFNR), elicits a host cellular antiviral response and inhibits HSV replication in vitro and in vivo. Previous studies have demonstrated that C. muridarum infection stimulates genital tract (GT) IFN-β production; therefore, we hypothesized that chlamydial pre-infection protects mice from HSV-2 challenge via the IFN-β/IFNR-induced antiviral response. To test this prediction, we quantified IFN-β levels in vaginal swab samples. Detection of IFN-β in C. muridarum singly infected, but not in mock-infected animals, prompted the use of the super-infection model in IFNR knockout (IFNR-/-) mice. We observed that C. muridarum pre-infection reduces HSV-2-induced mortality by 40% in wild-type mice and by 60% IFNR-/-mice. Severity of HSV-2 disease symptoms and viral shedding was also similarly reduced by C. muridarum pre-infection. These data indicate that, while chlamydial infection induces GT production of IFN-β, type I IFN-induced antiviral responses are likely not required for the observed protective effect

    β-catenin and transforming growth factor β have distinct roles regulating fibroblast cell motility and the induction of collagen lattice contraction

    <p>Abstract</p> <p>Background</p> <p>β-catenin and transforming growth factor β signaling are activated in fibroblasts during wound healing. Both signaling pathways positively regulate fibroblast proliferation during this reparative process, and the effect of transforming growth factor β is partially mediated by β-catenin. Other cellular processes, such as cell motility and the induction of extracellular matrix contraction, also play important roles during wound repair. We examined the function of β-catenin and its interaction with transforming growth factor β in cell motility and the induction of collagen lattice contraction.</p> <p>Results</p> <p>Floating three dimensional collagen lattices seeded with cells expressing conditional null and stabilized β-catenin alleles, showed a modest negative relationship between β-catenin level and the degree of lattice contraction. Transforming growth factor β had a more dramatic effect, positively regulating lattice contraction. In contrast to the situation in the regulation of cell proliferation, this effect of transforming growth factor β was not mediated by β-catenin. Treating wild-type cells or primary human fibroblasts with dickkopf-1, which inhibits β-catenin, or lithium, which stimulates β-catenin produced similar results. Scratch wound assays and Boyden chamber motility studies using these same cells found that β-catenin positively regulated cell motility, while transforming growth factor β had little effect.</p> <p>Conclusion</p> <p>This data demonstrates the complexity of the interaction of various signaling pathways in the regulation of cell behavior during wound repair. Cell motility and the induction of collagen lattice contraction are not always coupled, and are likely regulated by different intracellular mechanisms. There is unlikely to be a single signaling pathway that acts as master regulator of fibroblast behavior in wound repair. β-catenin plays dominant role regulating cell motility, while transforming growth factor β plays a dominant role regulating the induction of collagen lattice contraction.</p

    Host Nectin-1 Promotes Chlamydial Infection in the Female Mouse Genital Tract, But is Not Required for Infection in a Novel Male Murine Rectal Infection Model

Chlamydia trachomatis is the most common bacterial sexually transmitted pathogen, but more than 70% of patients fail to seek treatment due to the asymptomatic nature of these infections. Women suffer from numerous complications from chronic chlamydial infections, which include pelvic inflammatory disease and infertility. We previously demonstrated in culture that host cell nectin-1 knockdown significantly reduced chlamydial titers and inclusion size. Here, we sought to determine whether nectin-1 was required for chlamydial development in vivo by intravaginally infecting nectin-1-/- mice with Chlamydia muridarum and monitoring chlamydial shedding by chlamydial titer assay. We observed a significant reduction in chlamydial shedding in female nectin-1-/- mice compared to nectin-1+/+ control mice, an observation that was confirmed by PCR. Immunohistochemical staining in mouse cervical tissue confirmed that there are fewer chlamydial inclusions in Chlamydia-infected nectin-1-/- mice. Notably, anorectal chlamydial infections are becoming a substantial health burden, though little is known regarding the pathogenesis of these infections. We therefore established a novel male murine model of rectal chlamydial infection, which we used to determine whether nectin-1 is required for anorectal chlamydial infection in male mice. In contrast to the data from vaginal infection, no difference in rectal chlamydial shedding was observed when male nectin-1+/+ and nectin-1-/- mice were compared. Through the use of these two models, we have demonstrated that nectin-1 promotes chlamydial infection in the female genital tract but does not appear to contribute to rectal infection in male mice. These models could be used to further characterize tissue and sex related differences in chlamydial infection

    Keeping it real: Virtual connection with SToP trial community navigators

    Building trust and forging relationships with remote Aboriginal communities is an essential element of culturally informed, reciprocal research. Historically these relationships have been formed over-time where community members and researchers come together face to face to share their knowledge and yarn in both an informal and formal manner. Researchers from Telethon Kids Institute are partnering with local stakeholders and remote Aboriginal communities in the Kimberley, Western Australia (WA) to support healthy skin through the SToP (See, Treat, Prevent skin sores and scabies) Trial. The SToP trial, a collaboration between Telethon Kids Institute, Kimberley Aboriginal Medical Services (KAMS), Nirrumbuk Environmental Health Services and Western Australia Country Health Services (WACHS) – Kimberley is a clustered randomised trial with a stepped-wedge design. SToP trial consultation with stakeholders and communities commenced in 2016 to proceed consenting in 2018 and trial commencement in 2019. Since that time, the SToP trial team have been conducting intermittent fieldwork in nine remote Aboriginal communities in the Kimberley. However, due to the COVID-19 pandemic when Aboriginal health leaders recommended a cessation to research related travel in northern WA from March 11, 2020 to prevent the incursion of COVID-19 into Aboriginal communities with health vulnerabilities, crucial face-to-face yarning was no longer possible. At the time it appeared the existing relationships with communities involved in our research (the SToP trial) would be challenging to maintain without this ability to visit the communities. Fortunately, when tested, this assumption was erroneous. Here we report the successful use of technology to bridge the inability to visit communities in 2020 due to COVID-19. The Telethon Kulunga Aboriginal Unit (Kulunga) and SToP trial team members were able to connect virtually with Community Navigators from the Dampier Peninsula communities. The initial virtual meeting using Microsoft Teams technology involved four Community Navigators and their mentor, three Telethon Kids Institute and five Kulunga staff members. Community Navigators joined Microsoft Teams from their respective communities and Kulunga and Telethon Kids Institute staff joined from their homes. Not only was this an exciting new way of communicating, it enabled existing relationships to continue to be strengthened. Since the initial meeting, the teams have continued to meet virtually, and plan SToP trial health promotion activities including a community-driven, collaborative music video. While the significance of face-to-face yarning can never be overstated, having to adjust to a new way of yarning has reiterated the importance of connection, albeit virtually. Unfortunately, due to technical limitations, intermittent internet connectivity and various other challenges, there has been no opportunity to engage virtually with SToP trial communities in the East Kimberley. However, we continue to seek ways where virtual communication in these communities is possible

    Development of high amylose wheat through TILLING

    BACKGROUND: Wheat (Triticum spp.) is an important source of food worldwide and the focus of considerable efforts to identify new combinations of genetic diversity for crop improvement. In particular, wheat starch composition is a major target for changes that could benefit human health. Starches with increased levels of amylose are of interest because of the correlation between higher amylose content and elevated levels of resistant starch, which has been shown to have beneficial effects on health for combating obesity and diabetes. TILLING (Targeting Induced Local Lesions in Genomes) is a means to identify novel genetic variation without the need for direct selection of phenotypes. RESULTS: Using TILLING to identify novel genetic variation in each of the A and B genomes in tetraploid durum wheat and the A, B and D genomes in hexaploid bread wheat, we have identified mutations in the form of single nucleotide polymorphisms (SNPs) in starch branching enzyme IIa genes (SBEIIa). Combining these new alleles of SBEIIa through breeding resulted in the development of high amylose durum and bread wheat varieties containing 47-55% amylose and having elevated resistant starch levels compared to wild-type wheat. High amylose lines also had reduced expression of SBEIIa RNA, changes in starch granule morphology and altered starch granule protein profiles as evaluated by mass spectrometry. CONCLUSIONS: We report the use of TILLING to develop new traits in crops with complex genomes without the use of transgenic modifications. Combined mutations in SBEIIa in durum and bread wheat varieties resulted in lines with significantly increased amylose and resistant starch contents
