    KIF2A silencing inhibits the proliferation and migration of breast cancer cells and correlates with unfavorable prognosis in breast cancer

    Background; Kinesin family member 2a (KIF2A), a type of motor protein found in eukaryotic cells, is associated with development and progression of various human cancers. The role of KIF2A during breast cancer tumorigenesis and progression was studied. Methods; Immunohistochemical staining, real time RT-PCR and western blot were used to examine the expression of KIF2A in cancer tissues and adjacent normal tissues from breast cancer patients. Patients’ survival in relation to KIF2A expression was estimated using the Kaplan–Meier survival and multivariate analysis. Breast cancer cell line, MDA-MB-231 was used to study the proliferation, migration and invasion of cells following KIF2A-siRNA transfection. Results; The expression of KIF2A in cancer tissues was higher than that in normal adjacent tissues from the same patient (P < 0.05). KIF2A expression in cancer tissue with lymph node metastasis and HER2 positive cancer were higher than that in cancer tissue without (P < 0.05). A negative correlation was found between KIF2A expression levels in breast cancer and the survival time of breast cancer patients (P < 0.05). In addition, multivariate analysis indicated that KIF2A was an independent prognostic for outcome in breast cancer (OR: 16.55, 95% CI: 2.216-123.631, P = 0.006). The proliferation, migration and invasion of cancer cells in vitro were suppressed by KIF2A gene silencing (P < 0.05). Conclusions; KIF2A may play an important role in breast cancer progression and is potentially a novel predictive and prognostic marker for breast cancer

    Etiologic Diagnosis of Lower Respiratory Tract Bacterial Infections Using Sputum Samples and Quantitative Loop-Mediated Isothermal Amplification

    Etiologic diagnoses of lower respiratory tract infections (LRTI) have been relying primarily on bacterial cultures that often fail to return useful results in time. Although DNA-based assays are more sensitive than bacterial cultures in detecting pathogens, the molecular results are often inconsistent and challenged by doubts on false positives, such as those due to system- and environment-derived contaminations. Here we report a nationwide cohort study on 2986 suspected LRTI patients across P. R. China. We compared the performance of a DNA-based assay qLAMP (quantitative Loop-mediated isothermal AMPlification) with that of standard bacterial cultures in detecting a panel of eight common respiratory bacterial pathogens from sputum samples. Our qLAMP assay detects the panel of pathogens in 1047(69.28%) patients from 1533 qualified patients at the end. We found that the bacterial titer quantified based on qLAMP is a predictor of probability that the bacterium in the sample can be detected in culture assay. The relatedness of the two assays fits a logistic regression curve. We used a piecewise linear function to define breakpoints where latent pathogen abruptly change its competitive relationship with others in the panel. These breakpoints, where pathogens start to propagate abnormally, are used as cutoffs to eliminate the influence of contaminations from normal flora. With help of the cutoffs derived from statistical analysis, we are able to identify causative pathogens in 750 (48.92%) patients from qualified patients. In conclusion, qLAMP is a reliable method in quantifying bacterial titer. Despite the fact that there are always latent bacteria contaminated in sputum samples, we can identify causative pathogens based on cutoffs derived from statistical analysis of competitive relationship

    Analysis of the Potential Impacts on China’s Industrial Structure in Energy Consumption

    Industrial structure is one of the main factors that determine energy consumption. Based on China’s energy consumption in 2015 and the goals in 13th Five-Year Plan for Economic and Social Development of the People’s Republic of China (The 13th Five-Year Plan), this paper established an input–output fuzzy multi-objective optimization model to estimate the potential impacts of China’s industrial structure on energy consumption in 2015. Results showed that adjustments to industrial structure could save energy by 19% (1129.17 million ton standard coal equivalent (Mtce)). Second, China’s equipment manufacturing industry has a large potential to save energy. Third, the development of several high energy intensive and high carbon intensive sectors needs to be strictly controlled, including Sector 25 (electricity, heat production, and supply industry), Sector 11 (manufacture of paper and stationery, printing), and Sector 14 (non-metallic mineral products industry). Fourth, the territory industry in China has a great potential for energy saving, while its internal structure still needs to be upgraded. Finally, we provide policy suggestions that may be adopted to reduce energy consumption by adjusting China’s industrial structure

    Comparative Life Cycle Energy and GHG Emission Analysis for BEVs and PhEVs: A Case Study in China

    Battery electric vehicles (BEVs) and plug-in hybrid electric vehicles (PHEVs) are seen as the most promising alternatives to internal combustion vehicles, as a means to reduce the energy consumption and greenhouse gas (GHG) emissions in the transportation sector. To provide the basis for preferable decisions among these vehicle technologies, an environmental benefit evaluation should be conducted. Lithium iron phosphate (LFP) and lithium nickel manganese cobalt oxide (NMC) are two most often applied batteries to power these vehicles. Given this context, this study aims to compare life cycle energy consumption and GHG emissions of BEVs and PHEVs, both of which are powered by LFP and NMC batteries. Furthermore, sensitivity analyses are conducted, concerning electricity generation mix, lifetime mileage, utility factor, and battery recycling. BEVs are found to be less emission-intensive than PHEVs given the existing and near-future electricity generation mix in China, and the energy consumption and GHG emissions of a BEV are about 3.04% (NMC) to 9.57% (LFP) and 15.95% (NMC) to 26.32% (LFP) lower, respectively, than those of a PHEV

    Allocation of Energy Consumption among Provinces in China: A Weighted ZSG-DEA Model

    To realize the sustainable development of energy, the Chinese government has formulated a series of national goals of total energy control and energy structure optimization. Under the national constraints, how to efficiently allocate the constrained total amount of energy consumption to each province is a fundamental problem to be solved. Based on a data envelopment analysis (DEA) model and a zero-sum game theory (ZSG), this paper constructs a weighted zero-sum game data envelopment analysis (ZSG-DEA) model to allocate the energy consumption quota. Additionally, this paper compares the results with the current administrative targets, to examine the efficiency and feasibility of each allocation mechanism. Finally, this paper employs the proposed model to determine the optimal energy structure for each province in China. The results indicate that by 2020, the national goal of energy structure adjustment will be realized, and energy structure will be diversified in most regions, whereas the coal-dominated status in primary energy consumption will not change. Additionally, the weighted ZSG-DEA model focuses on allocation efficiency while the government considers more regional economic disparity. Therefore, this study suggests a mixture of the two allocation mechanisms in accordance with specific conditions

    Oxygen effects on rhamnolipids production by Pseudomonas aeruginosa

    Abstract Background Rhamnolipids are the most extensively studied biosurfactants and has been successfully used in various areas from bioremediation to industrial fields. Rhamnolipids structural composition decide their physicochemical properties. Different physicochemical properties influence their application potential. Rhamnolipids can be produced at both aerobic conditions and anaerobic conditions by Pseudomonas aeruginosa. This study aims to evaluate the oxygen effects on the rhamnolipids yield, structural composition, physicochemical properties and the rhl-genes expression in P. aeruginosa SG. Results will guide researchers to regulate microbial cells to synthesize rhamnolipids with different activity according to diverse application requirements. Results Quantitative real-time PCR analysis revealed that rhlAB genes were down-regulated under anaerobic conditions. Therefore, strain P. aeruginosa SG anaerobically produced less rhamnolipids (0.68 g/L) than that (11.65 g/L) under aerobic conditions when grown in media containing glycerol and nitrate. HPLC–MS analysis showed that aerobically produced rhamnolipids mainly contained Rha-C8-C10, Rha–Rha-C10-C12:1 and Rha–Rha-C8-C10; anaerobically produced rhamnolipids mainly contained Rha-C10-C12 and Rha-C10-C10. Anaerobically produced rhamnolipids contained more mono-rhamnolipids (94.7%) than that (54.8%) in aerobically produced rhamnolipids. rhlC gene was also down-regulated under anaerobic conditions, catalyzing less mono-rhamnolipids to form di-rhamnolipids. Aerobically produced rhamnolipids decreased air–water surface tension (ST) from 72.2 to 27.9 mN/m with critical micelle concentration (CMC) of 60 mg/L; anaerobically produced rhamnolipids reduced ST to 33.1 mN/m with CMC of 80 mg/L. Anaerobically produced rhamnolipids emulsified crude oil with EI24 = 80.3%, and aerobically produced rhamnolipids emulsified crude oil with EI24 = 62.3%. Both two rhamnolipids products retained surface activity (ST  60.0%) under temperatures (4–121 °C), pH values (4–10) and NaCl concentrations less than 90 g/L. Conclusions Oxygen affected the rhl-genes expression in P. aeruginosa, thus altering the rhamnolipids yield, structural composition and physicochemical properties. Rhamnolipids produced at aerobic or anaerobic conditions was structurally distinct. Two rhamnolipids products had different application potential in diverse biotechnologies. Although both rhamnolipids products were thermo-stable and halo-tolerant, aerobically produced rhamnolipids possessed better surface activity, implying its well wetting activity and desorption property; anaerobically produced rhamnolipids exhibited better emulsifying activity, indicating its applicability for enhanced oil recovery and bioremediation of petroleum pollution

    Dynamics of Fractional Vegetation Coverage and Its Relationship with Climate and Human Activities in Inner Mongolia, China

    Long-term remote sensing normalized difference vegetation index (NDVI) datasets have been widely used in monitoring vegetation changes. In this study, the NASA Global Inventory Modeling and Mapping Studies (GIMMS) NDVI3g dataset was used as the data source, and the dimidiate pixel model, intensity analysis, and residual analysis were used to analyze the changes of vegetation coverage in Inner Mongolia—from 1982 to 2010—and their relationships with climate and human activities. This study also explored vegetation changes in Inner Mongolia with respect to natural factors and human activities. The results showed that the estimated vegetation coverage exhibited a high correlation (0.836) with the actual measured values. The increased vegetation coverage area (49.2% of the total area) was larger than the decreased area (43.3%) from the 1980s to the 1990s, whereas the decreased area (57.1%) was larger than the increased area (35.6%) from the 1990s to the early 21st century. This finding indicates that vegetation growth in the 1990s was better than that in the other two decades. Intensity analysis revealed that changes in the average annual rate from the 1990s to the early 21st century were relatively faster than those in the 1980s–1990s. During the 1980s–1990s, the gain of high vegetation coverage areas was active, and the loss was dormant; in contrast, the gain and loss of low vegetation coverage areas were both dormant. In the 1990s to the early 21st century, the gains of high and low vegetation coverage areas were both dormant, whereas the losses were active. During the study period, areas of low vegetation coverage were converted into ones with higher coverage, and areas of high vegetation coverage were converted into ones with lower coverage. The vegetation coverage exhibited a good correlation (R2 = 0.60) with precipitation, and the positively correlated area was larger than the negatively correlated area. Human activities not only promote the vegetation coverage, but also have a destructive effect on vegetation, and the promotion effect during 1982 to 2000 was larger than from 2001 to 2010, while, the destructive effect was larger from 2000 to 2010

    Analysis of Drought Characteristics in Xilingol Grassland of Northern China Based on SPEI and Its Impact on Vegetation

    This research is based on the standardized precipitation evapotranspiration index (SPEI) and normalized difference vegetation index (NDVI) which represent the drought and vegetation condition on land. Take the linear regression method and Pearson correlation analysis to study the spatial and temporal evolution of SPEI and NDVI and the drought effect on vegetation. The results show that (1) during 1961–2015, SPEI values at different time scales showed a downward trend; SPEI-12 has a mutation in 1997 and the SPEI value significantly decreased after this year. (2) During 2000–2015, the annual growing season SPEI has an obvious upward trend in time and the apparent wetting spatially. (3) In the recent 16 years, the growing season NDVI showed an upward trend and more than 80% of the total area’s vegetation increased in Xilingol. (4) Vegetation coverage in Xilingol grew better in humid years and opposite in arid years. SPEI and NDVI had a significant positive correlation; 98% of the region showed positive correlation, indicating that meteorological drought affects vegetation growth more in arid and semiarid region.
