    Learning to Hash-tag Videos with Tag2Vec

    User-given tags or labels are valuable resources for semantic understanding of visual media such as images and videos. Recently, a new type of labeling mechanism known as hash-tags have become increasingly popular on social media sites. In this paper, we study the problem of generating relevant and useful hash-tags for short video clips. Traditional data-driven approaches for tag enrichment and recommendation use direct visual similarity for label transfer and propagation. We attempt to learn a direct low-cost mapping from video to hash-tags using a two step training process. We first employ a natural language processing (NLP) technique, skip-gram models with neural network training to learn a low-dimensional vector representation of hash-tags (Tag2Vec) using a corpus of 10 million hash-tags. We then train an embedding function to map video features to the low-dimensional Tag2vec space. We learn this embedding for 29 categories of short video clips with hash-tags. A query video without any tag-information can then be directly mapped to the vector space of tags using the learned embedding and relevant tags can be found by performing a simple nearest-neighbor retrieval in the Tag2Vec space. We validate the relevance of the tags suggested by our system qualitatively and quantitatively with a user study

    Complex regulation of neutrophil-derived MMP-9 secretion in central nervous system tuberculosis.

    BACKGROUND: Central nervous system tuberculosis (CNS-TB) may be fatal even with treatment. Neutrophils are the key mediators of TB immunopathology, and raised CSF matrix metalloproteinase-9 (MMP-9) which correlates to neutrophil count in CNS-TB is associated with neurological deficit and death. The mechanisms by which neutrophils drive TB-associated CNS matrix destruction are not clearly defined. METHODS: Human brain biopsies with histologically proven CNS-TB were stained for neutrophils, neutrophil elastase, and MMP-9. Neutrophil MMP-9 secretion and gene expression were analyzed using Luminex and real-time PCR. Type IV collagen degradation was evaluated using confocal microscopy and quantitative fluorescent assays. Intracellular signaling pathways were investigated by immunoblotting and chemical inhibitors. RESULTS: MMP-9-expressing neutrophils were present in tuberculous granulomas in CNS-TB and neutrophil-derived MMP-9 secretion was upregulated by Mycobacterium tuberculosis (M.tb). Concurrent direct stimulation by M.tb and activation via monocyte-dependent networks had an additive effect on neutrophil MMP-9 secretion. Destruction of type IV collagen, a key component of the blood-brain barrier, was inhibited by neutralizing neutrophil MMP-9. Monocyte-neutrophil networks driving MMP-9 secretion in TB were regulated by MAP-kinase and Akt-PI3 kinase pathways and the transcription factor NF-kB. TNFα neutralization suppressed MMP-9 secretion to baseline while dexamethasone did not. CONCLUSIONS: Multiple signaling paths regulate neutrophil-derived MMP-9 secretion, which is increased in CNS-TB. These paths may be better targets for host-directed therapies than steroids currently used in CNS-TB

    Online user-generated content (UGC) includes various forms of computer-mediated online reviews, ratings, and feedback. The impact of online consumer reviews has been widely studied particularly in e-commerce, online marketing, and consumer behavior domains using text-mining and sentiment analysis. Such studies often assume that consumer-submitted online reviews truly represent consumer experiences, but multiple studies on online social networks suggest otherwise. Drawing from the social network literature, this paper investigates the impact of peers on consumer willingness to write and submit online reviews. An empirical study using data from “Yelp,” a globally used online restaurant review website, shows that the number of friends and fans positively impacts the number of online consumer reviews written. Implications for research and practice are discussed

    Arf GTPase interplay with Rho GTPases in regulation of the actin cytoskeleton

    The Arf and Rho subfamilies of small GTPases are nucleotide-dependent molecular switches that act as master regulators of vesicular trafficking and the actin cytoskeleton organization. Small GTPases control cell processes with high fidelity by acting through distinct repertoires of binding partners called effectors. While we understand a great deal about how these GTPases act individually, relatively little is known about how they cooperate, especially in the control of effectors. This review highlights how Arf GTPases collaborate with Rac1 to regulate actin cytoskeleton dynamics at the membrane via recruiting and activating the Wave Regulatory Complex (WRC), a Rho effector that underpins lamellipodia formation and macropinocytosis. This provides insight into Arf regulation of the actin cytoskeleton, whilst putting the spotlight on small GTPase cooperation with emerging evidence of its importance in fundamental cell biology and interactions with pathogenic bacteria

    RepSeq-A database of amino acid repeats present in lower eukaryotic pathogens

    BACKGROUND Amino acid repeat-containing proteins have a broad range of functions and their identification is of relevance to many experimental biologists. In human-infective protozoan parasites (such as the Kinetoplastid and Plasmodium species), they are implicated in immune evasion and have been shown to influence virulence and pathogenicity. RepSeq http://repseq.gugbe.com is a new database of amino acid repeat-containing proteins found in lower eukaryotic pathogens. The RepSeq database is accessed via a web-based application which also provides links to related online tools and databases for further analyses. RESULTS The RepSeq algorithm typically identifies more than 98% of repeat-containing proteins and is capable of identifying both perfect and mismatch repeats. The proportion of proteins that contain repeat elements varies greatly between different families and even species (3 - 35% of the total protein content). The most common motif type is the Sequence Repeat Region (SRR) - a repeated motif containing multiple different amino acid types. Proteins containing Single Amino Acid Repeats (SAARs) and Di-Peptide Repeats (DPRs) typically account for 0.5 - 1.0% of the total protein number. Notable exceptions are P. falciparum and D. discoideum, in which 33.67% and 34.28% respectively of the predicted proteomes consist of repeat-containing proteins. These numbers are due to large insertions of low complexity single and multi-codon repeat regions. CONCLUSION The RepSeq database provides a repository for repeat-containing proteins found in parasitic protozoa. The database allows for both individual and cross-species proteome analyses and also allows users to upload sequences of interest for analysis by the RepSeq algorithm. Identification of repeat-containing proteins provides researchers with a defined subset of proteins which can be analysed by expression profiling and functional characterisation, thereby facilitating study of pathogenicity and virulence factors in the parasitic protozoa. While primarily designed for kinetoplastid work, the RepSeq algorithm and database retain full functionality when used to analyse other species

    A novel inhaled phosphodiesterase 4 inhibitor (CHF6001) reduces the allergen challenge response in asthmatic patients

    CHF6001 is an inhaled phosphodiesterase 4 (PDE4) inhibitor in development for the treatment of obstructive lung diseases. We investigated the efficacy and safety of CHF6001 using the allergen challenge model in a double blind, placebo controlled, 3-way cross-over study. Thirty six atopic asthmatics who were not taking inhaled corticosteroids and who demonstrated a late asthmatic response (LAR) to inhaled allergen at screening were randomised to receive CHF6001 400 μg or 1200 μg or placebo administered once a day using a dry powder inhaler. The three treatment periods were 9 days; allergen challenges were performed on day 9 and induced sputum was obtained after 10 h from challenge. Washout periods between treatments were up to 5 weeks. Both CHF6001 doses significantly attenuated the LAR; the primary endpoint analysis showed that CHF6001 400 μg and 1200 μg caused reductions of 19.7% (p = 0.015) and 28.2% (p < 0.001) respectively of the weighted FEV1 AUC4-10h compared with placebo. The difference between the CHF6001 doses was not statistically significant (p = 0.2). Compared with placebo, CHF 6001 caused greater reduction in sputum eosinophil counts, although these changes were not statistically significant. CHF6001 was well tolerated, with similar numbers of adverse events in each treatment period. This inhaled PDE4 inhibitor has the potential to provide clinical benefits in patients with atopic asthma

    Ethnic differences in BMI among Dutch adolescents: what is the role of screen-viewing, active commuting to school, and consumption of soft drinks and high-caloric snacks?

    <p>Abstract</p> <p>Background</p> <p>The threats posed by the rising prevalence of overweight and obesity on public health have been widely acknowledged. Several population groups, which deserve special attention because of their higher prevalence rates, have been identified. These include adolescents and ethnic sub-groups. The aim of the present study was twofold: (1) to assess ethnic differences in body mass index (BMI) and in behaviours that are related to both energy intake and energy expenditure, and (2) to examine whether these behaviours explain the relationship between ethnicity and BMI.</p> <p>Methods</p> <p>We conducted a cross-sectional data analysis among 957 Dutch adolescents (mean age = 12.7 years). Body height and weight were measured using a standardized protocol. Adolescents completed a questionnaire on screen-viewing behaviour, physical activity, consumption of sugar-containing beverages, and consumption of high-caloric snacks.</p> <p>Results</p> <p>In our study sample 121 adolescents (= 13%) were of Non-Western origin. BMI was significantly higher in Non-Western adolescents (boys: 19.9 kg/m<sup>2</sup>, SD = 3.0, girls: 20.9 kg/m<sup>2</sup>, SD = 3.8) compared to Dutch adolescents (boys: 18.4 kg/m<sup>2</sup>, SD = 2.8, girls: 19.0 kg/m<sup>2</sup>, SD = 3.0). Our results show that time spent on television viewing, active commuting to school, and consumption of fruit juices partially mediated the association between BMI and ethnicity.</p> <p>Conclusion</p> <p>Behaviours related to both energy expenditure and energy intake may contribute to the ethnic differences in BMI in adolescents and should be considered when tailoring overweight prevention programs to ethnic subpopulations.</p> <p>Trial registration</p> <p>International Standard Randomised Controlled Trial Number ISRCTN87127361</p

    The impact of product attributes and emerging technologies on firms’ international configuration

    International business literature has largely explained the international dispersion of firms’ activities as a choice based on trade-offs between cost minimisation, knowledge seeking, managing transaction costs and maintaining control. By incorporating insights from operations management, we propose a framework that explicitly takes into account products’ physical and knowledge attributes that constrain the viable international configuration options available to firms. Linking the characteristics of a product to the scope for horizontal and vertical decoupling in a value network allows us to re-frame recent discussions in the literature about fragmentation of activities vs tasks and to develop an overall picture of the way industry-specific peculiarities characterise (and also constrain) viable international configurations. We show how our framework can be used to interpret data on the scope for decoupling and dispersion collected from industry experts and elucidate the relationships between configuration options and measures of product characteristics. We then utilise this framework to predict how emerging technologies will reshape the international configuration options available to firms.EPSR
