Palladium-catalyzed domino carbopalladation/5-exo-allylic amination of \u3b1-amino allenamides: an efficient entry to enantiopure imidazolidinones
Allenamides of alpha-amino acids were converted into enantiopure 2-vinylimidazolidin-4-ones by a carbopalladation/exo-cyclization process. The products were obtained in 2.5:1-5.5:1 dr, with 94-99% ee. The palladium-catalyzed carbonylative cyclization of the same substrates afforded enone structures. Starting from properly substituted allenamides, an intramolecular carbopalladation followed by intramolecular amination gave rise to tricyclic fused-ring imidazolidinones
Efficacy of Esthetic Retainers: Clinical Comparison between Multistranded Wires and Direct-Bond Glass Fiber-Reinforced Composite Splints
The purpose of this longitudinal prospective randomized study was to evaluate the reliability of two different types of orthodontic retainers in clinical use: a multistrand stainless steel wire and a polyethylene ribbon-reinforced resin composite. Moreover the level of satisfaction of the patient about the esthetic result was also analyzed by means of a Visual Analogue Scale (VAS). 34 patients (9 boys and 25 girls, mean age 14.3), in the finishing phase of orthodontic treatment, were selected for the study. Since splints were applied the number, cause, and date of splint failures were recorded for each single tooth over 12 months. Statistical analysis was performed using a paired -test, Kaplan Meier survival estimates, and the log-rank test. Kruskal Wallis test was performed to analyze VAS recordings. Differences between the bond failure rates were not statistically significant. Esthetic result of VAS was significantly higher for polyethylene ribbon-reinforced resin retainers than for stainless steel wires
Glycosylation tunes neuroserpin physiological and pathological properties
Neuroserpin (NS) is a member of the serine protease inhibitors superfamily. Specific point
mutations are responsible for its accumulation in the endoplasmic reticulum of neurons that leads to
a pathological condition named familial encephalopathy with neuroserpin inclusion bodies (FENIB).
Wild-type NS presents two N-glycosylation chains and does not form polymers in vivo, while
non-glycosylated NS causes aberrant polymer accumulation in cell models. To date, all in vitro
studies have been conducted on bacterially expressed NS, de facto neglecting the role of glycosylation
in the biochemical properties of NS. Here, we report the expression and purification of human
glycosylated NS (gNS) using a novel eukaryotic expression system, LEXSY. Our results confirm the
correct N-glycosylation of wild-type gNS. The fold and stability of gNS are not altered compared
to bacterially expressed NS, as demonstrated by the circular dichroism and intrinsic tryptophan
fluorescence assays. Intriguingly, gNS displays a remarkably reduced polymerisation propensity
compared to non-glycosylated NS, in keeping with what was previously observed for wild-type NS
in vivo and in cell models. Thus, our results support the relevance of gNS as a new in vitro tool to
study the molecular bases of FENIB
Sbiancamento dentale postortodontico in pazienti adulti
Scopo del lavoro. Valutare i risultati ottenuti in seguito all’applicazione di un nuovo prodotto
per lo sbiancamento dentale professionale in un paziente adulto, precedentemente sottoposto
a trattamento ortodontico al fine di migliorare l’estetica del sorriso.
Materiali e metodi. Il prodotto utilizzato in questo studio è Whiteness HP Maxx, un gel a base
di perossido d’idrogeno (30-35 per cento), dotato di elevato potere sbiancante, utilizzabile
sia da solo sia con l’ausilio di vari tipi di sorgenti luminose.
Risultati. Nel caso clinico esaminato, i risultati ottenuti con Whiteness HP Maxx sembrano
molto positivi: infatti è stata sufficiente una sola applicazione di gel a seduta, per un totale di
due sedute, per raggiungere un risultato giudicato molto soddisfacente dal paziente e dall’odontoiatra.
Conclusioni. È possibile affermare che il prodotto Whiteness HP Maxx è efficace nello sbiancamento
vitale dopo trattamento ortodontico fisso. Il ridotto numero di applicazioni necessarie
per ottenere tale risultato, in aggiunta all’assenza di effetti collaterali (ipersensibilità),
rappresentano sicuramente dati positivi nella valutazione complessiva dell’agente sbiancante
