    Yield improvement of exopolysaccharides by screening of the Lactobacillus acidophilus ATCC and optimization of the fermentation and extraction conditions

    Exopolysacharides (EPS) produced by Lactobacillus acidophilus play an important role in food processing with its well-recognized antioxidant activity. In this study, a L. acidophilus mutant strain with high-yielding EPS (2.92±0.05 g/L) was screened by chemical mutation (0.2 % diethyl sulfate). Plackett-Burman (PB) design and response surface methodology (RSM) were applied to optimize the EPS fermentation parameters and central composite design (CCD) was used to optimize the EPS extraction parameters. A strain with high-yielding EPS was screened. It was revealed that three parameters (Tween 80, dipotassium hydrogen phosphate and trisodium citrate) had significant influence (P < 0.05) on the EPS yield. The optimal culture conditions for EPS production were: Tween 80 0.6 mL, dipotassium hydrogen phosphate 3.6 g and trisodium citrate 4.1 g (with culture volume of 1 L). In these conditions, the maximum EPS yield was 3.96±0.08 g/L. The optimal extraction conditions analyzed by CCD were: alcohol concentration 70 %, the ratio of material to liquid (M/L ratio) 1:3.6 and the extraction time 31 h. In these conditions, the maximum EPS extraction yield was 1.48±0.23 g/L. It was confirmed by the verification experiments that the EPS yield from L. acidophilus mutant strains reached 5.12±0.73 g/L under the optimized fermentation and extraction conditions, which was 3.8 times higher than that of the control (1.05±0.06 g/L). The results indicated that the strain screening with high-yielding EPS was successful and the optimized fermentation and extraction conditions significantly enhanced EPS yield.     Abstract-A 5-DoF 3R2T (three dimensional rotation and two dimensional translation degrees of freedom) fully-symmetrical parallel manipulator can be adopted in many applications such as simulating the motion of spinal column. However, kinematics of this type parallel manipulator has not been studied enough because of short history. The study of kinematics of the manipulators leads inevitably to the problem of singular configuration. Singularity of a 5-DoF 3R2T fully-symmetrical parallel manipulator, 5-RRR(RR), is illustrated in this study. According to the singularity classification by Fang and Tsai, both limb singularity and actuation singularity are illustrated by screw theory and Grassmann geometry. The result of this study will be helpful for singularity analysis of 5-DoF 3R2T fully-symmetrical parallel manipulators because of their similar constraint property

    Removal of point source leakage from time-order data filtering

    Time-ordered data (TOD) from ground-based CMB experiments are generally filtered before map-making to remove or reduce the contamination from the ground and the atmospheric emissions. However, when the observation region contains strong point sources, the filtering process will result in considerable leakage around the point sources in a measured CMB map, and leave spurious polarization signals. Therefore, such signals need to be assessed and removed before CMB science exploitation. In this work, we present a new method that we call "template fitting" and can effectively remove these leakage signals in pixel domain, not only satisfying the requirement for measuring primordial gravitational waves from CMB-BB modes, but also avoiding time-consuming operations on TOD.Comment: 13pages, 6 figures, 5 table

    Alteration in the sensitivity to crizotinib by Na+/H+ exchanger regulatory factor 1 is dependent to its subcellular localization in ALK-positive lung cancers

    BackgroundNa+/H+ exchanger regulatory factor 1 (NHERF1) is an important scaffold protein participates in the modulation of a variety of intracellular signal pathways. NHERF1 was able to enhance the effects of chemo-drugs in breast and cervical cancer cells. Anaplastic lymphoma kinase (ALK) fusion mutations are validated molecules targeted therapy in lung cancers, where crizotinib can be used as the specific inhibitor to suppress tumor progression. However, due to the less frequent occurrence of ALK mutations and the complexity for factors to determine drug responses, the genes that could alter crizotinib sensitivity are unclear.MethodsBoth ALK-translocated and ALK-negative lung adenocarcinoma specimens in tissue sections were collected for immunohistochemistry. The possible mechanisms of NHERF1 and its role in the cell sensitivity to crizotinib were investigated using an ALK-positive and crizotinib-sensitive lung adenocarcinoma cell line H3122. Either a NHERF1 overexpression vector or agents for NHERF1 knockdown was used for crizotinib sensitivity measures, in association with cell viability and apoptosis assays.ResultsThe expression level of NHERF1 in ALK-translocated NSCLC was significantly higher than that in other lung cancer tissues. NHERF1 expression in ALK positive lung cancer cells was regulated by ALK activities, and was in return able to alter the sensitivity to crizotinib. The function of NHERF1 to influence crizotinib sensitivity was depending on its subcellular distribution in cytosol instead of its nucleus localized form.ConclusionEctopically overexpressed NHERF1 could be a functional protein for consideration to suppress lung cancers. The determination of NHERF1 levels in ALK positive NSCLC tissues might be useful to predict crizotinib resistance, especially by distinguishing cytosolic or nuclear localized NHERF1 for the overexpressed molecules

    Cognitive Representation Learning of Self-Media Online Article Quality

    The automatic quality assessment of self-media online articles is an urgent and new issue, which is of great value to the online recommendation and search. Different from traditional and well-formed articles, self-media online articles are mainly created by users, which have the appearance characteristics of different text levels and multi-modal hybrid editing, along with the potential characteristics of diverse content, different styles, large semantic spans and good interactive experience requirements. To solve these challenges, we establish a joint model CoQAN in combination with the layout organization, writing characteristics and text semantics, designing different representation learning subnetworks, especially for the feature learning process and interactive reading habits on mobile terminals. It is more consistent with the cognitive style of expressing an expert's evaluation of articles. We have also constructed a large scale real-world assessment dataset. Extensive experimental results show that the proposed framework significantly outperforms state-of-the-art methods, and effectively learns and integrates different factors of the online article quality assessment.Comment: Accepted at the Proceedings of the 28th ACM International Conference on Multimedi
