    Sumptuary Legislation and the Fabric Construction of National Identity in Early Modern England

    The sumptuary legislation is quite specific, outlining exactly which fabrics were permitted for each social class and in what types of clothing. For example, Elizabeth’s 1562 proclamation asserted, “None shall wear in his apparel any silk of the color purple, cloth of gold tissue, but only the King, Queen . . . except dukes and marquises who may wear in doublets and sleeveless coats cloth of gold of tissue not exceeding £5 the yard, and purple in mantles of the Garter.” The language of exclusion reflects the common conceptions of social hierarchy in early modern England. The most lavish accoutrements were reserved for the royal family, including the traditional imperial colors of purple and gold, although certain levels of nobility were allowed to sparingly use those colors. Expressing the desire to halt the use of apparel as a tool for untoward social imitation, writer William Prynne called for a law regulating apparel, not only in the playhouse, but also throughout society “which would well befit our Nation, our times, which Proteus-like are always changing shape and fashion, and like the Moone, appeare from day to day in different formes.

    In Shakespeare\u27s canon, Twelfth Night is considered one of his great comedies. According to formalist critic Milton Crane, The great comedies such as Twelfth Night show ... Shakespeare working effectively within the tradition of classical comedy and enlarging it to encompass a rich and harmonious development of fundamentally comic matter (Crane 8). However, Crane\u27s conclusion is problematic in that it does not reconcile individual\u27s romantic fulfillment with an overarching resolution for the play. Antonio, Malvolio, and Feste are three prominent characters in this comedy who are not comic. Antonio\u27s love for Sebastian leads him into danger, and the audience never knows his final fate. The trick played on Malvolio is initially amusing, but quickly it becomes unsavory mistreatment. Feste, the clown of the play, is rarely funny in a traditional sense, instead performing a melancholic truth-telling role. It is not only these characters who contribute to the problematic ending: the three marriages are also inherently unstable. The cases of mistaken and hidden identities create romantic pairings based on a troubling foundation of deception. Despite its comedic elements, the ambiguous ending of Twelfth Night is not decisive or satisfying due to a lack of resolution in the case of characters such as Antonio, Malvolio, and Feste, and due to underlying problems in the three marriages

    Neurovirulent Vaccine-Derived Polioviruses in Sewage from Highly Immune Populations

    BACKGROUND: Vaccine-derived polioviruses (VDPVs) have caused poliomyelitis outbreaks in communities with sub-optimal vaccination. Israeli environmental surveillance of sewage from populations with high (>95%) documented vaccine coverage of confirmed efficacy identified two separate evolutionary clusters of VDPVs: Group 1 (1998–2005, one system, population 1.6×10(6)) and Group 2 (2006, 2 systems, populations 0.7×10(6) and 5×10(4)). PRINCIPAL FINDINGS: Molecular analyses support evolution of nine Group 1 VDPVs along five different lineages, starting from a common ancestral type 2 vaccine-derived Sabin-2/Sabin-1 recombinant strain, and independent evolution of three Group 2 VDPVs along one lineage starting from a different recombinant strain. The primary evidence for two independent origins was based on comparison of unique recombination fingerprints, the number and distribution of identical substitutions, and evolutionary rates. Geometric mean titers of neutralizing antibodies against Group 1 VDPVs were significantly lower than against vaccine strains in all age-group cohorts tested. All individuals had neutralizing titers >1∶8 against these VDPVs except 7% of the 20–50 year cohort. Group 1 VDPVs were highly neurovirulent in a transgenic mouse model. Intermediate levels of protective immunity against Group 2 VDPVs correlated with fewer (5.0+1.0) amino acid substitutions in neutralizing antigenic sites than in Group 1 VDPV's (12.1±1.5). SIGNIFICANCE: VDPVs that revert from live oral attenuated vaccines and reacquire characteristics of wild-type polioviruses not only threaten populations with poor immune coverage, but are also a potential source for re-introduction of poliomyelitis into highly immune populations through older individuals with waning immunity. The presence of two independently evolved groups of VDPVs in Israel and the growing number of reports of environmental VDPV elsewhere make it imperative to determine the global frequency of environmental VDPV. Our study underscores the importance of the environmental surveillance and the need to reconsider the global strategies for polio eradication and the proposed cessation of vaccination

    Regulation of adipose tissue inflammation by interleukin 6

    Obesity is associated with a chronic state of low-grade inflammation and progressive tissue infiltration by immune cells and increased expression of inflammatory cytokines. It is established that interleukin 6 (IL6) regulates multiple aspects of metabolism, including glucose disposal, lipolysis, oxidative metabolism, and energy expenditure. IL6 is secreted by many tissues, but the role of individual cell types is unclear. We tested the role of specific cells using a mouse model with conditional expression of the Il6 gene. We found that IL6 derived from adipocytes increased, while IL6 derived from myeloid cells and muscle suppressed, macrophage infiltration of adipose tissue. These opposite actions were associated with a switch of IL6 signaling from a canonical mode (myeloid cells) to a noncanonical trans-signaling mode (adipocytes and muscle) with increased expression of the ADAM10/17 metalloprotease that promotes trans-signaling by the soluble IL6 receptor alpha. Collectively, these data demonstrate that the source of IL6 production plays a major role in the physiological regulation of metabolism

    A feed-forward regulatory loop in adipose tissue promotes signaling by the hepatokine FGF21

    The cJun NH2-terminal kinase (JNK) signaling pathway is activated by metabolic stress and promotes the development of metabolic syndrome, including hyperglycemia, hyperlipidemia, and insulin resistance. This integrated physiological response involves cross-talk between different organs. Here we demonstrate that JNK signaling in adipocytes causes an increased circulating concentration of the hepatokine fibroblast growth factor 21 (FGF21) that regulates systemic metabolism. The mechanism of organ crosstalk is mediated by a feed-forward regulatory loop caused by JNK-regulated FGF21 autocrine signaling in adipocytes that promotes increased expression of the adipokine adiponectin and subsequent hepatic expression of the hormone FGF21. The mechanism of organ cross-talk places circulating adiponectin downstream of autocrine FGF21 expressed by adipocytes and upstream of endocrine FGF21 expressed by hepatocytes. This regulatory loop represents a novel signaling paradigm that connects autocrine and endocrine signaling modes of the same hormone in different tissues

    Evolution of echovirus 11 in a chronically infected immunodeficient patient.

    Deep sequencing was used to determine complete nucleotide sequences of echovirus 11 (EV11) strains isolated from a chronically infected patient with CVID as well as from cases of acute enterovirus infection. Phylogenetic analysis showed that EV11 strains that circulated in Israel in 1980-90s could be divided into four clades. EV11 strains isolated from a chronically infected individual belonged to one of the four clades and over a period of 4 years accumulated mutations at a relatively constant rate. Extrapolation of mutations accumulation curve into the past suggested that the individual was infected with circulating EV11 in the first half of 1990s. Genomic regions coding for individual viral proteins did not appear to be under strong selective pressure except for protease 3C that was remarkably conserved. This may suggest its important role in maintaining persistent infection

    Reversal of Hypertriglyceridemia, Fatty Liver Disease, and Insulin Resistance by a Liver-Targeted Mitochondrial Uncoupler

    SummaryNonalcoholic fatty liver disease (NAFLD) affects one in three Americans and is a major predisposing condition for the metabolic syndrome and type 2 diabetes (T2D). We examined whether a functionally liver-targeted derivative of 2,4-dinitrophenol (DNP), DNP-methyl ether (DNPME), could safely decrease hypertriglyceridemia, NAFLD, and insulin resistance without systemic toxicities. Treatment with DNPME reversed hypertriglyceridemia, fatty liver, and whole-body insulin resistance in high-fat-fed rats and decreased hyperglycemia in a rat model of T2D with a wide therapeutic index. The reversal of liver and muscle insulin resistance was associated with reductions in tissue diacylglycerol content and reductions in protein kinase C epsilon (PKCε) and PKCθ activity in liver and muscle, respectively. These results demonstrate that the beneficial effects of DNP on hypertriglyceridemia, fatty liver, and insulin resistance can be dissociated from systemic toxicities and suggest the potential utility of liver-targeted mitochondrial uncoupling agents for the treatment of hypertriglyceridemia, NAFLD, metabolic syndrome, and T2D
