    Promoter-Level Transcriptome Identifies Stemness Associated With Relatively High Proliferation in Pancreatic Cancer Cells

    Both pancreatic intraepithelial neoplasia (PanIN), a frequent precursor of pancreatic cancer, and intraductal papillary mucinous neoplasm (IPMN), a less common precursor, undergo several phases of molecular conversions and finally develop into highly malignant solid tumors with negative effects on the quality of life. We approached this long-standing issue by examining the following PanIN/IPMN cell lines derived from mouse models of pancreatic cancer: Ptf1a-Cre; KrasG12D; p53f/+ and Ptf1a-Cre; KrasG12D; and Brg1f/f pancreatic ductal adenocarcinomas (PDAs). The mRNA from these cells was subjected to a cap analysis of gene expression (CAGE) to map the transcription starting sites and quantify the expression of promoters across the genome. Two RNA samples extracted from three individual subcutaneous tumors generated by the transplantation of PanIN or IPMN cancer cell lines were used to generate libraries and Illumina Seq, with four RNA samples in total, to depict discrete transcriptional network between IPMN and PanIN. Moreover, in IPMN cells, the transcriptome tended to be enriched for suppressive and inhibitory biological processes. In contrast, the transcriptome of PanIN cells exhibited properties of stemness. Notably, the proliferation capacity of the latter cells in culture was only minimally constrained by well-known chemotherapy drugs such as GSK690693 and gemcitabine. The various transcriptional factor network systems detected in PanIN and IPMN cells reflect the distinct molecular profiles of these cell types. Further, we hope that these findings will enhance our mechanistic understanding of the characteristic molecular alterations underlying pancreatic cancer precursors.     We studied the consciousness about weight reduction behavior, dietary habits and motivation to do sports of young females by using a self-report questionnaire survey. The subjects were 386 female college students (fiscal 2007, Grade 1, 18.6±0.6 years old) in Nishinomiya district. Although their BMIs were almost within normal range( 20.2±2.0), 84% of female students had a desire to get slimmer. Most of female students were aware of the necessity of daily exercise, and recognized that the amount of their exercise had been inadequate or unsatisfactory so far

Although its significance is often overlooked, it is associated with poor QOL, and decreased pulmonary and digestive function. Thus osteoporotic fracture is a preventive disease. Unfortunately, the significance of osteoporosis is not fully acknowledged in the general population.     Radiation Myelopathy Caused by Palliative Radiotherapy and Intrathecal Methotrexate

    Radiation myelopathy is a rare, late-stage adverse event that develops following irradiation at or above 50 Gy. Here, we report a case of irreversible paraplegia caused by palliative radiation (20 Gy in 5 fractions) to the spinal cord combined with intrathecal methotrexate (IT-MTX). A 69-year-old man presented with back pain, prompting a diagnosis of acute myeloid leukemia. At the first visit, he complained of muscle weakness and hypoesthesia in both legs; spinal magnetic resonance imaging (MRI) revealed an epidural mass compressing the spinal cord at the fifth to seventh level of the thoracic vertebrae. This was considered to be an extramedullary lesion of leukemia, and he received remission induction therapy including IT-MTX; palliative radiation (20 Gy in 5 fractions) of the epidural mass was initiated the following day. Then, during the course of consolidation therapy, a second IT-MTX was performed after 1 month and a third after 3 months. While the consolidation therapy was complete, yielding remission, he developed sudden paraplegia, as well as bladder and bowel dysfunction (BBD), 10 months later. Spinal MRI showed extensive intramedullary high signal intensity on T2-weighted image, including the irradiation field. It was thought myelopathy was due to irradiation of the spinal cord combined with IT-MTX. He immediately received steroid pulse therapy; however, the paraplegia and BBD did not improve. It is extremely rare for irreversible radiation myelopathy to occur with IT-MTX and palliative radiation to the spinal cord. We believe that even with low-dose palliative radiation, caution is required for combined use with IT-MTX

    We evaluated the physiological activity of soybean fermented with mushroom mycelia.Without heating,fibrinolytic activity was observed in Schizophyllum commune(1418mm2),Pleurotus cornucopiae( 1635mm2),Hericium ramosum(1608mm2)and NAPA(Ganodermataceae produced in Thailand) (1284mm2). The cell-free crude extract form Schizophyllum commune(563mm2)and Pleurotus cornucopiae( 369mm2)showed the fibrinolytic activity.Concerning the antithrombin activity,the thrombin clotting time was more than 600 seconds for Coriolus versicolor,Pleurotus cornucopiae,Lenzites betulina,Wolfiporia cocos,Schizophyllum commune, NAPA, Hericium erinaceum,Wynnea gigantea, Ramaria botrytis,Cordyceps militaris,Hericium ramosum and Stropharia rugosoannulata. Even after heating,Wolfiporia cocos and Macrolepiota procera showed the clotting time of more than 600 seconds. Compared with the blank(soybeans before fermentation),the total amino acid level increased to 38-fold for Schizophyllum commune and Pleurotus cornucopiae,to 20-fold for Hericium ramosum and to 15-fold for NAPA. In addition,the possible conversion from the glycoside type of isoflavone to the aglycon type was suggested

    The fusion method using PEG as a fusogenic agent has a number of disadvantages in comparison with the newly developed electrical fusion method. However, there has been little application yet of this electrical fusion method to the fusion of protoplasts of mushrooms. In this report, we describe the pearl chain formation and effect of pulse field strength on protoplast breakdown of protoplasts prepared from Hericium erinaceum. The protoplasts density favorable for pearl chain formation was about 10^8protoplasts/ml, and the yield of single pairs was maximum (30%) after 60sec dielectrophoresis at 100V/cm and 1MHz. The field intensity required to decompose half of the protoplast from H. erinaceum was 8kV/cm in a 0.7M mannitol solution

    Utilization of Enokitake and Shiitake in fiber-bread processing and their characteristics

    Bread containing 5% enokitake (Flammulina velutipes) produced manually the expanded most, however, in the case of shiitake (Lentinus edodes), the loaf volume of bread decreased with an increasing concentration of shiitake. In the case of dough without sugar, the bread containing 10% enokitake the expanded most, however, that of more than 10% enokitake decreased with an increasing concentration of enokitake. The loaf volume of shiitake bread without sugar also decreased with an increasing concentration of shiitake because shiitake strongly inhibited the growth of baker\u27s yeast. The firmness of the bread increased with an increasing concentration of enokitake or shiitake until the concentration was 30%. Fermentable sugars such as glucose, mannitol, maltose and trehalose from enokitake and glucose from shiitake were consumed with fermentation by yeast (Saccharomyces cerevisiae) during the first 4 hours of incubation
