    Short-term effects of Theracurmin dose and exercise type on pain, walking ability, and muscle function in patients with knee osteoarthritis

    The purpose of this study was to investigate the short-term of Theracurmin dose and exercise type on pain, walking ability, and muscle function in patients with knee osteoarthritis. Twenty-five patients with knee osteoarthritis randomly selected to Theracurmin intake (T) group and Theracurmin in combined with exercise (T+E) group. T group (n= 13) was taken orally a capsule of 700 mg, 3 times per day, (total 2,100 mg, 35 mg/kg-body weight). T+E group (n= 12) performed aerobic training of 30-min walking and weight training for increasing leg muscular strength. After treatment, the number of steps, muscle mass, range of motion of knee, and the muscle strength in flexion and extension significantly increased. The percent body fat, visual analogue scale, The Western Ontario and McMaster score, centers of pressure with closed eye, 10-m walking ability, stair ascending speed were significantly decreased after treatment. Although no difference observed between the T and T+E groups, the 4-week intake of Theracurmin with and without exercise appeared to be effective in reducing the pain and enhancing muscular and balancing function. Therefore, Theracurmin intake for early symptoms and additional exercise as symptoms alleviate might be an effective way of delaying and managing osteoarthritis, and additional studies investigating the effects of Theracurmin and exercise on osteoarthritis could be beneficial

    Bone regeneration potential of sub-microfibrous membranes with osteogenic induction of rBMSC for tissue engineering

    Purpose: To examine the biocompatibility and osteoinductive potential ofĀ  sub-microfibrous membranes with cells in vitro and in vivo.Methods: Polylactic acid (PLA) and poly-Īµ-caprolactone (PCL) were blended at various volume ratios (PLA:PCL = 100:0, 70:30, 50:50, 30:70 and 0:100) and each membrane form was prepared by electrospinning. Cell viability,Ā  biocompatibility, and bone regeneration were measured.Results: The membranes from the PLA/PCL blends prepared by an electrospinning process showed a range of diameter distribution ranging from 1,580 to 550 nm. The cells of 100 % PCL membrane (smallest diameter) exhibited significantly higher adhesion and proliferation than those of the other membranes. Among theĀ  membranes from PLA/PCL blends, PCL membrane showed weak inflammatory changes in the early stages of implantation without acute or chronic inflammation. PCL membranes with osteogenically-induced cells successfully stimulated new bone formation in a rate calvarial defect model.Conclusion: The results indicate that biodegradable PCL sub-microfibrous membrane produced by electrospinning process seems to have excellent biocompatibility, and may be used as a scaffold for bone tissue engineering.Keywords: Biocompatibility, Hard tissue, Biomaterial availability, Bone remodeling, Polylactic acid, Poly-Īµ-caprolactone, Osteoinductive potential, Sub-microfibrous membrane

The next generation of recently introduced TVT-SECUR is intended to minimize the incidence of complications. Herein we report a case of internal pudendal artery injury sustained during this procedure that was successfully treated by radiological embolization. Angiography with vessel embolization, when available, should be considered when the arterial injury is suspected

    Evaluation of Food Web Structure and Complexity in the Process of Kelp Bed Recovery Using Stable Isotope Analysis

    Kelp forests have declined gradually all over the world. Understanding the trophic structure of such a productive and diverse ecosystem is crucial for its restoration and effective management. Few studies, however, have focused on the trophic structure and functional recovery of kelp forests in the process of restoration. This study was conducted in the eastern coast of Korea where kelp bed restoration was in process with the removal of sea urchins. In addition to quantitative measurement for recovery using common community parameters, we investigated how fast the stability of a food web structure could be established in the newly restored kelp beds with an initially barren condition, using stable isotope analysis, in comparison with a nearby natural bed and barren site. At the restored bed, total algal biomass and diversity reached the levels of the natural bed within 1 year. While the Ī“13C and Ī“15N values of macroalgae and organic matter were separated isotopically, they were similar among sites, excluding organic matter at the natural bed in 2019. Most consumers showed similar isotopic values among sites, with overlapping range for Ī“13C of producers. However, some herbivores showed higher Ī“15N values as predators/omnivores, particularly at barrens, which could be explained by trophic plasticity depending on the macroalgal structure. In the restored bed, for the first year, higher trophic diversity (CR, CD) and lower trophic redundancy (MNND, SDNND) showed non-overlapping >50% CIs among sites. However, this distinctive stage moved toward the natural bed upon entering the second year as trophic diversity decreased and trophic redundancy increased while natural bed overlapped at<50% CIs. The recovery speed in this system was fast (1 year for quantitative perspectives and 2 years for functional completion). After removing the sea urchins, recruitment of macroalgae quickly increased concomitantly with consumer groups, boosting the diversity and trophic structure of the restored bed. The trophic structure of the first year of restoration was not an intermediate stage toward the completion, but a transient over-shooting state. This might have been triggered by the fast introduction of diverse macroalgae to the urchin-free bare rock space, providing an interesting finding needed to be tested in other temperate marine systems

    Visfatin exerts angiogenic effects on human umbilical vein endothelial cells through the mTOR signaling pathway

    AbstractThe biologically active factors known as adipocytokines are secreted primarily by adipose tissues and can act as modulators of angiogenesis. Visfatin, an adipocytokine that has recently been reported to have angiogenic properties, is upregulated in diabetes, cancer, and inflammatory diseases. Because maintenance of an angiogenic balance is critically important in the management of these diseases, understanding the molecular mechanism by which visfatin promotes angiogenesis is very important. In this report, we describe our findings demonstrating that visfatin stimulates the mammalian target of the rapamycin (mTOR) pathway, which plays important roles in angiogenesis. Visfatin induced the expression of hypoxia-inducible factor 1Ī± (HIF1Ī±) and vascular endothelial growth factor (VEGF) in human endothelial cells. Inhibition of the mTOR pathway by rapamycin eliminated the angiogenic and proliferative effects of visfatin. The visfatin-induced increase in VEGF expression was also eliminated by RNA interference-mediated knockdown of the 70-kDa ribosomal protein S6 kinase (p70S6K), a downstream target of mTOR. Visfatin inactivated glycogen synthase kinase 3Ī² (GSK3Ī²) by phosphorylating it at Ser-9, leading to the nuclear translocation of Ī²-catenin. Both rapamycin co-treatment and p70S6K knockdown inhibited visfatin-induced GSK3Ī² phosphorylation at Ser-9 and nuclear translocation of Ī²-catenin. Taken together, these results indicate that mTOR signaling is involved in visfatin-induced angiogenesis, and that this signaling leads to visfatin-induced VEGF expression and nuclear translocation of Ī²-catenin

    Vardenafil Enhances Oxytocin Expression in the Paraventricular Nucleus without Sexual Stimulation

    PurposeOxytocin is associated with the ability to form normal social attachments. c-Fos is an immediate early gene whose expression is used as a marker for stimulus-induced changes in neurons. The effect of phosphodiesterase-5 (PDE-5) inhibitors on oxytocin activation in the brain without sexual stimuli has not yet been reported. In the present study, we investigated the effects of vardenafil on oxytocin and c-Fos expression in the paraventricular nucleus (PVN) of conscious rats.MethodsMale Sprague-Dawley rats weighing 300Ā±10 g were divided into 6 groups (n=5 in each group): the control group, the 1-day-0.5 mg/kg, the 1-day-1 mg/kg, the 1-day-2 mg/kg, the 3-day-1 mg/kg, and the 7-day-1 mg/kg vardenafil administration group. The experiment was conducted without sexual stimulation. Vardenafil was orally administered. The animals in the control group received an equivalent amount of distilled water orally. The expression of oxytocin and c-Fos in the PVN was detected by immunohistochemistry.ResultsOxytocin expression in the PVN was increased by 1 day administration of 2 mg/kg vardenafil, and this effect of vardenafil appeared in a duration-dependent manner. c-Fos in the oxytocin neurons of the PVN was increased by 1 day administration of 2 mg/kg vardenafil, and this effect of vardenafil also appeared in a duration-dependent manner. These results showed that vardenafil augments the expression of oxytocin with activation of oxytocin neurons in the PVN.ConclusionsIn this study, we showed that the PDE-5 inhibitor, vardenafil directly enhances oxytocin expression and also activates oxytocin neurons in the PVN, which indicates that vardenafil may exert positive effects on affiliation behavior and social interaction

    A case of back pain caused by Salmonella spondylitis -A case report-

    Salmonella spondylitis is a rare illness, and it generally occurs in patients who have already had sickle cell anemia, and it is even rarer in patients who are without sickle cell anemia. A 61-year-old male patient was hospitalized for the evaluation of his renal function and then treatment was started for his back pain. His back pain had developed about 2 months previously without any specific trauma. Only a bulging disc was detected on the initial lumbar MRI. Regarding his fever, it was diagnosed as possible atypical pneumonia, scrub typhus, etc., and multiple antibiotic therapy was administered. At the time of transfer, the leucocytes and hs-CRP were normal and the ESR was elevated. A diagnostic epidural block was performed for his back pain, but his symptoms were not improved. Lumbar MRI was performed again and it showed findings of infective spondylitis. Salmonella D was identified on the abscess culture and so he was diagnosed as suffering from Salmonella spondylitis. After antibiotic treatment, his back pain was improved and the patient was able to walk
