    33.8 GHz CCS Survey of Molecular Cores in Dark Clouds

    We have conducted a survey of the CCS JN=3221J_N = 3_2-2_1 line toward 11 dark clouds and star-forming regions at 30 arcsec spatial resolution and 0.054 km/s velocity resolution. CCS was only detected in quiescent clouds, not in active star-forming regions. The CCS distribution shows remarkable clumpy structure, and 25 clumps are identified in 7 clouds. Seven clumps with extremely narrow nonthermal linewidths < 0.1 km/s are among the most quiescent clumps ever found. The CCS clumps tend to exist around the higher density regions traced by NH_3 emission or submillimeter continuum sources, and the distribution is not spherically symmetric. Variation of the CCS abundance was suggested as an indicator of the evolutionary status of star formation. However, we can only find a weak correlation between N(CCS) and nH2,virn_{H_2,vir}. The velocity distributions of CCS clouds reveal that a systematic velocity pattern generally exists. The most striking feature in our data is a ring structure in the position-velocity diagram of L1544 with an well-resolved inner hole of 0.04 pc x 0.13 km/s and an outer boundary of 0.16 pc x 0.55 km/s. This position-velocity structure clearly indicates an edge-on disk or ring geometry, and it can be interpreted as a collapsing disk with an infall velocity \gtrsim 0.1 km/s and a rotational velocity less than our velocity resolution. Nonthermal linewidth distribution is generally coherent in CCS clouds, which could be evidence for the termination of Larson's Law at small scales, \sim 0.1 pc.Comment: 21 pages, 25 ostscript figures, accepted for publication in the Supplement Series of the Astrophysical Journal (May 2000

    Control of RelB during dendritic cell activation integrates canonical and noncanonical NF-κB pathways.

    The NF-κB protein RelB controls dendritic cell (DC) maturation and may be targeted therapeutically to manipulate T cell responses in disease. Here we report that RelB promoted DC activation not as the expected RelB-p52 effector of the noncanonical NF-κB pathway, but as a RelB-p50 dimer regulated by canonical IκBs, IκBα and IκBɛ. IκB control of RelB minimized spontaneous maturation but enabled rapid pathogen-responsive maturation. Computational modeling of the NF-κB signaling module identified control points of this unexpected cell type-specific regulation. Fibroblasts that we engineered accordingly showed DC-like RelB control. Canonical pathway control of RelB regulated pathogen-responsive gene expression programs. This work illustrates the potential utility of systems analyses in guiding the development of combination therapeutics for modulating DC-dependent T cell responses

    Fine-Tuning of PI3K/AKT Signalling by the Tumour Suppressor PTEN Is Required for Maintenance of Flight Muscle Function and Mitochondrial Integrity in Ageing Adult Drosophila melanogaster

    Insulin/insulin-like growth factor signalling (IIS), acting primarily through the PI3-kinase (PI3K)/AKT kinase signalling cassette, plays key evolutionarily conserved regulatory roles in nutrient homeostasis, growth, ageing and longevity. The dysfunction of this pathway has been linked to several age-related human diseases including cancer, Type 2 diabetes and neurodegenerative disorders. However, it remains unclear whether minor defects in IIS can independently induce the age-dependent functional decline in cells that accompany some of these diseases or whether IIS alters the sensitivity to other aberrant signalling. We identified a novel hypomorphic allele of PI3K’s direct antagonist, Phosphatase and tensin homologue on chromosome 10 (Pten), in the fruit fly, Drosophila melanogaster. Adults carrying combinations of this allele, Pten5, combined with strong loss-of-function Pten mutations exhibit subtle or no increase in mass, but are highly susceptible to a wide range of stresses. They also exhibit dramatic upregulation of the oxidative stress response gene, GstD1, and a progressive loss of motor function that ultimately leads to defects in climbing and flight ability. The latter phenotype is associated with mitochondrial disruption in indirect flight muscles, although overall muscle structure appears to be maintained. We show that the phenotype is partially rescued by muscle-specific expression of the Bcl-2 homologue Buffy, which in flies, maintains mitochondrial integrity, modulates energy homeostasis and suppresses cell death. The flightless phenotype is also suppressed by mutations in downstream IIS signalling components, including those in the mechanistic Target of Rapamycin Complex 1 (mTORC1) pathway, suggesting that elevated IIS is responsible for functional decline in flight muscle. Our data demonstrate that IIS levels must be precisely regulated by Pten in adults to maintain the function of the highly metabolically active indirect flight muscles, offering a new system to study the in vivo roles of IIS in the maintenance of mitochondrial integrity and adult ageing

    Chiral and herringbone symmetry breaking in water-surface monolayers

    We report the observation from monolayers of eicosanoic acid in the L′2 phase of three distinct out-of-plane first-order diffraction peaks, indicating molecular tilt in a nonsymmetry direction and hence the absence of mirror symmetry. At lower pressures the molecules tilt in the direction of their nearest neighbors. In this region we find a structural transition, which we tentatively identify as the rotator-herringbone transition L2d−L2h

    It has been claimed that ``the use of entangled photons in an imaging system can exhibit effects that cannot be mimicked by any other two-photon source, whatever strength of the correlations between the two photons'' [A. F. Abouraddy, B. E. A. Saleh, A. V. Sergienko, and M. C. Teich, Phys. Rev. Lett. 87, 123602 (2001)].     Staggered-vorticity correlations in a lightly doped t-J model: a variational approach

    We report staggered vorticity correlations of current in the d-wave variational wave function for the lightly-doped t-J model. Such correlations are explained from the SU(2) symmetry relating d-wave and staggered-flux mean-field phases. The correlation functions computed by the variational Monte Carlo method suggest that pairs are formed of holes circulating in opposite directions.Comment: ReVTeX, 4 pages, 3 figure
