Synthesis of graphene oxideâmethacrylic acidâsodium allyl sulfonate copolymer and its tanning properties
AbstractGraphite oxide nanosheets (GONs) and the copolymer of GONs with methacrylic acid (MAA) and sodium allyl sulfonate (SAS) (poly(GONâMAAâSAS)) were prepared. The GONs in poly(GONâMAAâSAS) are smaller and uniformly dispersed, allowing them to penetrate into collagen fibers of leather and produce better tanning effects than current nano-tanning agents. Tanning effects due to chemical bonding and nanoeffects are elucidated by measuring the shrinkage temperature (Ts) of wet and dry leather. The results indicate that poly(GONâMAAâSAS) could be used alone as a tanning agent to provide excellent mechanical properties, especially good elasticity and softness, although the Ts is slightly lower than that of chrome-tanned leather. Poly(GONâMAAâSAS) in combination with a chrome tanning agent could allow the dosage of the latter to be halved. These results indicate the potential for new nano-tanning agents to reduce the pollution caused by tanning agents
Research on Deformation Evolution of a Large Toppling Based on Comprehensive Remote Sensing Interpretation and Real-Time Monitoring
Deep, unstable slopes are highly developed in mountainous areas, especially in the Minjiang River Basin, Sichuan Province, China. In this study, to reveal their deformation evolution characteristics for stability evaluation and disaster prevention, multi-period optical remote sensing images (2010â2019), SBAS-InSAR data (January 2018âDecember 2019), and on-site real-time monitoring (December 2017âSeptember 2020) were utilized to monitor the deformation of a large deep-seated toppling, named the Tizicao (TZC) Toppling. The obtained results by different techniques were cross-validated and synthesized in order to introduce the spatial and temporal characteristics of the toppling. It was found that the displacements on the north side of the toppling are much larger than those on the south side, and the leading edge exhibits a composite damage pattern of âcollapse failureâ and âbulging crackingâ. The development process of the toppling from the formation of a tensile crack at the northern leading edge to the gradual pulling of the rear edge was revealed for a time span of up to ten years. In addition, the correlation between rainfall, earthquakes, and GNSS time series showed that the deformation of the toppling is sensitive to rainfall but does not change under the effect of earthquakes. The surface-displacement-monitoring method in this study can provide a reference for the evolution analysis of unstable slopes with a large span of deformation.</p
Gamma-tocotrienol stimulates the proliferation, differentiation, and mineralization in osteoblastic MC3T3-E1 cells
Gamma-tocotrienol, a major component of tocotrienol-rich fraction of palm oil, has been suggested to exhibit bone protective effects in vivo. However, the effects of γ-tocotrienol on osteoblast cells are still unclear. In this study, the effects of γ-tocotrienol on the proliferation, differentiation, and mineralization in osteoblastic MC3T3-E1 cells were investigated. Our results showed that γ-tocotrienol (2â8âÎŒmol/L) significantly improved the cell proliferation (), but it didânotâaffectâcell cycle progression. γ-Tocotrienol significantly increased alkaline phosphatase (ALP) activity (), secretion levels of osteocalcin (OC) and osteonectin (ON), and mRNA levels of collagen type I (Col I) of MC3T3-E1 cells. Meanwhile, we found that γ-tocotrienol is promoted in differentiation MC3T3-E1 cells by upregulation of the expression of Runx2 protein. Moreover, the number of bone nodules increased over 2.5-fold in cells treated with γ-tocotrienol (2â8âÎŒmol/L) for 24âd compared to control group. These results indicated that γ-tocotrienol at low dose levels, especially 4âÎŒmol/L, could markedly enhance the osteoblastic function by increasing the proliferation, differentiation, and mineralization of osteoblastic MC3T3-E1 cells. Moreover, our data also indicated that Runx2 protein may be involved in these effects. Further studies are needed to determine the potential of γ-tocotrienol as an antiosteoporotic agent
Earthquake-Induced Stress Amplification and Rock Fragmentation within a Deep-Seated Bedding Fault: Case Study of the Daguangbao Landslide Triggered by the 2008 Wenchuan Earthquake (Ms=8.0)
AbstractThe 2008 Wenchuan Ms 8.0 earthquake triggered the Daguangbao (DGB) landslide, of which the shear surface belongs to a thrust bedding fault 400âm below the carbonate slope. After the landslide, a 1.8âkm-long inclined sliding face (0.3âkm2) was exposed in the south source area. By using shaking table test, the contributions of the fault to the landslide sliding have been studied in this paper. The bedding fault in the test model is simplified as a weak layer with small elasticity and the carbonate layers as a hard layer with high elastic modulus, which is 296 times the weak one. The test records larger displacement amplitude in the upper hard layer than that in the lower one and larger pressure amplitude in the weak layer than that in the hard ones. We ascribed the stress amplification in the weak layer to time delay of shaking wave as wave velocity in the weak layer is only 1/15 of that in the hard layers. Such time delay gives rise to phase differences between the hard layers during shaking. The compressive stress amplification occurs in the weak layer when the upper hard layer moves downwards relative to the lower one; otherwise, tensile stress amplification occurs. It is suggested that this kind of stress amplification triggered an extensive fragmentation of the bedding fault rock mass during the Wenchuan earthquake, which can be verified by a good deal of gentle-dip and steep-dip cracks observed on site. It is proposed that stress amplification had caused a fast dropping of shear strength in the bedding fault to enhance the suddenness of DGB landslide initiation
Selenium Yeast Dietary Supplement Affects Rumen Bacterial Population Dynamics and Fermentation Parameters of Tibetan Sheep (
Selenium (Se) deficiency is a widespread and seasonally chronic phenomenon observed in Tibetan sheep (Ovis aries) traditionally grazed on the QinghaiâTibet Plateau (QTP). Effects of the dietary addition of Se-enriched yeast (SeY) on the bacterial community in sheep rumen and rumen fermentation were evaluated with the aim of gaining a better understanding of the rumen prokaryotic community. Twenty-four yearling Tibetan rams [initial average body weight (BW) of 31.0 ± 0.64 kg] were randomly divided into four treatment groups, namely, control (CK), low Se (L), medium Se (M), and high Se (H). Each group comprised six rams and was fed a basic diet of fresh forage cut from the alpine meadow, to which SeY was added at prescribed dose rates. This feed trial was conducted for over 35 days. On the final day, rumen fluid was collected using a transesophageal sampler for analyzing rumen pH, NH3-N content, volatile fatty acid (VFA) level, and the rumen microbial community. Our analyses showed that NH3-N, total VFA, and propionate concentrations in the M group were significantly higher than in the other groups (P < 0.05). Both the principal coordinates analysis (PCoA) and the analysis of similarities revealed that the bacterial population structure of rumen differed among the four groups. The predominant rumen bacterial phyla were found to be Bacteroidetes and Firmicutes, and the three dominant genera in all the samples across all treatments were Christensenellaceae R7 group, Rikenellaceae RC9 gut group, and Prevotella 1. The relative abundances of Prevotella 1, Rikenellaceae RC9 gut group, Ruminococcus 2, Lachnospiraceae XPB1014 group, Carnobacterium, and Hafnia-Obesumbacterium were found to differ significantly among the four treatment groups (P < 0.05). Moreover, Tax4fun metagenome estimation revealed that gene functions and metabolic pathways associated with carbohydrate and other amino acids were overexpressed in the rumen microbiota of SeY-supplemented sheep. To conclude, SeY significantly affects the abundance of rumen bacteria and ultimately affects the rumen microbial fermentation
Rumen microbiota of Tibetan sheep (Ovis aries) adaptation to extremely cold season on the Qighai-Tibetan Platea
Publication history: Accepted - 3 May 2021; Published online - 25 may 2021.The Qinghai-Tibet Plateau is characterized by low temperatures and hypoxia, and this
feature is more obvious in the winter. However, it is not clear how Tibetan sheep
adapt to extreme cold climates. To address this, we used physiological methods
combined with next-generation sequencing technology to explore the differences in
growth performance, forage nutrient digestion, serum biochemical indexes, and rumen
microbial communities of Tibetan sheep (Ovis aries) between the summer and winter. In
the summer, owing to the high nutritional quality of the forage, the Tibetan sheep showed
enhanced forage degradation and fermentation though increased counts of important
bacteria in the rumen, such as Bacteroidetes, Prevotella_1, Prevotellaceae_UCG-003,
Ruminococcus_1, Saccharofermentans, and Ruminococcaceae_UCG-014, to improve
the growth performance and increase serum immunity and antioxidant status. In the
winter, owing to the low nutritional quality of the forage, the Tibetan sheep presented
low values of forage degradation and fermentation indicators. The relative abundance
of Firmicutes, the Firmicutes/Bacteroidetes ratio, microbial diversity, interactive activity
between microorganisms, and metabolism were significantly increased, implying that
the rumen microbiota could promote the decomposition of forage biomass and the
maintenance of energy when forage nutritional value was insufficient in the winter.
Our study helps in elucidating the mechanism by which Tibetan sheep adapt to the
high-altitude harsh environments, from the perspective of the rumen microbiota. This work was supported by the Program for Strategic Priority Research Program of the Chinese Academy of Sciences (Grant No. XDA20100102), the Second Tibetan Plateau Scientific Expedition and Research: Grassland Ecosystem and Ecological Animal Husbandry (Grant No. 2019QZKK0302), and Innovative Research Team of the Ministry of Education (Grant No. IRT_17R50)
Induction of Escherichia coli Into a VBNC State by Continuous-Flow UVC and Subsequent Changes in Metabolic Activity at the Single-Cell Level
A viable but non-culturable (VBNC) state of bacteria induced by disinfection in water treatment poses serious health risks because of possible resuscitation of VBNC cells during transportation. In this study, a setup using continuous-flow ultraviolet (UVC) irradiation ranging from 0 to 172.2 mJ cm-2 was designed to simulate real-world disinfection in both drinking water (SDW) and reclaimed water (SRW) treatment plants. A systematic investigation of UVC-induced VBNC bacteria, including occurrence, resuscitation, and time-dependent recovery of metabolic activity during post-incubation, was conducted. Different techniques including two new ones of âsingle cell cultureâ and D2O-labeled single-cell Raman spectroscopy were employed to gain comprehensive insights into VBNC cells. Heterotrophic plate counts (HPC) and 5-cyano-2,3-ditoyl tetrazolium chloride flow cytometry (CTC-FCM) assay demonstrated that exposure to continuous-flow UVC can induce E. coli into a VBNC state. Membranes integrity and 16S rRNA transcription level of VBNC bacteria were demonstrated to be unaffected by UVC exposure even at a high dose of 172.2 mJ cm-2. Resuscitation of VBNC bacteria was identified in a more accurate way based on âsingle cell culture.â Finally, time-dependent evolution of metabolic activity of UVC-treated cells during post-incubation was examined by D2O-labeled Raman spectroscopy at a high-resolution of single-cell level. C-D Raman bands resulting from incorporation of D2O-derived D into bacterial biomass were used as a sensitive and quantitative indicator of bacterial metabolic activity. A lower UVC dose, longer post-incubation time, and higher initial number of bacteria were demonstrated to result in a faster recovery of metabolic activity. Heterogeneous metabolic activity and subpopulation with higher metabolic activity were also revealed by single-cell Raman, even for UVC-treated cells losing cultivability. The comprehensive assessment of VBNC bacteria in UVC-disinfected drinking and reclaimed water points out treatment deficiencies of UVC and the necessity to develop more effective strategies to eliminate VBNC cells
Preâsymptomatic transmission of novel coronavirus in community settings
We used contact tracing to document how COVIDâ19 was transmitted across 5 generations involving 10 cases, starting with an individual who became ill on January 27. We calculated the incubation period of the cases as the interval between infection and development of symptoms. The median incubation period was 6.0Â days (interquartile range, 3.5â9.5Â days). The last two generations were infected in public places, 3 and 4Â days prior to the onset of illness in their infectors. Both had certain underlying conditions and comorbidity. Further identification of how individuals transmit prior to being symptomatic will have important consequences.Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/163478/2/irv12773.pdfhttp://deepblue.lib.umich.edu/bitstream/2027.42/163478/1/irv12773_am.pd
Research on HC-LSSVM Model for Soft Soil Settlement Prediction Based on Homotopy Continuation Method
Prediction of soft soil settlement is an important research topic in the field of civil engineering, and the least square support vector machine is one of the commonly used prediction methods at present. Nonetheless, the existing LSSVM models have problems of low search efficiency in the search process and lack of global optimal solution in the search results. In order to solve this problem, based on the leave-one-out cross-validation method, the homotopy continuation method was used to optimize the LSSVM model parameters, and then the HC-LSSVM model was constructed with the goal of minimizing the sum of squares of the prediction error of the full sample retention one. Finally, the rationality and correctness of the model are verified by engineering application. The results show that the HC-LSSVM model constructed in this study can accurately predict the settlement of soft ground, which is superior to the common LSSVM model and solves the problem that the parameters of LSSVM model cannot be solved optimally. The research results provide a new method for prediction of soft soil settlement
