    BIOAVAILABILITY ENHANCEMENT OF POORLY SOLUBLE RALOXIFENE BY DESIGNING INCLUSION COMPLEX WITH β–CYCLODEXTRIN

    Objective: Raloxifene hydrochloride (RLX) is widely used in the treatment of osteoporosis, but due to its extensive first pass metabolism bioavailability of RLX is remaining only 2%. In addition of that being from BCS class II, RLX has poor water solubility. Therefore the objective of present research work was to enhance solubility and dissolution rate of RLX by formulating inclusion complex with β cyclodextrin (β-CD) as a carrier.Methods: Inclusion complex was prepared by various methods like physical mixture, co-precipitation method and kneading method using different drug to carrier ratios (1:1, 1:2 and 1:3).Results: Inclusion complex prepared with co-precipitation method had shown 5.5 fold improvements in water solubility and significant increment in dissolution rate when compared with plain RLX. Optimized formulation was characterized by Fourier transform infrared spectroscopy, Differential scanning calorimetry, X-ray diffraction and Scanning electron microscopy studies for their compatibility, thermal analysis, crystallinity and surface morphology determination, respectively. Results of DSC and XRD study suggested the conversion of RLX from crystalline form to amorphous form. Stability studies showed stable formulation up to the period of 6 months. In vivo pharmacokinetic study was also conducted in wistar rats for optimized drug loaded inclusion complex that showed nearby two fold increments in drug bioavailability compared to plain drug suspension.Conclusion: From these studies, it can be concluded that solubility and dissolution rate of poorly soluble raloxifene could be remarkably increased by formulating into the inclusion complex with β-CD which results in significant improvement in bioavailability of poorly soluble RLX.Â

    Zinc oxide nanoparticles (ZnONPs) biosynthesis is gaining popularity since it is environmentally safe and can withstand a wide range of environmental conditions. The widely known medicinal herb Aloe barbadensis was employed to create ZnONPs in this work. XRD (X-Ray Diffraction), EDAX (Energy dispersive X-ray microanalysis), and TEM (Transmission Electron Microscopy) were also used to characterise the produced ZnONPs. In XRD, the produced ZnONPs revealed crystalline character, with an average size of 30 50 nm. TEM was used to determine spherical morphology. Under ultraviolet irradiation, the photocatalytic decolorization of Sudan IV, Crystal Violet (CV), and Acridine Orange (AO) by biogenic produced ZnONPs was studied. Using all three dyes (10-50 ppm) throughout a 4-hour incubation time, the produced ZnONPs showed 100% photocatalytic decolorization activity    &nbsp

    Thyroid Function and Thyroglobulin Level in Iodine-Deficient Children of Eastern Nepal

    Iodine deficiency during childhood affects physical and mental development. Iodine deficiency or excess both can negatively impact thyroid function. We conducted this study to assess iodine nutrition and thyroid function in children with insufficient urinary iodine concentration. A community-based cross-sectional study was conducted among the selected schools of Udayapur district. Urinary iodine concentration (UIC) was measured in 1012 school children (6-14 years). Based on UIC data, 83 blood samples were collected to measure serum thyroglobulin (Tg), thyroid-stimulating hormone (TSH), free triiodothyronine (fT3), and free thyroxine (fT4). UIC was measured by ammonium persulfate digestion method, and Tg, TSH, fT4, and fT3 were measured using ELISA kits. The median UIE was 236 µg/L, and 11.1% of the children had insufficient UIC. The mean fT3, fT4, and TSH in children with insufficient UIC were 2.55±0.43 pg/mL, 0.96±0.28 ng/dL, and 3.60±1.44 mIU/L respectively. Among children with low UIC levels, the median Tg was 17.5 ng/mL. Overt hypothyroidism was seen in 6%, and subclinical hypothyroidism in 3.6%. The children had sufficient iodine nutrition, and the frequency of thyroid dysfunction was low among the children with insufficient UIC

    Popliteal aneurysms are the most common peripheral arterial aneurysm and occur most commonly among older men with established cardiovascular disease. Popliteal aneurysms are asymptomatic or otherwise present with intermittent claudication, pressure symptoms in the popliteal fossa, distal embolization, and, rarely, rupture. We present a patient with a remarkably large popliteal aneurysm of 10 cm presenting as a popliteal swelling with foot drop and no signs of limb ischemia. According to our literature search, it is the largest reported popliteal aneurysm, and its corresponding symptoms are unusual. The diagnostic workup and treatment are presented

    Labetalol HCl competitively blocks adrenergic stimulation of β-receptors within the myocardium (β1-receptors) and within bronchial and vascular smooth muscle (β2-receptors), and α1-receptors within vascular smooth muscle. Mouth dissolving drug delivery systems (MDTs) have acquired an important position in the market by overcoming previously encountered administration problems and contributing to extension of patent life. MDTs have the unique property of rapidly disintegrating and/or dissolving and releasing the drug as soon as they come in contact with saliva, thus obviating the requirement of water during administration. Taste masking was done by using Kyron-T 134 in ratio 1:3. The tablets were prepared by using direct compression method, using different Superdisintegrantsand they were then evaluated for pre and post compression parameters. More than 80% of drug was released from almost all the formulations within 5 min. Results of this study indicate among the superdisintegrants tried, Indion-414 showed the best result in 2% concentration
