    Consensus of Corporate E-Learning System Stakeholders Regarding the Satisfaction of End-Users

    The purpose of this study is to call attention to the consensus of stakeholders of corporate e-Learning system regarding success. We identified the critical success factors (contents, technical features, management, and organizational support) as major components of corporate eLearning systems and questioned whether stakeholdersā€™ consensus on the importance of these components facilitates the implementation of these components to achieve good quality or well. We also questioned whether the influence of these components on user satisfaction could be moderated by contextual factors. Based on empirical testing of 18 eLearning user companies, we verified that the consensus of stakeholders regarding the importance of content, technological features, and organizational support has a positive influence on the perceived quality of these factors in their e-Learning systems, which in turn is positively related to user satisfaction. The learning subjects and learning style did significantly moderate the influences of these perceived qualities on user satisfaction

    Prediction of Advanced Fibrosis in Nonalcoholic Fatty Liver Disease: An Enhanced Model of BARD Score.

    UNLABELLED: BACKGROUNDAIMS: The BARD score is a model to detect advanced liver fibrosis in nonalcoholic fatty liver disease (NAFLD) patients. The aims of this study were to identify additional factors and then to build an enhanced version of the BARD score. METHODS: One hundred seven patients with biopsy-proven NAFLD were enrolled retrospectively. Logistic regressions were performed to identify independent risk factors for advanced liver fibrosis (stage 3 or 4). An enhanced model of the BARD score (BARDI score) was built and evaluated with a receiver operating characteristic (ROC) curve. RESULTS: In multivariate analysis, age (odds ratio [OR], 0.89; p=0.04), aspartate aminotransferase/alanine aminotransferase ratio (OR, 1.73; p CONCLUSIONS: The BARDI score had an improved PPV over the BARD score and maintained an excellent NPV. Further study is warranted for its external validation and comparison with other models

    Solvent-induced porosity control of carbon nanofiber webs for supercapacitor

    A simple and scalable method is reported for fabricating a porosity-controlled carbon nanofibers with a skin-core texture by electrospinning a selected blend of polymer solutions. Simple thermal treatment of the electrospun nanofibers from solution blends of various compositions creates suitable ultramicropores on the surface of carbon nanofibers that can accommodate many ions, removing the need for an activation step. The intrinsic properties of the electrode (e.g., nanometre-size diameter, high specific surface area, narrow pore size distribution, tuneable porosity, shallow pore depth, and good ionic accessibility) enable construction of supercapacitors with large specific capacitance (130.7 Fg(-1)), high power (100 kW kg(-1)), and energy density (15.0 Wh kg(-1)).     The Socio-economic Effect of Community-Driven Development in Conflict-affected Regions: Evidence from Cambodia

    Community-driven development (CDD) has become increasingly common approach used by mul-tilateral development banks, governments, and NGOs to address the needs of the poor local communities in developing countries. CDD is characterized by the approach of giving control of decision and resources to the local community. CDD approaches are particularly prominent in conflict-affected communities which enables local community to regain sense of social cohesion and to restore livelihood. While much of literature focused on documenting the challenges and benefits of CDD approach in conflict affected context, what is yet to be established is whether CDD in conflict affected context can generate change in social and economic outcomes. Thus, this paper explores whether or not community-driven development (CDD) project af-fects a villages social capital and economic outcomes in post-conflict Cambodia. We compare the impact of CDD by looking at data from two rounds of surveys (baseline and endline). The results indicate that the project significantly strengthens the capacity of self-reliance, especially in vulnerable groups such as low-income, less educated and ethnic and religious minority house-holds. However, CDD was shown to have very limited impact on improving economic condition. Although the project slightly increases the household monthly income, the CDD project does not generate remarkable changes in community-level economic conditions. The paper is organized as follows. Section 2 explains the historical background of Cambodia and the unique features of this CDD project. Section 3 describes the intervention and data collec-tion. Section 4 outlines the estimation models used in the analysis. In Section 5, we present the results of the program and explain their implications

    Anti-inflammatory effects of enzymatic hydrolysates of velvet antler in RAW 264.7 cells in vitro and zebrafish model

    Enzymatic hydrolysis has been successfully used for the extraction of numerous biologically active components from a wide variety of natural sources. In the present study, velvet antler was subjected to the extraction process using Alcalase protease. We analyzed bioactive components, such as uronic acid, sulfated-glycosaminoglycans (sulfated-GAGs), and sialic acid, present in the velvet antler Alcalase hydrolysate (VAAH) and assessed their anti-inflammatory effects in zebrafish as well as in vitro using cell lines. VAAH mainly contained uronic acid (78.22 mg/g) and sulfated-GAGs (50.47 mg/g), while the amount of sialic acid was negligible (5.55 mg/g). VAAH inhibited the production of nitric oxide (NO) by lipopolysaccharide (LPS)-induced cells in a dosedependent manner and the inhibitory effect of VAAH on NO production was higher than that of hot water extracts. VAAH treatment also reduced the expression of inflammatory mediators such as nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2). Furthermore, we evaluated anti-inflammatory effects of VAAH using LPS-stimulated zebrafish. Treatment with LPS significantly increased cell death, NO, and reactive oxygen species (ROS) levels in zebrafish. Notably, VAAH significantly inhibited the extent of LPS-stimulated cell death and generation of NO and ROS in zebrafish. These results suggest that VAAH alleviated inflammation and cell death by inhibiting the generation of ROS induced by LPS treatment. Thus, VAAH could be used as a potential natural remedy with a trong anti-inflammatory effect. Taken together, we believe that based on our present results, enzymatic hydrolysis of velvet antler may be an effective process to make antler products acceptable as elements of health foods and nutraceutical components with increased biological activity

    Development of a Novel Orthotopic Gastric Cancer Mouse Model.

    BACKGROUND: Gastric cancer metastasis is a highly fatal disease with a five-year survival rate of less than 5%. One major obstacle in studying gastric cancer metastasis is the lack of faithful models available. The cancer xenograft mouse models are widely used to elucidate the mechanisms of cancer development and progression. Current procedures for creating cancer xenografts include both heterotopic (i.e., subcutaneous) and orthotopic transplantation methods. Compared to the heterotopic model, the orthotopic model has been shown to be the more clinically relevant design as it enables the development of cancer metastasis. Although there are several methods in use to develop the orthotopic gastric cancer model, there is not a model which uses various types of tumor materials, such as soft tissues, semi-liquid tissues, or culture derivatives, due to the technical challenges. Thus, developing the applicable orthotopic model which can utilize various tumor materials is essential. RESULTS: To overcome the known limitations of the current orthotopic gastric cancer models, such as exposure of tumor fragments to the neighboring organs or only using firm tissues for the orthotopic implantation, we have developed a new method allowing for the complete insertion of soft tissue fragments or homogeneously minced tissues into the stomach submucosa layer of the immunodeficient NOD.Cg-Prkdc CONCLUSION: Here we describe a new protocol, step by step, for the establishment of orthotopic xenograft of gastric cancer. This novel technique will be able to increase the use of orthotopic models in broader applications for not only gastric cancer research but also any research related to the stomach microenvironment

    Hemeoxygenase-1 Mediates an Adaptive Response to Spermidine-Induced Cell Death in Human Endothelial Cells

    Spermidine (SPD) is a ubiquitous polycation that is commonly distributed in living organisms. Intracellular levels of SPD are tightly regulated, and SPD controls cell proliferation and death. However, SPD undergoes oxidation in the presence of serum, producing aldehydes, hydrogen peroxide, and ammonia, which exert cytotoxic effect on cells. Hemeoxygenase-1 (HO-1) is thought to have a protective effect against oxidative stress. Upregulation of HO-1 in endothelial cells is considered to be beneficial in the cardiovascular disease. In the present study, we demonstrate that the ubiquitous polyamine, SPD, induces HO-1 in human umbilical vein endothelial cells (HUVECs). SPD-induced HO-1 expression was examined by Western blot and reverse transcription-polymerase chain reaction (RT-PCR). Involvement of reactive oxygen species, serum amine oxidase, PI3K/Akt signaling pathway, and transcription factor Nrf2 in the induction of HO-1 by SPD was also investigated. Furthermore, small interfering RNA knockdown of Nrf2 or HO-1 and treatment with the specific HO-1 inhibitor ZnPP exhibited a noteworthy increase of death of SPD-stimulated HUVECs. In conclusion, these results suggest that SPD induces PI3K/Akt-Nrf2-mediated HO-1 expression in human endothelial cells, which may have a role in cytoprotection of the cells against oxidative stress-induced death

    Taurine chloramine differentially inhibits matrix metalloproteinase 1 and 13 synthesis in interleukin-1Ī² stimulated fibroblast-like synoviocytes

    It has been suggested that taurine chloramine (TauCl) plays an important role in the downregulation of proinflammatory mediators. However, little is known about its effect on the expression of matrix metalloproteinases (MMPs). In this study, we investigated the effects of TauCl on synovial expression of MMPs. The effects of TauCl on MMP expression in IL-1Ī² stimulated fibroblast-like synoviocytes (FLSs) were studied using the following techniques. Real-time PCR and semi-quantitative PCR were employed to analyze the mRNA expression of MMPs. ELISA was used to determine protein levels of MMPs. Western blot analyses were performed to analyze the mitogen-activated protein kinase and inhibitor of nuclear factor-ĪŗB (IĪŗB) kinase signalling pathways. Finally, electrophoretic mobility shift assay and immunohistochemistry were used to assess localization of transcription factors. IL-1Ī² increased the transcriptional and translational levels of MMP-1 and MMP-13 in rheumatoid arthritis FLSs, whereas the levels of MMP-2 and MMP-9 were unaffected. TauCl at a concentration of 400 to 600 Ī¼mol/l greatly inhibited the transcriptional and translational expression of MMP-13, but the expression of MMP-1 was significantly inhibited at 800 Ī¼mol/l. At a concentration of 600 Ī¼mol/l, TauCl did not significantly inhibit phosphorylation of mitogen-activated protein kinase or IĪŗB degradation in IL-1Ī² stimulated rheumatoid arthritis FLSs. The degradation of IĪŗB was significantly inhibited at a TauCl concentration of 800 Ī¼mol/l. The inhibitory effect of TauCl on IĪŗB degradation was confirmed by electrophoretic mobility shift assay and immunochemical staining for localization of nuclear factor-ĪŗB. TauCl differentially inhibits the expression of MMP-1 and MMP-13, and inhibits expression of MMP-1 primarily through the inhibition of IĪŗB degradation, whereas it inhibits expression of MMP-13 through signalling pathways other than the IĪŗB pathway
