    Расширенная ролевая модель безопасности для веб-приложений, основанная на иерархии путей

    Web applications security is a complex problem with several aspects. One aspect is access control according to specified security policy. Access control is accomplished by security model restrictions. This research is dedicated to developing security access control model for web applications. This work describes path-based RBAC model, which improves RBAC and allows flexible access control using request path (URI). Authors created guidelines to apply model’s elements for real-world web applications. Developing web applications with model described allows reducing security risksОбеспечение безопасности веб-приложений – комплексная проблема, имеющая несколько аспектов. Одним из аспектов является разграничение доступа в соответствии с заданной политикой безопасности. Разграничение доступа осуществляется путем применения ограничений, накладываемых моделью безопасности. Данная работа посвящена разработке модели безопасности для разграничения доступа в веб-приложениях. В работе описана основанная на иерархии путей ролевая модель безопасности, которая улучшает базовую модель RBAC и обеспечивает гибкое разграничение доступа на основе пути запроса (URI). Разработаны рекомендации по применению модели для веб-приложений. Разработка веб-приложений с применением описанной модели позволяет снизить риски, связанные с безопасность

    Broadness and specificity: ArdB, ArdA, and Ocr against various restriction-modification systems

    ArdB, ArdA, and Ocr proteins inhibit the endonuclease activity of the type I restriction-modification enzymes (RMI). In this study, we evaluated the ability of ArdB, ArdA, and Ocr to inhibit different subtypes of Escherichia coli RMI systems (IA, IB, and IC) as well as two Bacillus licheniformis RMI systems. Furthermore we explored, the antirestriction activity of ArdA, ArdB, and Ocr against a type III restriction-modification system (RMIII) EcoPI and BREX. We found that DNA-mimic proteins, ArdA and Ocr exhibit different inhibition activity, depending on which RM system tested. This effect might be linked to the DNA mimicry nature of these proteins. In theory, DNA-mimic might competitively inhibit any DNA-binding proteins; however, the efficiency of inhibition depend on the ability to imitate the recognition site in DNA or its preferred conformation. In contrast, ArdB protein with an undescribed mechanism of action, demonstrated greater versatility against various RMI systems and provided similar antirestriction efficiency regardless of the recognition site. However, ArdB protein could not affect restriction systems that are radically different from the RMI such as BREX or RMIII. Thus, we assume that the structure of DNA-mimic proteins allows for selective inhibition of any DNA-binding proteins depending on the recognition site. In contrast, ArdB-like proteins inhibit RMI systems independently of the DNA recognition site

    Dicyemida and Orthonectida: Two Stories of Body Plan Simplification

    Two enigmatic groups of morphologically simple parasites of invertebrates, the Dicyemida (syn. Rhombozoa) and the Orthonectida, since the 19th century have been usually considered as two classes of the phylum Mesozoa. Early molecular evidence suggested their relationship within the Spiralia (=Lophotrochozoa), however, high rates of dicyemid and orthonectid sequence evolution led to contradicting phylogeny reconstructions. Genomic data for orthonectids revealed that they are highly simplified spiralians and possess a reduced set of genes involved in metazoan development and body patterning. Acquiring genomic data for dicyemids, however, remains a challenge due to complex genome rearrangements including chromatin diminution and generation of extrachromosomal circular DNAs, which are reported to occur during the development of somatic cells. We performed genomic sequencing of one species of Dicyema, and obtained transcriptomic data for two Dicyema spp. Homeodomain (homeobox) transcription factors, G-protein-coupled receptors, and many other protein families have undergone a massive reduction in dicyemids compared to other animals. There is also apparent reduction of the bilaterian gene complements encoding components of the neuromuscular systems. We constructed and analyzed a large dataset of predicted orthologous proteins from three species of Dicyema and a set of spiralian animals including the newly sequenced genome of the orthonectid Intoshia linei. Bayesian analyses recovered the orthonectid lineage within the Annelida. In contrast, dicyemids form a separate clade with weak affinity to the Rouphozoa (Platyhelminthes plus Gastrotricha) or (Entoprocta plus Cycliophora) suggesting that the historically proposed Mesozoa is a polyphyletic taxon. Thus, dramatic simplification of body plans in dicyemids and orthonectids, as well as their intricate life cycles that combine metagenesis and heterogony, evolved independently in these two lineages

    Serum Cytokine Profile in a Patient Diagnosed with Dysferlinopathy

    Limb-girdle muscular dystrophy type 2 (LGMD2B) is a mild form of dysferlinopathy, characterized by limb weakness and wasting. It is an autosomal recessive disease, with currently 140 mutations in the LGMD2B gene identified. Lack of functional dysferlin inhibits muscle fiber regeneration in voluntary muscles, the main pathological finding in LGMD2B patients. However, the immune system has been suggested to contribute to muscle cell death and tissue regeneration. Serum levels of 27 cytokines were evaluated in a dysferlinopathy patient. Levels of 8 cytokines differed in patient serum compared to controls. Five cytokines (IL-10, IL-17, CCL2, CXCL10, and G-CSF) were higher while 3 were lower in the patient than in controls (IL-2, IL-8, and CCL11). Together, these data on serum cytokine profile of this dysferlinopathy patient suggest immune response activation, which could explain leukocyte infiltration in the muscle tissue

    Pilot study of the safety and efficacy of angiogenic therapy in diabetic foot syndrome

    BACKGROUND: The syndrome of diabetic foot remains the main cause of non-traumatic amputation of the lower extremity in the world. Even with the provision of comprehensive medical care in the conditions of a specialized center, 10-15% of patients do not succeed in healing the ulcerative defect due to the ischemic component. AIMS: The objective of this study is evaluation of safety and efficacy of pl-VEGF165 transfer in patients with neuroischemic type of diabetic foot syndrome. METHODS: The pilot study included 35 diabetic patients with neuroischemic foot ulcers (Wagner stage 1-2) who were not candidates for revascularization procedures (NCT02538705). The patients were closely monitored after repeated pl-VEGF165 intramuscular gene transfer (2,4 mg) at 1, 3, and 6 months after treatment. The primary efficacy endpoint was the surface area of the ulcers (sq.cm), the secondary endpoints were transcutaneous oxygen tension (Tcp02), ankle-brachial index (ABI), neuropathy disability score (NDS), neuropathy symptoms score (NSS), and Michigan neuropathy screening instrument (MNSI). Adverse events were monitored throughout the study. RESULTS: The use of pl-VEGF165 as part of complex treatment allowed to achieve wound healing in 65,7% of patients with chronic ulcerative defects, the safety of the target limb was 84%. Carrying out therapeutic angiogenesis as a part of the combined therapy ensured a reduction in the average area of the resistant to treatment defects from 3.6 [1.0; 7.05] cm2 to 0.0 [0.0;2.0] cm2 (p=0,001), which correlated with an increase in the TcPo2 index by 15% from 35 [29.5; 40.5] to 40.5 [36.0; 46.5] mm Hg (p= p=0,005) and in the ABI by 16% from 0.96 [0.82;1.08] to 1.11 [0.85; 1.24] (p=0,062). The decrease in the signs of diabetic neuropathy was determined - the scores of NSS scales and VAT decreased from 6,5 [5.75; 8.0) to 6.0 [5.25; 7.0] (p=0,004) and from 9.0 [8.0; 13.5] to 8.0 [7.0; 12.7] (p=0,001), respectively. No adverse effects associated with the use of pl-VEGF165 were recorded. CONCLUSIONS: Thus, preliminary results of the pilot study show that the use of pl-VEGF165 gene transfer in combination therapy allows for complete healing of neuroischemic diabetic foot ulcers in the majority of patients

    Quantitative and qualitative indicators of developing anticipation skills in wrestling athletes

    Actual problems of modern psychology are fundamental studies of the phenomenon of anticipation, the search for new methodological approaches to the study of its mechanisms and processes of development are considered. There is a particular interest in the anticipation issue in sport, where the probabilistic forecast of the situation is crucial for winning. The paper presents the results of testing methods for anticipation skills in wrestling. The main objective of the study was to find psycho-physiological and behavioral indicators to quantitatively and qualitatively evaluate the degree of anticipation. The skills formation procedure was based on the reinforcement of the correct choice of behaviour in simulated situations of decision-making using multiple choices technique. Stimuli were videos of simulated situations in wrestling. Simultaneous recording of oculomotor activity and registration of multi-channel electroencephalogram (EEG) was carried out. The results showed the effectiveness of the proposed method. After completing training the number of errors and the decision taking time span reduced. Expert assessment of the main qualifying factors showed a significant increase of the test group. On the psychophysiological level, there is a reduction of oculomotor activity in selecting the right answers, reducing the number of fixations, the number of fixations and saccades reverse on the text of questions and answers. Analysis of EEG parameters showed a gradual decrease in the index of brain activation when analysing simulated situations and dynamic movement of the peaks of activity in the frontal areas of the temporal and occipital areas during the series of trainings

    Modelling and Thropology of Fullerenes

    The dodecahedron C20 structure has been modified to all possible fullerenes up to C40 with the help of simple isoperimetric transformations. The new type of rectangular 5õ12 matrix has been suggested for fullerenes. It has been shown that all modelled fullerenes have toroidal matrices (T-matrices). The symmetry character of T-matrices is correlated with the symmetry of corresponding fullerenes
