Overexpression of Arabidopsis thaliana brassinosteroid-related acyltransferase 1 gene induces brassinosteroid-deficient phenotypes in creeping bentgrass

Brassinosteroids (BRs) are naturally occurring steroidal hormones that play diverse roles in various processes during plant growth and development. Thus, genetic manipulation of endogenous BR levels might offer a way of improving the agronomic traits of crops, including plant architecture and stress tolerance. In this study, we produced transgenic creeping bentgrass (Agrostis stolonifera L.) overexpressing a BR-inactivating enzyme, Arabidopsis thaliana BR-related acyltransferase 1 (AtBAT1), which is known to catalyze the conversion of BR intermediates to inactive acylated conjugates. After putative transgenic plants were selected using herbicide resistance assay, genomic integration of the AtBAT1 gene was confirmed by genomic PCR and Southern blot analysis, and transgene expression was validated by northern blot analysis. The transgenic creeping bentgrass plants exhibited BR-deficient phenotypes, including reduced plant height with shortened internodes (i.e., semi-dwarf), reduced leaf growth rates with short, wide, and thick architecture, high chlorophyll contents, decreased numbers of vascular bundles, and large lamina joint bending angles (i.e., erect leaves). Subsequent analyses showed that the transgenic plants had significantly reduced amounts of endogenous BR intermediates, including typhasterol, 6-deoxocastasterone, and castasterone. Moreover, the AtBAT1 transgenic plants displayed drought tolerance as well as delayed senescence. Therefore, the results of the present study demonstrate that overexpression of an Arabidopsis BR-inactivating enzyme can reduce the endogenous levels of BRs in creeping bentgrass resulting in BR-deficient phenotypes, indicating that the AtBAT1 gene from a dicot plant is also functional in the monocot crop.111Ysciescopu

Enhancement of phase separation in the InGaN layer for self-assembled In-rich quantum dots

    Get PDF
The enhancement of phase separation in the InGaN layer grown on a GaN layer with a rough surface was investigated for the formation of self-assembled In-rich quantum dots(QDs) in the InGaN layer. Transmission electron microscopy images showed that In-rich QDs with a size of 2–5 nm were formed even in an InGaN layer with a low indium content, and a layer thickness less than the critical thickness. The room-temperature photoluminescence(PL) spectrum of this layer showed emission peaks corresponding to In-rich QDs. The temperature-dependent PL spectra showed dominant peak shifts to the lower energy side, indicating that the self-assembled In-rich QDs are formed in the InGaN layer grown on a rough GaNsurface and that the carriers are localized in In-rich QDs

A Parallelized Model For Simulating A Vertical Closed-Loop Geothermal Heat Pump System

ABSTRACT A borehole heat exchanger (BHE) is one of the important factors that can affect performance of the vertical closedloop geothermal heat pump (GHP) system. Its construction costs are also a large percentage of the overall initial costs of the GHP system. When constructing BHEs, several parameters should be determined, such as length of the borehole heat exchanger, the number of the BHEs, and spacing between BHEs. Numerical simulation can be used to find optimal parameters. However, it takes a long time to find optimal parameters with a large domain. To reduce the calculation time, a massively parallel computing procedure is introduced into the serial simulator for the vertical closed-loop GHP system. It can simulate temperature changes in the BHE with circulating fluid through the Utubes and compute groundwater flow and aquifer temperature changes. By increasing the number of processors, the total execution time was reduced from 7,031 seconds using two processors to 1,145 seconds using 32 processors. The total execution time of the serial model is 13,902 seconds

Size distributions of atmospheric particulate matter and associated trace metals in the multi-industrial city of Ulsan, Korea

Particulate matter (PM) was collected using micro-orifice uniform deposit impactors from a residential (RES) site and an industrial (IND) site in Ulsan, South Korea, in September-October 2014. The PM samples were measured based on their size distributions (11 stages), ranging from 0.06 ??m to over 18.0 ??m. Nine trace metals (As, Se, Cr, V, Cd, Pb, Ba, Sb, and Zn) associated with PM were analyzed. The PM samples exhibited weak bimodal distributions irrespective of sampling sites and events, and the mean concentrations of total PM (TPM) measured at the IND site (56.7 ??g/m3) was higher than that measured at the RES site (38.2 ??g/m3). The IND site also showed higher levels of nine trace metals, reflecting the influence of industrial activities and traffic emissions. At both sites, four trace metals (Ba, Zn, V, and Cr) contributed to over 80% of the total concentrations in TPM. The modality of individual trace metals was not strong except for Zn; however, the nine trace metals in PM2.5 and PM10 accounted for approximately 50% and 90% of the total concentrations in TPM, respectively. This result indicates that the size distributions of PM and trace metals are important to understand how respirable PM affects public health

How Does the SST Variability over the Western North Atlantic Ocean Control Arctic Warming over the Barents-Kara Seas?

Arctic warming over the BarentsKara Seas and its impacts on the mid-latitude circulations have been widely discussed. However, the specific mechanism that brings the warming still remains unclear. In this study, a possible cause of the regional Arctic warming over the BarentsKara Seas during early winter (OctoberDecember) is suggested. We found that warmer sea surface temperature anomalies over the western North Atlantic Ocean (WNAO) modulate the transient eddies overlying the oceanic frontal region. The altered transient eddy vorticity flux acts as a source for the Rossby wave straddling the western North Atlantic and the BarentsKara Seas (Scandinavian pattern), and induces a significant warm advection, increasing surface and lower-level temperature over the Eurasian sector of the Arctic Ocean. The importance of the sea surface temperature anomalies over the WNAO and subsequent transient eddy forcing over the WNAO was also supported by both specially designed simple model experiments and general circulation model experiments

Journey of a Swallowed Toothbrush to the Colon

    Get PDF
Toothbrush swallowing is a rare event. Because no cases of spontaneous passage have been reported, prompt removal is recommended to prevent the development of complications. Most swallowed toothbrushes have been found in the esophagus or the stomach of affected patients, and there has been no previously reported case of a toothbrush in the colon. Here, we report a case of a swallowed toothbrush found in the ascending colon that caused a fistula between the right colon and the liver, with a complicating small hepatic abscess. This patient was successfully managed using exploratory laparotomy. To our knowledge, this is the first documented case of a swallowed toothbrush found in the colon

Generalized Lichen Nitidus Successfully Treated with Narrow-band UVB Phototherapy : Two Cases Report

    Get PDF
Lichen nitidus (LN) is an uncommon chronic inflammatory skin disease composed of numerous, tiny, shiny, flesh-colored papules that are predominantly observed on the chest, abdomen, glans penis and upper extremities. The distribution of LN is most often localized, but in some cases it can become generalized. Because LN tends to be asymptomatic and presents spontaneous resolution within several years, it usually does not require treatment except in symptomatic, persistent and generalized cases. We describe a 28-yr-old man and a 7-yr-old boy with generalized LN where both cases improved with narrow-band ultraviolet B (NB-UVB) phototherapy plus topical steroid ointment. Both patients noted improvement within the first three treatments and showed almost complete resolution after 18 and 20 treatments, respectively. NB-UVB phototherapy may be an effective alternative therapy for the treatment of generalized LN, even for those patients in their childhood
