    Nationalism, Nationalistic Demos and Democracy: East Asian Experiences

    This research is an attempt to offer a new theoretical framework to understand the political dynamics of East Asian nationalism(s), a topic overlooked by both historians and political scientists. The political dynamics of nationalism shown in the two historical case studies investigated here, the bottom-up ultra-right-wing nationalism in 1930s Japan and the anti-state left-wing/anti-imperial nationalism in 1980s Korea, poses a strong anti-thesis against our commonsensical understanding of nationalism. From the Eurocentric perspectives, the nationalist projects of nation-making always create a homogeneous –either real or fictive – population that is willing to fight and die for the state. The historical case studies shown in this study, however, refute the monolithic interpretation of nationalism in the modern history. Assuming that the nation-making projects in Japan and Korea were very successful, the histories of 1930s Japan and 1980s Korea show a hidden face of nationalism – the more nationalized, the more rebellious – as the nationalized subjects claimed ownership of the state. The experiences of nationalistic fever in Japan and Korea provide a prism to analyze contemporary Chinese neo-nationalism, which has become one of the most important research subjects in China. The experiences of Japan and Korea suggest that the only outcome we can predict from the surge of nationalism is the vitiated and weakened state capability to control the ideological realm of the society. Therefore, we can expect that the surge of nationalistic sentiments from the bottom up in Chinese societies pose a threat to the domestic stability managed by the Chinese Communist Party

    Genetic and Metabolic Characterization of Insomnia

    Insomnia is reported to chronically affect 10∼15% of the adult population. However, very little is known about the genetics and metabolism of insomnia. Here we surveyed 10,038 Korean subjects whose genotypes have been previously profiled on a genome-wide scale. About 16.5% reported insomnia and displayed distinct metabolic changes reflecting an increase in insulin secretion, a higher risk of diabetes, and disrupted calcium signaling. Insomnia-associated genotypic differences were highly concentrated within genes involved in neural function. The most significant SNPs resided in ROR1 and PLCB1, genes known to be involved in bipolar disorder and schizophrenia, respectively. Putative enhancers, as indicated by the histone mark H3K4me1, were discovered within both genes near the significant SNPs. In neuronal cells, the enhancers were bound by PAX6, a neural transcription factor that is essential for central nervous system development. Open chromatin signatures were found on the enhancers in human pancreas, a tissue where PAX6 is known to play a role in insulin secretion. In PLCB1, CTCF was found to bind downstream of the enhancer and interact with PAX6, suggesting that it can probably inhibit gene activation by PAX6. PLCB4, a circadian gene that is closely located downstream of PLCB1, was identified as a candidate target gene. Hence, dysregulation of ROR1, PLCB1, or PLCB4 by PAX6 and CTCF may be one mechanism that links neural and pancreatic dysfunction not only in insomnia but also in the relevant psychiatric disorders that are accompanied with circadian rhythm disruption and metabolic syndrome

    Impacts of salt stress on locomotor and transcriptomic responses in the intertidal Gastropod Batillaria attramentaria

    Salinity is one of the most crucial environmental factors that structures biogeographic boundaries of aquatic organisms, affecting distribution, abundance, and behavior. However, the association between behavior and gene regulation underlying acclimation to changes in salinity remains poorly understood. In this study, we investigated the effects of salinity stress on behavior (movement distance) and patterns of gene expression (using RNA sequencing) of the intertidal gastropod Batillaria attramentaria. We examined responses to short-term (1-hour) and long-term (30-day) acclimation to a range of salinities (43, 33 [control], 23, 13, and 3 psu). We found that the intertidal B. attramentaria is able to tolerate a broad range of salinity from 13 to 43 psu but not the acute low salinity of 3 psu. Behavioral experiments showed that salt stress significantly influenced snails’ movement, with lower salinity resulting in shorter movement distance. Transcriptomic analyses revealed critical metabolic pathways and genes potentially involved in acclimation to salinity stress, including ionic and osmotic regulation, signal and hormonal transduction pathways, water exchange, cell protection, and gene regulation or epigenetic modification. In general, our study presents a robust, integrative laboratory-based approach to investigate the effects of salt stress on a nonmodel gastropod facing detrimental consequences of environmental change. The current genetic results provide a wealth of reference data for further research on mechanisms of ionic and osmotic regulation and adaptive evolution of this coastal gastropod

    Underwater Optical-Sonar Image Fusion Systems

    Unmanned underwater operations using remotely operated vehicles or unmanned surface vehicles are increasing in recent times, and this guarantees human safety and work efficiency. Optical cameras and multi-beam sonars are generally used as imaging sensors in underwater environments. However, the obtained underwater images are difficult to understand intuitively, owing to noise and distortion. In this study, we developed an optical and sonar image fusion system that integrates the color and distance information from two different images. The enhanced optical and sonar images were fused using calibrated transformation matrices, and the underwater image quality measure (UIQM) and underwater color image quality evaluation (UCIQE) were used as metrics to evaluate the performance of the proposed system. Compared with the original underwater image, image fusion increased the mean UIQM and UCIQE by 94% and 27%, respectively. The contrast-to-noise ratio was increased six times after applying the median filter and gamma correction. The fused image in sonar image coordinates showed qualitatively good spatial agreement and the average IoU was 75% between the optical and sonar pixels in the fused images. The optical-sonar fusion system will help to visualize and understand well underwater situations with color and distance information for unmanned works

    Hypoglycemic Effect of an Herbal Decoction (Modified Gangsimtang) in a Patient with Severe Type 2 Diabetes Mellitus Refusing Oral Anti-Diabetic Medication: A Case Report

    There is growing interest in alternative therapies for type 2 diabetes mellitus (T2DM) because some patients refuse to receive conventional therapies. In East Asia, herbal medicines are often used to treat T2DM, and modified Gangsimtang (mGST) is prescribed to treat a condition called wasting thirst (消渴), which resembles T2DM. This study reported the treatment of hyperglycemia using herbal medicines without oral hypoglycemic agents or insulin therapy. Case presentation: A 36-year-old man with obesity was diagnosed with T2DM four years prior to hospitalization and experienced blood glucose level reduction from 22.2–27.8 mmol/L (400–500 mg/dL) to 5.6–11.1 mmol/L (100–200 mg/dL) by using herbal medicines. He visited D Korean Medicine Hospital with chronic polydipsia and general weakness as chief complaints. He was diagnosed with T2DM on the basis of a hemoglobin A1c level of 11.7% and 2 h postprandial blood glucose level of >25.0 mmol/L (450 mg/dL). Moreover, he was diagnosed with a “dual deficiency of qi and yin” (氣陰兩虛) because of ordinary symptoms (素證). During his 30-day inpatient treatment, the patient received mGST 120 mL thrice daily; as a result, his postprandial blood glucose level decreased from 25.3 mmol/L (455 mg/dL) to 8.6 mmol/L (154 mg/dL), polydipsia decreased (visual analog scale score decreased from six to one), and triglyceride levels decreased from 11.7 mmol/L (1031 mg/dL) to 2.0 mmol/L (174 mg/dL). Plasma glucose levels remained stable for 6 months after the treatment, and no adverse events were observed over 200 days. We administered an herbal decoction to decrease plasma glucose levels without using oral hypoglycemic agents or insulin. Conclusions: Herbal decoctions such as mGST can reduce hyperglycemia in patients with T2DM who refuse conventional therapy

    Impact of Transcriptional Properties on Essentiality and Evolutionary Rate

    We characterized general transcriptional activity and variability of eukaryotic genes from global expression profiles of human, mouse, rat, fly, plants, and yeast. The variability shows a higher degree of divergence between distant species, implying that it is more closely related to phenotypic evolution, than the activity. More specifically, we show that transcriptional variability should be a true indicator of evolutionary rate. If we rule out the effect of translational selection, which seems to operate only in yeast, the apparent slow evolution of highly expressed genes should be attributed to their low variability. Meanwhile, rapidly evolving genes may acquire a high level of transcriptional variability and contribute to phenotypic variations. Essentiality also seems to be correlated with the variability, not the activity. We show that indispensable or highly interactive proteins tend to be present in high abundance to maintain a low variability. Our results challenge the current theory that highly expressed genes are essential and evolve slowly. Transcriptional variability, rather than transcriptional activity, might be a common indicator of essentiality and evolutionary rate, contributing to the correlation between the two variables
