    Reprogramming of various cell types to a beta-like state by Pdx1, Ngn3 and MafA

    The three transcription factors, PDX1, NGN3 and MAFA, are very important in pancreatic development. Overexpression of these three factors can reprogram both pancreatic exocrine cells and SOX9-positive cells of the liver into cells resembling pancreatic beta cells. In this study we investigate whether other cell types can be reprogrammed. Eight cell types are compared and the results are consistent with the idea that reprogramming occurs to a greater degree for developmentally related cells (pancreas, liver) than for other types, such as fibroblasts. Using a line of mouse hepatocyte-derived cells we screened 13 compounds for the ability to increase the yield of reprogrammed cells. Three are active and when used in combination they can increase the yield of insulin-immunopositive cells by a factor of six. These results should contribute to the eventual ability to develop a new cure for diabetes based on the ability to reprogram other cells in the body to a beta cell phenotype

    Generators and commutators in finite groups; abstract quotients of compact groups

    Let N be a normal subgroup of a finite group G. We prove that under certain (unavoidable) conditions the subgroup [N,G] is a product of commutators [N,y] (with prescribed values of y from a given set Y) of length bounded by a function of d(G) and |Y| only. This has several applications: 1. A new proof that G^n is closed (and hence open) in any finitely generated profinite group G. 2. A finitely generated abstract quotient of a compact Hausdorff group must be finite. 3. Let G be a topologically finitely generated compact Hausdorff group. Then G has a countably infinite abstract quotient if and only if G has an infinite virtually abelian continuous quotient.Comment: This paper supersedes the preprint arXiv:0901.0244v2 by the first author and answers the questions raised there.     A graph-based integration of multimodal brain imaging data for the detection of early mild cognitive impairment (E-MCI)

    Alzheimer's disease (AD) is the most common cause of dementia in older adults. By the time an individual has been diagnosed with AD, it may be too late for potential disease modifying therapy to strongly influence outcome. Therefore, it is critical to develop better diagnostic tools that can recognize AD at early symptomatic and especially pre-symptomatic stages. Mild cognitive impairment (MCI), introduced to describe a prodromal stage of AD, is presently classified into early and late stages (E-MCI, L-MCI) based on severity. Using a graph-based semi-supervised learning (SSL) method to integrate multimodal brain imaging data and select valid imaging-based predictors for optimizing prediction accuracy, we developed a model to differentiate E-MCI from healthy controls (HC) for early detection of AD. Multimodal brain imaging scans (MRI and PET) of 174 E-MCI and 98 HC participants from the Alzheimer's Disease Neuroimaging Initiative (ADNI) cohort were used in this analysis. Mean targeted region-of-interest (ROI) values extracted from structural MRI (voxel-based morphometry (VBM) and FreeSurfer V5) and PET (FDG and Florbetapir) scans were used as features. Our results show that the graph-based SSL classifiers outperformed support vector machines for this task and the best performance was obtained with 66.8% cross-validated AUC (area under the ROC curve) when FDG and FreeSurfer datasets were integrated. Valid imaging-based phenotypes selected from our approach included ROI values extracted from temporal lobe, hippocampus, and amygdala. Employing a graph-based SSL approach with multimodal brain imaging data appears to have substantial potential for detecting E-MCI for early detection of prodromal AD warranting further investigation

    Asymptotic and small sample statistical properties of random frailty variance estimates for shared gamma frailty models

    This paper concerns maximum likelihood estimation for the semiparametric shared gamma frailty model; that is the Cox proportional hazards model with the hazard function multiplied by a gamma random variable with mean 1 and variance θ. A hybrid ML-EM algorithm is applied to 26400 simulated samples of 400 to 8000 observations with Weibull hazards. The hybrid algorithm is much faster than the standard EM algorithm, faster than standard direct maximum likelihood (ML, Newton Raphson) for large samples, and gives almost identical results to the penalised likelihood method in S-PLUS 2000. When the true value θ0 of θ is zero, the estimates of θ are asymptotically distributed as a 50-50 mixture between a point mass at zero and a normal random variable on the positive axis. When θ0 > 0> the asymptotic distribution is normal. However, for small samples, simulations suggest that the estimates of θ are approximately distributed as an x -(100 -x)% mixture, 0≤ x ≤50, between a point mass at zero and a normal random variable on the positive axis even for θ0 > 0. In light of this, p-values and confidence intervals need to be adjusted accordingly. We indicate an approximate method for carrying out the adjustment

    Treatment of venous thromboembolism with low-molecular-weight heparin: a synthesis of the evidence published in systematic literature reviews

    Objective: To evaluate the methodology and cumulative evidence presented in systematic reviews of clinical trials comparing low-molecular-weight heparin (LMWH) with unfractionated heparin (UFH) for the treatment of venous thromboembolism. Methods: We reviewed all systematic reviews of clinical trials published until March 2002. Fourteen systematic literature reviews were published between 1994 and 2000. Deficiencies in methodological quality were common, particularly in the description of search strategies, assessment of clinical trial quality, and methods used to combine results. Results: Results of reviews indicate that LMWH is superior to UFH for the treatment of venous thromboembolism, particularly in reducing mortality. Patients with isolated deep venous thrombosis or deep venous thrombosis with concomitant pulmonary embolism seemed to have similar benefit. However, the benefits of LMWH over UFH were smaller in magnitude in reviews that included more recent clinical trials
