Determinação da vitamina E em avelãs: optimização e validação do método analÃtico.
Neste trabalho foi desenvolvido e validado um método analÃtico para a separação,
identificação e quantificação simultânea de 4 tocoferóis e 4 tocotrienóis, posteriormente
aplicado a amostras de avelãs de diferentes cultivares.Foram ensaiados três métodos
extractivos (extracção com soxtec, extracção sólido-liquido com e sem saponificação). Os
melhores resultados foram obtidos com uma extracção sólido-lÃquido simplificada
utilizando n-hexano para a extracção dos lÃpidos. A separação cromatográfica realizou-se
numa coluna de fase normal (Inertsil 5 SI) utilizando, como fase móvel, uma mistura de nhexano/
1,4-dioxano (95,5:4,5, v/v) com um fluxo de 0,7 ml/min (eluição isocrática). A
eluição foi monitorizada por um detector de fluorescência ligado em série com um detector
de dÃodos (DAD). A quantificação dos compostos foi feita pelo método do padrão interno
utilizando tocol como substância de referência. Todos os compostos foram separados
num curto perÃodo de tempo (17 min). O método mostrou ser rápido, sensÃvel, preciso e
reprodutÃvel, permitindo a determinação simultânea de todos os constituintes da vitamina
E.
Nas amostras de avelãs foram identificados e quantificados 6 compostos: α-, β-, γ- e δ-
tocoferóis e β- e γ-tocotrienóis. α-Tocotrienol foi detectado apenas em algumas das
cultivares estudadas
BenefÃcios potenciais do consumo de nozes
Os frutos secos, na sua generalidade, apresentam uma composição similar do ponto de vista nutricional sendo, caracteristicamente, alimentos com elevado teor de lÃpidos. Por este motivo, a generalidade dos consumidores evita o seu consumo, uma vez que associa este tipo de alimentos ao aumento de peso corporal. No entanto, estudos epidemiológicos e clÃnicos realizados nas últimas décadas sugerem que o consumo frequente de quantidades moderadas de frutos secos pode ter efeitos benéficos para a saúde humana, principalmente a nÃvel do perfil lipÃdico sanguÃneo.13
Quando comparadas com a maioria dos outros frutos secos, os -quais contêm principalmente ácidos gordos monoinsaturados, as nozes destacam-se pelos seus teores elevados em ácidos gordos polinsaturados E -3 e 0-6, nomeadamente em termos dos ácidos gordos essenciais linoleico e 0-linolénico. Para além do seu perfil favorável em ácidos gordos, as nozes contêm ainda vários outros compostos bioactivos, tais como fitoesteróis, tocoferóis e tocotrienóis.
Neste trabalho procedeu-se ao estudo da fracção lipÃdica das nozes em termos do seu perfil em ácidos gordos, triacilgliceróis, fitoesteróis, tocoferóis e tocotrienóis. As composições em ácidos gordos e fitoesteróis foram analisadas por GC/FID, os triacilgliceróis por HPLC/ELSD e os
tocoferóis e tocotrienóis por HPLC/DAD/FL. Os detalhes relativos à composição nos referidos compostos serão apresentados bem como a sua relação com possÃveis efeitos benéficos para a saúde
Influência da torra de avelãs sua composição em vitamina E
Neste trabalho avaliou-se o efeito da torra da avelã (Corylus avellana L.) na sua composição em tocoferóis e tocotrienóis. Para tal, procedeu-se à quantificação dos diferentes vitâmeros em avelãs submetidas a diversas torras, compreendendo diferentes temperaturas (125, 145, 165 e 185 e 200 ºC) e tempos de exposição (5, 15 e 30 min). As amostras foram analizadas utilizando-se uma metodologia de HPLC/DAD/FL previamente validada. Apesar de se verificar uma tendência de perdas crescentes com os aumentos de temperatura e tempo de torra, o decréscimo máximo do teor total comparativamente à amostra crua foi de 9,2%, tendo-se observado comportamentos distintos por parte dos diversos vitâmeros presentes na avelã
Development and evaluation of a normal phase liquid chromatographic method for the determination of tocopherols and tocotrienols in walnuts
A high performance liquid chromatographic (HPLC) method for the determination
of tocopherols and tocotrienols in walnut samples is described. The
compounds were extracted with n-hexane, using a simple solid-liquid extraction
procedure. Tocol was used as internal standard and BHT as anti-oxidant. The ehromatographic
separation was achieved using an Inertsil 5 SI normal phase column
operating with isocratic elution of n-hexane/1,4-dioxane (96.5:3.5, v/v), at a flow
rate of 0.7 mL/min. The effluent was monitored by a series arrangement of a diodearray
detector followed by a fluorescence detector. The detection limits were
low, between 0.037 and 0.266 (jig/mL. The method was precise (% CV less than2.8%), accurate (% CV less than 5.6%), and, as a general rule, the recovery values were
high (mean values ranging from 93.4% to 104%)
Avaliação nutricional e composição em ácidos gordos de 6 variedades de nozes, sementes de Juglans regia L., cultivadas em Trás-os-Montes
A dieta mediterrânea, considerada saudável e uma das melhores no que concerne à prevenção das doenças cardiovasculares inclui, como componentes importantes, os frutos secos. No entanto, e talvez devido ao facto deste tipo de alimentos ser rico em lÃpidos, assistiu-se a um declÃnio do seu consumo. Actualmente, associa-se o consumo de frutos secos à ingestão de ingredientes com propriedades funcionais. Quando comparadas com a maioria dos outros frutos secos, que contam principalmente ácidos gordos monoinsaturados, as nozes distinguem-se pelos teores elevados em ácidos gordos polinsaturados (omega-3 e omega-6)
Evaluation of a numerical method to predict the polyphenol content in monovarietal olive oils
Three monovarietal virgin olive oils obtained from olives grown under biological agricultural system, in Portugal, with different ripening
stages, were studied in what concerns the parameters usually related with oxidation status (total polyphenols, tocopherols, chlorophylls
and carotenoids, peroxide values, oxidative stability, specific extinction coefficients K232 and K270 and titratable acidity). A total
of 18 samples were analyzed: seven from Cv. Cobranc¸osa (maturation indices 1–7), five from Cv. Madural (maturation indices 3–7) and
six from Cv. Verdeal Transmontana (maturation indices 1–6). Oxidative stability and polyphenols profile presented high correlation coef-
ficients. Given this high correlation, a numerical method was developed and evaluated to predict total polyphenols contents in olive oil.
The method is based on the kinetic equation of the oxidation process in the presence of antioxidants and on Rancimat profiles. Total
polyphenols contents obtained with this method were similar to those obtained by the Folin-Ciocalteau method
Phenolic compounds, organic acids profiles and antioxidative properties of beefsteak fungus (Fistulina hepatica)
The phenolic compounds and the organic acids composition of the edible beefsteak fungus Fistulina hepatica was determined by HPLC/DAD and HPLC/UV, respectively. The results showed a profile composed by five phenolic compounds (caffeic, p-coumaric and ellagic acids, hyperoside and quercetin) and six organic acids (oxalic, aconitic, citric, malic, ascorbic and fumaric acids). The quantification of the identified compounds revealed that ellagic acid (ca. 49.7%) and malic acid (ca. 57.9%) are the main compounds in this species. In a general way the phenolic profile revealed to be more constant than the organic acids one and could be more useful for the quality control of the species. Beefsteak fungus was also investigated for its capacity to act as a scavenger of DPPH- radical and reactive oxygen species (superoxide radical, hydroxyl radical and hypochlorous acid). Good results were obtained against DPPH in a concentration-dependent manner. Beefsteak fungus also displayed good activity against superoxide radical, achieved by its capacity to act as both scavenger and xanthine oxidase inhibitor. A prooxidant effect was noticed for hydroxyl radical, which may be due to its capacity for iron ions reduction. Little ability for iron chelation was also observed. Beefsteak fungus showed a weak protective effect against hypochlorous acid
Determination of Sterol and Fatty Acid Compositions, Oxidative Stability, and Nutritional Value of Six Walnut (Juglans regia L.) Cultivars Grown in Portugal
Six cultivars (Franquette, Marbot, Mayette, Mellanaise, Lara, and Parisienne) of walnuts (Juglans regia L.) were collected during the 2001 crop, from Bragança, Portugal. Chemical composition,
including moisture, total oil content, crude protein, ash, carbohydrates, and nutritional value, was
evaluated. Fat was the predominant component, ranging from 62.3 to 66.5%. Total oil was extracted
and analyzed for fatty acids, sterols, oxidative stability, and peroxide value. Fatty acids and sterols
were determined by gas-liquid chromatography coupled to a flame ionization detector. Eighteen
fatty acids were quantified. Polyunsaturated fatty acids and, in particular, linoleic acid were
predominant. â-Sitosterol, ¢5-avenasterol, and campesterol were the major sterols found. Differences
were observed among the studied cultivars, especially in peroxide values and in the sterol profile
Chemical composition, fatty acid and sterol contents of nuts grown in Portugal
Nuts are relevant components of the Mediterranean diet, which is thought to be a healthy one and has been considered as one of the best on coronary heart disease prevention. Although total fat intake is certainly related to health risks, there is now general agreement that the more important issue, where health is concerned, is the type of fat or fatty acids that are consumed. Nuts are also rich in phytosterols, which have been associated with health benefits. Additionally the assessment of identity and quality of vegetable oils obtained from different sources generally involves many tests, including analysis of fatty acids and sterols, once these are probably the most important major and ninor components respectively.
In order to obtain data on composition and nutritional quality of different nuts cultivated in Portugal, different cultivars of hazetnuss (Corylus avellana L.) and walnuts (Juglans regia L.) were analyzed. Chemical composition concerning moisture, total oil content, crude protein, ash and carbohydrates was evaluated. The chemical study also included the determination of sterol and fatty acid compositions. These were determined by gas-liquid chromatography coupled to a flame ionisation detector. Polyunsaturated fatty acids were predominant in walnut samples while monounsaturated were the predominant fatty acids in hazelnuts (beta)-sitosterol, (delta)-avenasterol and campesterol were the major phytosterols in both cases
