    Current ecotoxicity testing needs among selected U.S. federal agencies

    U.S. regulatory and research agencies use ecotoxicity test data to assess the hazards associated with substances that may be released into the environment, including but not limited to industrial chemicals, pharmaceuticals, pesticides, food additives, and color additives. These data are used to conduct hazard assessments and evaluate potential risks to aquatic life (e.g., invertebrates, fish), birds, wildlife species, or the environment. To identify opportunities for regulatory uses of non-animal replacements for ecotoxicity tests, the needs and uses for data from tests utilizing animals must first be clarified. Accordingly, the objective of this review was to identify the ecotoxicity test data relied upon by U.S. federal agencies. The standards, test guidelines, guidance documents, and/or endpoints that are used to address each of the agencies’ regulatory and research needs regarding ecotoxicity testing are described in the context of their application to decision-making. Testing and information use, needs, and/or requirements relevant to the regulatory or programmatic mandates of the agencies taking part in the Interagency Coordinating Committee on the Validation of Alternative Methods Ecotoxicology Workgroup are captured. This information will be useful for coordinating efforts to develop and implement alternative test methods to reduce, refine, or replace animal use in chemical safety evaluations

    Evolutionary Trends of the Pharyngeal Dentition in Cypriniformes (Actinopterygii: Ostariophysi)

    International audienceBACKGROUND: The fish order Cypriniformes is one of the most diverse ray-finned fish groups in the world with more than 3000 recognized species. Cypriniformes are characterized by a striking distribution of their dentition: namely the absence of oral teeth and presence of pharyngeal teeth on the last gill arch (fifth ceratobranchial). Despite this limited localisation, the diversity of tooth patterns in Cypriniformes is astonishing. Here we provide a further description of this diversity using X-ray microtomography and we map the resulting dental characters on a phylogenetic tree to explore evolutionary trends. RESULTS: We performed a pilot survey of dental formulae and individual tooth shapes in 34 adult species of Cypriniformes by X-ray microtomography (using either conventional X-ray machine, or synchrotron microtomography when necessary) or by dissecting. By mapping morphological results in a phylogenetic tree, it emerges that the two super-families Cobitoidea and Cyprinoidea have followed two distinct evolutionary pathways. Furthermore, our analysis supports the hypothesis of a three-row dentition as ancestral for Cyprinoidea and a general trend in tooth row reduction in most derived lineages. Yet, this general scheme must be considered with caution as several events of tooth row gain and loss have occurred during evolutionary history of Cyprinoidea. SIGNIFICANCE: Dentition diversity in Cypriniformes constitutes an excellent model to study the evolution of complex morphological structures. This morphological survey clearly advocates for extending the use of X-ray microtomography to study tooth morphology in Cypriniformes. Yet, our survey also underlines that improved knowledge of Cypriniformes life traits, such as feeding habits, is required as current knowledge is not sufficient to conclude on the link between diet and dental morphology

    BHPR research: qualitative1. Complex reasoning determines patients' perception of outcome following foot surgery in rheumatoid arhtritis

    Background: Foot surgery is common in patients with RA but research into surgical outcomes is limited and conceptually flawed as current outcome measures lack face validity: to date no one has asked patients what is important to them. This study aimed to determine which factors are important to patients when evaluating the success of foot surgery in RA Methods: Semi structured interviews of RA patients who had undergone foot surgery were conducted and transcribed verbatim. Thematic analysis of interviews was conducted to explore issues that were important to patients. Results: 11 RA patients (9 ♂, mean age 59, dis dur = 22yrs, mean of 3 yrs post op) with mixed experiences of foot surgery were interviewed. Patients interpreted outcome in respect to a multitude of factors, frequently positive change in one aspect contrasted with negative opinions about another. Overall, four major themes emerged. Function: Functional ability & participation in valued activities were very important to patients. Walking ability was a key concern but patients interpreted levels of activity in light of other aspects of their disease, reflecting on change in functional ability more than overall level. Positive feelings of improved mobility were often moderated by negative self perception ("I mean, I still walk like a waddling duck”). Appearance: Appearance was important to almost all patients but perhaps the most complex theme of all. Physical appearance, foot shape, and footwear were closely interlinked, yet patients saw these as distinct separate concepts. Patients need to legitimize these feelings was clear and they frequently entered into a defensive repertoire ("it's not cosmetic surgery; it's something that's more important than that, you know?”). Clinician opinion: Surgeons' post operative evaluation of the procedure was very influential. The impact of this appraisal continued to affect patients' lasting impression irrespective of how the outcome compared to their initial goals ("when he'd done it ... he said that hasn't worked as good as he'd wanted to ... but the pain has gone”). Pain: Whilst pain was important to almost all patients, it appeared to be less important than the other themes. Pain was predominately raised when it influenced other themes, such as function; many still felt the need to legitimize their foot pain in order for health professionals to take it seriously ("in the end I went to my GP because it had happened a few times and I went to an orthopaedic surgeon who was quite dismissive of it, it was like what are you complaining about”). Conclusions: Patients interpret the outcome of foot surgery using a multitude of interrelated factors, particularly functional ability, appearance and surgeons' appraisal of the procedure. While pain was often noted, this appeared less important than other factors in the overall outcome of the surgery.     Laboratory study of the effects of citric and ascorbic acids on injections prepared with brown heroin

    The addition of acidic substances to brown street heroin to facilitate the solubility of diamorphine in the injection preparation process is commonplace amongst UK injectors. Knowledge of the chemistry behind this process supports the need for this stage in the injection preparation process. It is currently illegal, under the Misuse of Drugs Act 1971, section 9A, to supply acidifiers and other paraphernalia to injectors in the UK. In the current climate of evidence-based practice, any consideration given to changing the law would look for evidence to illustrate that the paraphernalia was necessary. Although the theory behind the use of acidifiers suggests they are essential, no previous work using street heroin has actually been reported to illustrate this fact. Anecdotal information has found that drug users are being told by some service providers that the addition of acids is unnecessary. It is important that drugs services give credible information to their clients. The provision of inaccurate information in one area may lead to a lack of trust of all information provided. The small study reported here investigated, under controlled laboratory conditions, the effects of citric and ascorbic acids (vitamin C) on injections prepared with brown heroin, in order to demonstrate the need for acidifiers in the injection preparation process

    Investigation into the effectiveness of filters for use by intravenous drug users

    Injecting drug users are at risk of and can suffer from serious health problems due to intravenous administration of insoluble particles from street drugs and tablets. Makeshift filters are used to try to remove particles from the injections. The decision to distribute filters from Harm Reduction Centres cannot be based on evidence of efficacy and safety as no such published data exists. The work presented here is part of a larger study which anticipates to provide such evidence. Laboratory methods were developed based on information gathered from drug users. Injections were prepared using drugs of abuse and filtered either through makeshift or commercially available filters. The resulting solutions were assayed for particle size and number (using Coulter Counter®). Comparisons were made with unfiltered injections. The results presented here are from the first part of this study, which investigated the effectiveness of various filters on reducing particulate count and size range of injections made from tablets. The commercially produced filter (Acrodisk®) showed a much greater reduction in particle number and size. This strongly suggests the risks to health could be reduced by the use of such filters. Of the makeshift filters, the Rizla® acetate filter showed the most satisfactory reduction in the number of particles, suggesting their use may be preferable to the currently popular cigarette filter. The ongoing work is looking at the effect of the filtration methods on amount of drug in injections made from tablets and repeating the work using street heroin
