Bei diesem Beitrag handelt es sich um die Schriftfassung der Dritten Wilhelm-Röpke-Vorlesung, die der stellvertretende Chefredaktor und Leiter der Wirtschaftsredaktion der Neuen Zürcher Zeitung, Dr. Gerhard Schwarz, am 12. Februar 2009 in der Thüringer Aufbaubank in Erfurt gehalten hat. Die jährlich am 12. Februar, dem Todestag des großen Wirtschafts- und Sozialwissenschaftlers, stattfindende Veranstaltung des Wilhelm-Röpke-Instituts e.V. soll dazu beitragen, Röpkes Wirken als Wissenschaftler, Politikberater und Publizist in Erinnerung zu rufen und seine Ideen auf aktuelle Probleme anzuwenden. --

Peptide–liposome association. A critical examination with mastoparan-X

AbstractMastoparan-X, a wasp venom factor, is a membrane active peptide whose binding to lipid vesicles is of basic interest towards an analysis of its functions. Titration of aqueous peptide solutions with liposomes allows the determination of the association isotherm, i.e. a plot of bound peptide per lipid versus the free peptide concentration. We have scrutinised the various steps in the evaluation procedure, considering circular dichroism as well as fluorescence intensity as possible signals for the binding process. First of all the measured data had to be corrected for light scattering effects which may otherwise appreciably falsify the final results. Uncertainties due to inherent difficulties regarding the reproducibility of lipid preparations and inevitable titration errors have to be considered. The consequences of these errors for the quantitative analysis of the titration curves were investigated. The plotted curves can be reasonably well fitted by a functional relationship derived from a Gouy–Chapman model approach that assumes a partitioning of monomeric peptide. The two relevant parameters, partition coefficient and effective charge number, and their error ranges have been determined for mastoparan-X and a series of phosphatidylcholine vesicle sizes and various ionic strengths. These findings show that the applied analysis implies a sufficient basis for calculations of the amount of lipid bound peptide in practice. However, the possible existence of peptide aggregates cannot generally be excluded from a formal monomer associated curve fit as indicated by computer simulations

Registerforschung: Verwaltungs- und Statistikdaten für die Wissenschaft

Die Verfügbarkeit von Daten aus der Verwaltung sowie der Bundesstatistik für die (Grundlagen-)Forschung wird zunehmend ein Faktor für die Wettbewerbsfähigkeit eines Forschungsstandortes – insbesondere in den Sozialwissenschaften aber auch der Gesundheitswissenschaft – und könnte einen wichtigen Beitrag für eine Evidenzorientierung in der Politik und Verwaltung liefern. In Österreich bildet vor allem die vergleichsweise unklare und zerklüftete Rechtslage in diesem Bereich sowie politische Widerstände gegen einen Zugang zu diesen Daten für die Wissenschaft eine wesentliche Herausforderung. Aus internationaler Perspektive ist aber die Öffnung dieser Daten geradezu notwendig und ein Gebot der Stunde

Ein Mathematiker mit universalem Anspruch : über Max Dehn und sein Wirken am Mathematischen Seminar

Für eine erste Blüte der Mathematik in Frankfurt gab Max Dehn (1878 –1952) in den Jahren ab 1921 bis 1935 entscheidende Impulse. Seine völlig neuen Ideen zur Knotentheorie und zur Topologie beeinflussten die Entwicklung der Mathematik weit über Deutschland hinaus. 1935 fand sein Wirken in Frankfurt durch den Terror der Nationalsozialisten ein jähes Ende. Nach einer gefahrvollen Flucht über Norwegen, Finnland, die Sowjetunion und Japan erreichte Dehn schließlich, 62-jährig, die Vereinigten Staaten von Nordamerika. Eine seinen Fähigkeiten entsprechende Stellung konnte er dort nicht mehr erlangen. Sein fünfzigster Todestag in diesem Jahr ist Anlass für diese Rückschau

Serum response factor is crucial for actin cytoskeletal organization and focal adhesion assembly in embryonic stem cells

The activity of serum response factor (SRF), an essential transcription factor in mouse gastrulation, is regulated by changes in actin dynamics. Using Srf(−/−) embryonic stem (ES) cells, we demonstrate that SRF deficiency causes impairments in ES cell spreading, adhesion, and migration. These defects correlate with defective formation of cytoskeletal structures, namely actin stress fibers and focal adhesion (FA) plaques. The FA proteins FA kinase (FAK), β1-integrin, talin, zyxin, and vinculin were downregulated and/or mislocalized in ES cells lacking SRF, leading to inefficient activation of the FA signaling kinase FAK. Reduced overall actin expression levels in Srf(−/−) ES cells were accompanied by an offset treadmilling equilibrium, resulting in lowered F-actin levels. Expression of active RhoA-V14 rescued F-actin synthesis but not stress fiber formation. Introduction of constitutively active SRF-VP16 into Srf(−/−) ES cells, on the other hand, strongly induced expression of FA components and F-actin synthesis, leading to a dramatic reorganization of actin filaments into stress fibers and lamellipodia. Thus, using ES cell genetics, we demonstrate for the first time the importance of SRF for the formation of actin-directed cytoskeletal structures that determine cell spreading, adhesion, and migration.
