Spatially Stable Mitochondrial Compartments Fuel Local Translation during Plasticity
Local translation meets protein turnover and plasticity demands at synapses, however, the location of its energy supply is unknown. We found that local translation in neurons is powered by mitochondria and not by glycolysis. Super-resolution microscopy revealed that dendritic mitochondria exist as stable compartments of single or multiple filaments. To test if these mitochondrial compartments can serve as local energy supply for synaptic translation, we stimulated individual synapses to induce morphological plasticity and visualized newly synthesized proteins. Depletion of local mitochondrial compartments abolished both the plasticity and the stimulus-induced synaptic translation. These mitochondrial compartments serve as spatially confined energy reserves, as local depletion of a mitochondrial compartment did not affect synaptic translation at remote spines. The length and stability of dendritic mitochondrial compartments and the spatial functional domain were altered by cytoskeletal disruption. These results indicate that cytoskeletally tethered local energy compartments exist in dendrites to fuel local translation during synaptic plasticity
Super-resolution imaging and estimation of protein copy numbers at single synapses with DNA-PAINT
In the brain, the strength of each individual synapse is defined by the complement of proteins present or the "local proteome." Activity-dependent changes in synaptic strength are the result of changes in this local proteome and posttranslational protein modifications. Although most synaptic proteins have been identified, we still know little about protein copy numbers in individual synapses and variations between synapses. We use DNA-point accumulation for imaging in nanoscale topography as a single-molecule super-resolution imaging technique to visualize and quantify protein copy numbers in single synapses. The imaging technique provides near-molecular spatial resolution, is unaffected by photobleaching, enables imaging of large field of views, and provides quantitative molecular information. We demonstrate these benefits by accessing copy numbers of surface AMPA-type receptors at single synapses of rat hippocampal neurons along dendritic segments
Teaching old NCATs new tricks: using non-canonical amino acid tagging to study neuronal plasticity
The non-canonical amino acid labeling techniques BONCAT (bioorthogonal non-canonical amino acid tagging) and FUNCAT (fluorescent non-canonical amino acid tagging) enable the specific identification and visualization of newly synthesized proteins. Recently, these techniques have been applied to neuronal systems to elucidate protein synthesis dynamics during plasticity, identify stimulation-induced proteomes and subproteomes and to investigate local protein synthesis in specific subcellular compartments. The next generation of tools and applications, reviewed here, includes the development of new tags, the quantitative identification of newly synthesized proteins, the application of NCAT to whole animals, and the ability to genetically restrict NCAT labeling. These techniques will enable not only improved detection but also allow new scientific questions to be tackled
A genetically encodable cell-type-specific protein synthesis inhibitor
Chemical inhibitors have revealed requirements for protein synthesis that drive cellular plasticity. We developed a genetically encodable protein synthesis inhibitor (gePSI) to achieve cell-type-specific temporal control of protein synthesis. Controlled expression of the gePSI in neurons or glia resulted in rapid, potent and reversible cell-autonomous inhibition of protein synthesis. Moreover, gePSI expression in a single neuron blocked the structural plasticity induced by single-synapse stimulation

Grondsmaak is wereldwijd een van de belangrijkste beperkende factoren voor de afzet van kweekvis afkomstig uit vijvers en recirculatiesystemen. Dit wordt vooral veroorzaakt door de lage waardering van consumenten voor kweekvis met grondsmaak. Marktprijs en marktvolume komen daardoor onder druk te staan. Daarnaast brengt de bestrijding van grondsmaak extra kosten met zich mee voor de producent. Ook in Nederlandse kweekvis komt grondsmaak voor. In Nederland wordt op verschillende fronten gewerkt aan het voorkomen en bestrijden van grondsmaak. In dit artikel (dat uit twee delen bestaat) wordt het grondsmaakprobleem en het daaraan gekoppelde onderzoek beschreven. Fiberoptic Colonoscopic Polypectomies
The development of fiberoptic instruments which permit direct visualization of the entire colonic lumen has made possible the safe removal of polyps from any part of the colon. The avoidance of transabdominal colotomy and polypectomy significantly decreases the mortality and morbidity as well as the duration of hospital stay. We wish to present our experience in performing the endoscopic removal of 128 colonic polyps larger than 0.5 cm in size in 103 patients, located from the cecum to the sigmoid colon and beyond the reach of the rigid sigmoidoscope. The polypectomy procedures via the colonoscope were done without mortality and with only three relatively minor complications, all of which responded to conservative measures. Endoscopic removal of colonic polyps is proposed as a safe, practical alternative to either laparotomy and colotomy or repeated barium enema studies in the management of the patient with a colonic polyp
