    Assessing the Hydraulics of Water Heaters by Adding Fluoride as a Tracer to Inform its Overall Effect on Water Quality

    As residential water heaters are the primary source of waterborne disease outbreaks in the U.S. there is a need to better understand how they are contributing to decreased water quality in buildings. Previous work has indicated that intermittent stagnation, low disinfection residual and operation temperatures promote the growth of pathogens in water heaters. The overall effect of hydrodynamics of water heaters on these factors remains unclear. Therefore, a tracer study was performed to determine the effect of and characterize hydraulics of the typical residential water heater on water quality. Controlling temperature and flow rate, a pilot-scale hot water system (50 gal) with specially adapted fittings was used. The fittings allowed for injection of fluoride as a tracer. 4.40 mg/L fluoride solution was injected at room temperature into the inlet line, set at 2.50 gal/min, to derive a step input fluoride concentration. Samples were obtained at the outlet of the tank, then fluoride concentration was measured by ion chromatography. A model was derived numerically in Matlab to validate if the experimental data reflected the behavior of a continuous stirred tank reactor. The data was consistent with the model but deviated at high flow rate and temperature conditions indicating that dispersion effects may be a factor. Additional tests with this tracer can help to better evaluate hydrodynamics of the system and thus its impact on water quality before it reaches the point of use

    Searching for epistatic interactions in nuclear families using conditional linkage analysis

    BACKGROUND: Genomic screens generally employ a single-locus strategy for linkage analysis, but this may have low power in the presence of epistasis. Ordered subsets analysis (OSA) is a method for conditional linkage analysis using continuous covariates. METHODS: We used OSA to evaluate two-locus interactions in the simulated Genetic Analysis Workshop 14 dataset. We used all nuclear families ascertained by Aipotu, Karangar, and Danacaa. Using the single-nucleotide polymorphism map, multipoint affected-sibling-pair (ASP) linkage analysis was performed on all 100 replicates for each chromosome using SIBLINK. OSA was used to examine linkage on each chromosome using LOD scores at each 3-cM location on every other chromosome as covariates. Two methods were used to identify positive results: one searching across the entire covariate chromosome, the other conditioning on location of known disease loci. RESULTS: Single-locus linkage analysis revealed very high LOD scores for disease loci D1 through D4, with mean LOD scores over 100 replicates ranging from 4.0 to 7.8. Although OSA did not obscure this linkage evidence, it did not detect the simulated interactions between any of the locus pairs. We found inflated type I error rates using the first OSA method, highlighting the need to correct for multiple comparisons. Therefore, using "null chromosome pairs" without simulated disease loci, we calculated a corrected alpha-level. CONCLUSION: We were unable to detect two-locus interactions using OSA. This may have been due to lack of incorporation of phenotypic subgroups, or because linkage evidence as summarized by LOD scores performs poorly as an OSA covariate. We found inflated type I error rates, but were able to calculate a corrected alpha-level for future analyses employing this strategy to search for two-locus interactions

    The effect of adding fluorine to the Vanguard first-stage oxidant was anlyzed. An increase in specific impulse of 5.74 percent may be obtained with 30 percent fluorine. This increase, coupled with increased mass ratio due to greater oxidant density, gave up to 24.6-percent increase in first-stage burnout energy with 30 percent fluorine added. However, a change in tank configuration is required to accommodate the higher oxidant-fuel ratio necessary for peak specific impulse with fluorine addition

    Solar Forcing of Florida Straits Surface Salinity During the Early Holocene

    Previous studies showed that sea surface salinity (SSS) in the Florida Straits as well as Florida Current transport covaried with changes in North Atlantic climate over the past two millennia. However, little is known about earlier Holocene hydrographic variability in the Florida Straits. Here, we combine Mg/Ca-paleothermometry and stable oxygen isotope measurements on the planktonic foraminifera Globigerinoides ruber (white variety) from Florida Straits sediment core KNR166-2 JPC 51 (24° 24.70\u27 N, 83° 13.14\u27 W, 198 m deep) to reconstruct a high-resolution (~25 yr/sample) early to mid Holocene record of sea surface temperature and δ18OSW)(a proxy for SSS) variability. After removing the influence of globalδ18OSW change due to continental ice volume variability, we find that early Holocene SSS enrichments are associated with increased evaporation/precipitation ratios in the Florida Straits during periods of reduced solar forcing, increased ice rafted debris in the North Atlantic and the development of more permanent El Niño-like conditions in the eastern equatorial Pacific. When considered with previous high-resolution reconstructions of Holocene tropical atmospheric circulation changes, our results provide evidence that variations in solar forcing over the early Holocene had a significant impact on the global tropical hydrologic cycle

    We explore why venture capital funds limit the amount of capital they raise and do not reinvest the proceeds. This structure is puzzling because it leads to a succession of several funds financing each new venture which multiplies the well known agency problems. We argue that an inside investor cannot provide a hard budget constraint while a less well informed outsider can. Therefore, the venture capitalist delegates the continuation decision to the outsider by ex ante restricting the amount of capital he has under management. The soft budget constraint problem becomes the more important the higher the entrepreneur’s private benefits are and the higher the probability of failure of a project is

    Clastic Sediments in the Butler Cave – Sinking Creek System, Virginia, USA

    The Butler Cave - Sinking Creek System in Bath County, Virginia, consists of a master trunk passage along the axis of a syncline with a trellis arrangement of dip-oriented side caves. The western set of dip passages contain a sequence of massively and chaotically bedded sand and cobble sediments. Massive cobble fills also occur in the strike-oriented trunk passage. Cave passages on the eastern side of the syncline contain mostly sand and silt. The light fraction of the sediments consists predominantly of quartz and rock fragments. The sediments contain several percent heavy minerals composed of iron oxides, zircon, rutile, tourmaline and other minerals. Measurement of the visible and near infrared diffuse reflectance spectra shows at least three populations of sediments to be present: an iron-rich, clay-poor group; a clay-rich group; and a gypsiferous sediment. The iron minerals provided a paleomagnetic signal. Sediments from the trunk passage, deposited by recent underground drainage, contained a normal pole direction. Sediments from the dip passages were paleomagnetically reversed, showing the deposition dates from prior to 780,000 years. In one instance reversed polarity deposits overlie normal polarity, implying a minimum age of 990,000 years for the reversed sediments.   

    Integrase-deficient lentiviral vectors mediate efficient gene transfer to human vascular smooth muscle cells with minimal genotoxic risk

    We have previously shown that injury-induced neointima formation was rescued by adenoviral-Nogo-B gene delivery. Integrase-competent lentiviral vectors (ICLV) are efficient at gene delivery to vascular cells but present a risk of insertional mutagenesis. Conversely, integrase-deficient lentiviral vectors (IDLV) offer additional benefits through reduced mutagenesis risk, but this has not been evaluated in the context of vascular gene transfer. Here, we have investigated the performance and genetic safety of both counterparts in primary human vascular smooth muscle cells (VSMC) and compared gene transfer efficiency and assessed the genotoxic potential of ICLVs and IDLVs based on their integration frequency and insertional profile in the human genome. Expression of enhanced green fluorescent protein (eGFP) mediated by IDLVs (IDLV-eGFP) demonstrated efficient transgene expression in VSMCs. IDLV gene transfer of Nogo-B mediated efficient overexpression of Nogo-B in VSMCs, leading to phenotypic effects on VSMC migration and proliferation, similar to its ICLV version and unlike its eGFP control and uninfected VSMCs. Large-scale integration site analyses in VSMCs indicated that IDLV-mediated gene transfer gave rise to a very low frequency of genomic integration compared to ICLVs, revealing a close-to-random genomic distribution in VSMCs. This study demonstrates for the first time the potential of IDLVs for safe and efficient vascular gene transfer
