    Continental drift, seafloor spreading, plate tectonics: These terms conjure up a picture of the whole of Earth\u27s lithospheric plates in motion, a picture that truly represents a revolution in the earth sciences that took place in the 1960s and permanently changed our view of a more static world. If someone were to ask which subdiscipline of the geosciences has provided the crucial quantitative evidence about the past locations of discrete parts of continental and oceanic plates, the answer would be geomagnetism and paleomagnetism. Polarity stratigraphy, based on radiometrically dated 180° reversals of the dipolar geomagnetic field, informs us about the locations of parts of the seafloor in the past, and paleomagnetically determined paleolatitudes of continental rocks provide similar information about past locations of continental plates

    Clastic Sediments in the Butler Cave – Sinking Creek System, Virginia, USA

    The Butler Cave - Sinking Creek System in Bath County, Virginia, consists of a master trunk passage along the axis of a syncline with a trellis arrangement of dip-oriented side caves. The western set of dip passages contain a sequence of massively and chaotically bedded sand and cobble sediments. Massive cobble fills also occur in the strike-oriented trunk passage. Cave passages on the eastern side of the syncline contain mostly sand and silt. The light fraction of the sediments consists predominantly of quartz and rock fragments. The sediments contain several percent heavy minerals composed of iron oxides, zircon, rutile, tourmaline and other minerals. Measurement of the visible and near infrared diffuse reflectance spectra shows at least three populations of sediments to be present: an iron-rich, clay-poor group; a clay-rich group; and a gypsiferous sediment. The iron minerals provided a paleomagnetic signal. Sediments from the trunk passage, deposited by recent underground drainage, contained a normal pole direction. Sediments from the dip passages were paleomagnetically reversed, showing the deposition dates from prior to 780,000 years. In one instance reversed polarity deposits overlie normal polarity, implying a minimum age of 990,000 years for the reversed sediments.

    A method is described for the production of chronic atrioventricular block in dogs, by incision of the region of the bundle of His through the open right atrium during temporary caval occlusion. Exercise tolerances, chest x-ray films, electrocardiograms, phonocardiograms, cardiac outputs, intracardiac pressures, femoral pressures, and left ventricular coronary flows were obtained preoperatively and from 1 to 10 months postoperatively. The majority of the animals developed clinical, laboratory, and pathological evidence of congestive heart failure. All animals had generalized myocardial hypertrophy

    Oceanographic Observations in Chilean Coastal Waters Between Valdivia and Concepcion

    The physical oceanography of the biologically productive coastal waters of central Chile (36 degrees to 40 degrees S) is relatively unknown. In December 1998 we made a short exploratory cruise between Valdivia (40 degrees S) and Concepcion (37.8 degrees S) taking temperature, salinity, oxygen, and current velocity profiles. Coincident sea surface temperature and color measurements were obtained by satellite. The results showed an area dominated by wind-induced coastal upwelling, river runoff, intrusion of offshore eddies, mixing, and heating. Upwelling centers were found over the shelf at three locations: inshore of Mocha Island, off Valdivia, and off Lavapie Point. At these centers, equatorial subsurface water (ESSW) intrudes into the coastal waters, sometimes affecting the surface waters. Since ESSW has characteristically low-oxygen and high-salinity values, it is easily detected. Off Valdivia, runoff imparts stratification, while farther north, solar heating and reduced mixing may facilitate stratification. In some areas, even strong winds would not destroy the stratification imparted by the advection of buoyancy that occurs during the upwelling process. Strong equatorward currents (\u3e1 m s(-1)) in the form of an upwelling jet were found off Lavapie Point. This is also the location of an intruding anticyclone. Elsewhere, currents were mainly northward but highly variable because of intrusions from offshore eddies. The sea surface temperature and ocean color images show a complex field of onshore and offshore intrusions combined with the effects of mixing on chlorophyll concentrations. The residence time of upwelled water on the shelf is estimated to be less than 1 week

    Long-term outcome prediction by clinicopathological risk classification algorithms in node-negative breast cancer—comparison between Adjuvant!, St Gallen, and a novel risk algorithm used in the prospective randomized Node-Negative-Breast Cancer-3 (NNBC-3) trial

    Background: Defining risk categories in breast cancer is of considerable clinical significance. We have developed a novel risk classification algorithm and compared its prognostic utility to the Web-based tool Adjuvant! and to the St Gallen risk classification. Patients and methods: After a median follow-up of 10 years, we retrospectively analyzed 410 consecutive node-negative breast cancer patients who had not received adjuvant systemic therapy. High risk was defined by any of the following criteria: (i) age 2 cm. All patients were also characterized using Adjuvant! and the St Gallen 2007 risk categories. We analyzed disease-free survival (DFS) and overall survival (OS). Results: The Node-Negative-Breast Cancer-3 (NNBC-3) algorithm enlarged the low-risk group to 37% as compared with Adjuvant! (17%) and St Gallen (18%), respectively. In multivariate analysis, both Adjuvant! [P = 0.027, hazard ratio (HR) 3.81, 96% confidence interval (CI) 1.16-12.47] and the NNBC-3 risk classification (P = 0.049, HR 1.95, 95% CI 1.00-3.81) significantly predicted OS, but only the NNBC-3 algorithm retained its prognostic significance in multivariate analysis for DFS (P < 0.0005).     Transduction of rat pancreatic islets with pseudotyped adeno-associated virus vectors

    <p>Abstract</p> <p>Background</p> <p>Pancreatic islet transplantation is a promising treatment for type I diabetes mellitus, but current immunosuppressive strategies do not consistently provide long-term survival of transplanted islets. We are therefore investigating the use of adeno-associated viruses (AAVs) as gene therapy vectors to transduce rat islets with immunosuppressive genes prior to transplantation into diabetic mice.</p> <p>Results</p> <p>We compared the transduction efficiency of AAV2 vectors with an AAV2 capsid (AAV2/2) to AAV2 vectors pseudotyped with AAV5 (AAV2/5), AAV8 (AAV2/8) or bovine adeno-associated virus (BAAV) capsids, or an AAV2 capsid with an insertion of the low density lipoprotein receptor ligand from apolipoprotein E (AAV2apoE), on cultured islets, in the presence of helper adenovirus infection to speed expression of a GFP transgene. Confocal microscopy and flow cytometry were used. The AAV2/5 vector was superior to AAV2/2 and AAV2/8 in rat islets. Flow cytometry indicated AAV2/5-mediated gene expression in approximately 9% of rat islet cells and almost 12% of insulin-positive cells. The AAV2/8 vector had a higher dependence on the helper virus multiplicity of infection than the AAV 2/5 vector. In addition, the BAAV and AAV2apoE vectors were superior to AAV2/2 for transducing rat islets. Rat islets (300 per mouse) transduced with an AAV2/5 vector harboring the immunosuppressive transgene, <it>tgfβ1</it>, retain the ability to correct hyperglycemia when transplanted into immune-deficient diabetic mice.</p> <p>Conclusion</p> <p>AAV2/5 vectors may therefore be useful for pre-treating donor islets prior to transplantation.</p

    Mutant PIK3CA promotes cell growth and invasion of human cancer cells

    SummaryPIK3CA is mutated in diverse human cancers, but the functional effects of these mutations have not been defined. To evaluate the consequences of PIK3CA alterations, the two most common mutations were inactivated by gene targeting in colorectal cancer (CRC) cells. Biochemical analyses of these cells showed that mutant PIK3CA selectively regulated the phosphorylation of AKT and the forkhead transcription factors FKHR and FKHRL1. PIK3CA mutations had little effect on growth under standard conditions, but reduced cellular dependence on growth factors. PIK3CA mutations resulted in attenuation of apoptosis and facilitated tumor invasion. Treatment with the PI3K inhibitor LY294002 abrogated PIK3CA signaling and preferentially inhibited growth of PIK3CA mutant cells. These data have important implications for therapy of cancers harboring PIK3CA alterations

    Ultraviolet astronomical spectrograph calibration with laser frequency combs from nanophotonic waveguides

    Astronomical precision spectroscopy underpins searches for life beyond Earth, direct observation of the expanding Universe and constraining the potential variability of physical constants across cosmological scales. Laser frequency combs can provide the critically required accurate and precise calibration to the astronomical spectrographs. For cosmological studies, extending the calibration with such astrocombs to the ultraviolet spectral range is highly desirable, however, strong material dispersion and large spectral separation from the established infrared laser oscillators have made this exceedingly challenging. Here, we demonstrate for the first time astronomical spectrograph calibrations with an astrocomb in the ultraviolet spectral range below 400 nm. This is accomplished via chip-integrated highly nonlinear photonics in periodically-poled, nano-fabricated lithium niobate waveguides in conjunction with a robust infrared electro-optic comb generator, as well as a chip-integrated microresonator comb. These results demonstrate a viable route towards astronomical precision spectroscopy in the ultraviolet and may contribute to unlocking the full potential of next generation ground- and future space-based astronomical instruments
