    Amphibians in agricultural landscapes: the habitat value of crop areas, linear plantings and remnant woodland patches

    Mitigating the negative impacts of agriculture on amphibians requires knowledge of how different land uses affect species distribution and community composition. In the case of frogs, there is currently insufficient information on their use of terrestrial habitats in cropping landscapes to inform conservation planning. We examined how four different farmland types (linear plantings, cereal crops, grazing paddocks and woody mulch) and crop harvesting influenced amphibian abundance, richness, body condition and movement. We found the abundance of frogs was significantly higher in linear plantings compared to grazing paddocks and adjacent patches of remnant woodland vegetation. However, species richness and abundance of three individual species did not vary significantly between farmland types. For the most common frog Uperoleia laevigata, body condition was higher at the edges of the woody debris treatment (coupled with higher abundance) and lower in farmland with debris and linear plantings. The body condition of Limnodynastes tasmaniensis and L. interioris was not influenced by farmland type. Frog abundance and condition was largely unaffected by crop harvesting. However, frogs were less common after harvesting at the edges of farmland and within remnant patches. Movement patterns did not suggest mass movement out of crops after harvest, where almost half of all individuals recaptured remained within the farmland. These results suggest that some generalist frog species may have an affinity for habitats within agricultural paddocks, particularly when key habitat features like plantings are present. However, we found overall frog richness was low and did not differ between remnant patches, edges and farmland which may be an indication of habitat degradation within terrestrial habitats across the landscape. Although protection of remnant native vegetation is important, conservation strategies for the protection of amphibians will be ineffective if they do not consider the variety of land uses and the relationships of different species and their microhabitats within and outside of patches.Funding was provided by the Australian Government Research Training Program, Central Tablelands Local Land Services, NSW Environmental Trust and the Lake Cowal Foundation

    Spatiotemporal effects of logging and fire on tall, wet temperate eucalypt forest birds

    Forests globally are subject to disturbances such as logging and fire that create complex temporal variation in spatial patterns of forest cover and stand age. However, investigations that quantify temporal changes in biodiversity in response to multiple forms of disturbance in space and time are relatively uncommon. Over a 10-yr period, we investigated the response of bird species to spatiotemporal changes in forest cover associated with logging and wildfire in the mountain ash (Eucalyptus regnans) forests of southeastern Australia. Specifically, we examined how bird occurrence changed with shifts in the proportion of area burned or logged in a 4.5 km radius surrounding our 88 long-term field survey sites, each measuring 1 ha in size. Overall species richness was greatest in older forest patches, but declined as the amount of fire around each site increased. At the individual species level, 31 of the 37 bird species we modeled exhibited a negative response to the amount of fire in the surrounding landscape, while one species responded positively to fire. Only nine species exhibited signs of recovery in the 6 yr of surveys following the fire. Five species were more likely to be detected as the proportion of logged forest surrounding a site increased, suggesting a possible "concentration effect" with displaced birds moving into unlogged areas following harvesting of adjacent areas. We also identified relationships between the coefficients of life history attributes and spatiotemporal changes in forest cover and stand age. Large-bodied birds and migratory species were associated with landscapes subject to large amounts of fire in 2009. There were associations between old growth stands and small-bodied bird species and species that were not insectivores. Our study shows that birds in mountain ash forests are strongly associated with old growth stands and exhibit complex, time-dependent, and species-specific responses to landscape disturbance. Despite logging and fire both being high-severity perturbations, no bird species exhibited similar responses to fire and logging in the landscape surrounding our sites.     Decline of an endangered amphibian during an extreme climatic event

    Climate change is a poorly understood, emerging threat to many amphibian species. One of the ways climate change is likely to affect amphibians is through increased recruitment failure associated with more frequent climatic extremes. To understand the risk posed by this threat, we combined 13 years of annual monitoring and multi-scaled habitat modelling at the site (n = 60), pool (n = 105) and nest (n = 170) levels to investigate the decline of the endangered northern corroboree frog (Pseudophryne pengilleyi), during the most severe drought on record in southern Australia. We documented the local extinction of 42% of P. pengilleyi breeding sites during the climatic extreme. Using logistic regression we investigated habitat variables associated with extinction sites. We found that locally extinct sites now resemble historically absent sites, with fewer pools, less water, and drying-related tree invasion. Extended periods of limited water availability at extinction sites is likely to have restricted breeding, contributing to localised extinctions. Habitat variables recorded at the pool and nest level did not significantly influence P. pengilleyi presence/absence, indicating that site level wetness had an overriding effect. We anticipate that increasing climate variability is likely to disproportionately threaten seasonal pool-breeding amphibian species, exacerbating the global amphibian biodiversity crisis. However, our work with P. pengilleyi suggests there are a range of simple habitat manipulations that could help to ameliorate the impacts

    Dysregulated autophagy in muscle precursor cells from humans with type 2 diabetes

    Autophagy is active during cellular remodeling including muscle differentiation. Muscle differentiation is dysregulated in type 2 diabetes and we therefore hypothesize that muscle precursor cells from people with type 2 diabetes (T2DM) have a dysregulation of their autophagy leading to impaired myogenesis. Muscle precursor cells were isolated from people with T2DM or healthy controls and differentiated in vitro. Autophagy marker levels were assessed by immunoblotting. Differentially expressed autophagy-related genes between healthy and T2DM groups were identified based on a previously published RNA-sequencing data-set, which we verified by RT-qPCR. siRNA was used to assess the function of differentially expressed autophagy genes. Basal autophagy increases during human muscle differentiation, while T2DM muscle cells have reduced levels of autophagy marker ATG7 and show a blunted response to starvation. Moreover, we demonstrate that the 3 non-canonical autophagy genes DRAM1, VAMP8 and TP53INP1 as differentially expressed between healthy and T2DM groups during myoblast differentiation, and that T53INP1 knock-down alters expression of both pro-and anti-apoptotic genes. In vitro differentiated T2DM muscle cells show differential expression of autophagy-related genes. These genes do not regulate myogenic transcription factors but may rather be involved in p53-associated myoblast apoptosis during early myogenesis

    ADP/ATP carrier protein was synthesized in heterologous cell-free systems programmed with Neurospora poly(A)-containing RNA and homologous cell-free systems from Neurospora. The apparent molecular weight of the product obtained in vitro was the same as that of the authentic mitochondrial protein. The primary translation product obtained in reticulocyte lysates starts with formylmethionine when formylated initiator methionyl-tRNA (fMet-tRNAfMet) was present. The product synthesized in vitro was released from the ribosomes into the postribosomal supernatant. The evidence presented indicates that the ADP/ATP carrier is synthesized as a polypeptide with the same molecular weight as the mature monomeric protein and does not carry an additional sequence

    Biosynthesis of Mitochondrial Porin and Insertion into the Outer Mitochondrial Membrane of Neuruspora crassa

    Mitochondrial porin, the major protein of the outer mitochondrial membrane is synthesized by free cytoplasmic polysomes. The apparent molecular weight of the porin synthesized in homologous or heterologous cell-free systems is the same as that of the mature porin. Transfer in vitro of mitochondrial porin from the cytosolic fraction into the outer membrane of mitochondria could be demonstrated. Before membrane insertion, mitochondrial porin is highly sensitive to added proteinase; afterwards it is strongly protected. Binding of the precursor form to mitochondria occurs at 4°C and appears to precede insertion into the membrane. Unlike transfer of many precursor proteins into or across the inner mitochondrial membrane, assembly of the porin is not dependent on an electrical potential across the inner membrane

    First detection of NH3 (1,0 - 0,0) from a low mass cloud core: On the low ammonia abundance of the rho Oph A core

    Odin has successfully observed the molecular core rho Oph A in the 572.5 GHz rotational ground state line of ammonia, NH3 (J,K = 1,0 - 0,0). The interpretation of this result makes use of complementary molecular line data obtained from the ground (C17O and CH3OH) as part of the Odin preparatory work. Comparison of these observations with theoretical model calculations of line excitation and transfer yields a quite ordinary abundance of methanol, X(CH3OH) = 3e-9. Unless NH3 is not entirely segregated from C17O and CH3OH, ammonia is found to be significantly underabundant with respect to typical dense core values, viz.     Conservation decisions under pressure: lessons from an exercise in rapid response to wildlife disease

    Novel outbreaks of emerging pathogens require rapid responses to enable successful mitigation. We simulated a 1‐day emergency meeting where experts were engaged to recommend mitigation strategies for a new outbreak of the amphibian fungal pathogen Batrachochytrium salamandrivorans. Despite the inevitable uncertainty, experts suggested and discussed several possible strategies. However, their recommendations were undermined by imperfect initial definitions of the objectives and scope of management. This problem is likely to arise in most real‐world emergency situations. The exercise thus highlighted the importance of clearly defining the context, objectives, and spatial–temporal scale of mitigation decisions. Managers are commonly under pressure to act immediately. However, an iterative process in which experts and managers cooperate to clarify objectives and uncertainties, while collecting more information and devising mitigation strategies, may be slightly more time consuming but ultimately lead to better outcomes

    Submillimeter Emission from Water in the W3 Region

    We have mapped the submillimeter emission from the 1(10)-1(01) transition of ortho-water in the W3 star-forming region. A 5'x5' map of the W3 IRS4 and W3 IRS5 region reveals strong water lines at half the positions in the map. The relative strength of the Odin lines compared to previous observations by SWAS suggests that we are seeing water emission from an extended region. Across much of the map the lines are double-peaked, with an absorption feature at -39 km/s; however, some positions in the map show a single strong line at -43 km/s. We interpret the double-peaked lines as arising from optically thick, self-absorbed water emission near the W3 IRS5, while the narrower blue-shifted lines originate in emission near W3 IRS4. In this model, the unusual appearance of the spectral lines across the map results from a coincidental agreement in velocity between the emission near W3 IRS4 and the blue peak of the more complex lines near W3 IRS5. The strength of the water lines near W3 IRS4 suggests we may be seeing water emission enhanced in a photon-dominated region.Comment: Accepted to A&A Letters as part of the special Odin issue; 4 page
