Development of a Disposable Gold Electrodes-Based Sensor for Electrochemical Measurements of cDNA Hybridization
AbstractThis work deals with the development of a disposable electrochemical biosensor for the speciïŹc detection of short DNA sequences. The sensor is an amperometric transducer with three planar electrodes, comprising a working, a counter and a pseudo-reference electrode, all made of a gold layer over a polycarbonate substrate. For the development of the genosensor, the working electrode was modiïŹed using thiol-tethered 33-mer DNA probe by chemisorptions, in a concentration range from 0.1 ÎŒM to 5 ÎŒM. Immobilization of ssDNA on gold surface was monitored with electrochemical impedance spectroscopy (EIS) and differential pulse voltammetry (DPV) in Fe(CN)64â/13â and Ruthenium(II)/(III) solutions. The time dependence of ssDNA probe immobilization was also studied. The hybridization detection is then compared with EIS and DPV measurements
Event-based, 6-DOF Camera Tracking from Photometric Depth Maps
Event cameras are bio-inspired vision sensors that output pixel-level brightness changes instead of standard intensity frames. These cameras do not suffer from motion blur and have a very high dynamic range, which enables them to provide reliable visual information during high-speed motions or in scenes characterized by high dynamic range. These features, along with a very low power consumption, make event cameras an ideal complement to standard cameras for VR/AR and video game applications. With these applications in mind, this paper tackles the problem of accurate, low-latency tracking of an event camera from an existing photometric depth map (i.e., intensity plus depth information) built via classic dense reconstruction pipelines. Our approach tracks the 6-DOF pose of the event camera upon the arrival of each event, thus virtually eliminating latency. We successfully evaluate the method in both indoor and outdoor scenes and show thatâbecause of the technological advantages of the event cameraâour pipeline works in scenes characterized by high-speed motion, which are still unaccessible to standard cameras
Sensitive methods for estimating the anchoring strength of nematic liquid crystals on Langmuir-Blodgett monolayers of fatty acids

N-(p-methoxybenzylidene)-p-butylaniline (MBBA) on Langmuir-Blodgett monolayers
of fatty acids (COOHCH) was studied as a function of the length
of the fatty acid alkyl chains, (). The monolayers were
deposited onto ITO-coated glass plates which were used to assemble sandwich
cells of various thickness that were filled with MBBA in the nematic phase. The
mechanism of relaxation from the flow-induced quasi-planar to the
surface-induced homeotropic alignment was studied for the four decreases
linearly with increasing the length of the alkyl chains which suggests that
the Langmuir-Blodgett film plays a role in the phenomenon. This fact was
confirmed by a sensitive estimation of the anchoring strength of MBBA on the
fatty acid monolayers after anchoring breaking which takes place at the
transition between two electric-field--induced turbulent states, denoted as
DSM1 and DSM2. It was found that the threshold electric field for the anchoring
breaking, which can be considered as a measure of the anchoring strength, also
decreases linearly as increases. Both methods thus possess a high
sensitivity in resolving small differences in anchoring strength. In cells
coated with mixed Langmuir-Blodgett monolayers of two fatty acids ( and
) a maximum of the relaxation speed was observed when the two acids were
present in equal amount. This observation homeotropic cells by changing the

Rethinking carbohydrate intake and time in range in children and adolescents with type 1 diabetes
none15The aim of this study was to evaluate the association between macronutrient intake and time in range (TIR) of 70â180 mg/dL in children and adolescents with type 1 diabetes (T1D). A multi-center study recruited patients with T1D using continuous glucose monitoring (CGM) between January 2019 and January 2020 from centers across Italy. Diet intake was recorded using three-day weighed food diaries. Nutrients were evaluated as percentages of total intake. TIR was considered at target if the percentage of readings was higher than 70%. Clinical and nutritional factors associated with TIR at target were analyzed using multiple correspondence analysis and multiple logistic regression. Data from 197 participants (53% male, median age 11.6 years, median HbA1c 55.2 mmol/mol, median TIR 60%) were analyzed. Macronutrient intake was 45.9% carbohydrates, 16.9% protein, 37.3% fat, and 13.1 g/day fiber (median values). TIR > 70% was observed in 28% of participants; their diet contained more protein (17.6%, p = 0.015) and fiber (14.4 g/day, p = 0.031) than those not at target. The probability of having a TIR > 70% was significantly higher with 40â44% consumption of carbohydrates compared with 45â50% consumption of carbohydrates and with the use of a carbohydrate counting system. Based on these results, a five percent reduction in the percentage of carbohydrate intake can help children and adolescents with T1D achieve the goal of a TIR > 70%. Both a lower and higher percentage of carbohydrate intake appears to reduce the probability of reaching the target TIR > 70%. Dual-regulated lentiviral vector for gene therapy of X-linked chronic granulomatosis
Regulated transgene expression may improve the safety and efficacy of hematopoietic stem cell (HSC) gene therapy. Clinical trials for X-linked chronic granulomatous disease (X-CGD) employing gammaretroviral vectors were limited by insertional oncogenesis or lack of persistent engraftment. Our novel strategy, based on regulated lentiviral vectors (LV), targets gp91(phox) expression to the differentiated myeloid compartment while sparing HSC, to reduce the risk of genotoxicity and potential perturbation of reactive oxygen species levels. Targeting was obtained by a myeloid-specific promoter (MSP) and posttranscriptional, microRNA-mediated regulation. We optimized both components in human bone marrow (BM) HSC and their differentiated progeny in vitro and in a xenotransplantation model, and generated therapeutic gp91(phox) expressing LVs for CGD gene therapy. All vectors restored gp91(phox) expression and function in human X-CGD myeloid cell lines, primary monocytes, and differentiated myeloid cells. While unregulated LVs ectopically expressed gp91(phox) in CD34(+) cells, transcriptionally and posttranscriptionally regulated LVs substantially reduced this off-target expression. X-CGD mice transplanted with transduced HSC restored gp91(phox) expression, and MSP-driven vectors maintained regulation during BM development. Combining transcriptional (SP146.gp91-driven) and posttranscriptional (miR-126-restricted) targeting, we achieved high levels of myeloid-specific transgene expression, entirely sparing the CD34(+) HSC compartment. This dual-targeted LV construct represents a promising candidate for further clinical development
Insulin pump failures in Italian children with Type 1 diabetes: retrospective 1-year cohort study
AimsInsulin pump failure and/or malfunction requiring replacement have not been thoroughly investigated. This study evaluated pump replacement in children and adolescents with Type 1 diabetes using insulin pump therapy.MethodsData were collected for all participants younger than 19 years, starting insulin pump therapy before 31 December 2013. For each child, age, disease duration, date of insulin pump therapy initiation, insulin pump model, failure/malfunction/replacement yes/no and reason were considered for the year 2013.ResultsData were returned by 40 of 43 paediatric centres belonging to the Diabetes Study Group of the Italian Society of Paediatric Endocrinology and Diabetology. In total, 1574 of 11 311 (13.9%) children and adolescents with Type 1 diabetes were using an insulin pump: 29.2% Animas VIBE, 9.4% Medtronic MiniMed 715/515, 34.3% Medtronic MiniMed VEO, 24.3% Accu-Check Spirit Combo and 2.8% other models. In 2013, 0.165 insulin pump replacements per patient-year (11.8% due to pump failure/malfunction and 4.7% due to accidental damage) were recorded. Animas VIBE (22.1%) and Medtronic MiniMed VEO (17.7%) were the most replaced.ConclusionsIn a large cohort of Italian children and adolescents with Type 1 diabetes, insulin pump failure/malfunction and consequent replacement are aligned with rates previously reported and higher in more sophisticated pump models
Land-use and land-cover mapping of the Brazilian cerrado based mainly on Landsat-8 satellite images.
The Brazilian Cerrado is one of the worldÂŽs biodiversity hotspot and hosts some of the most intensive agricultural activities for food production in the world. The objective of this study was to produce a land-use and land-cover (LULC) map of the Cerrado based on Landsat-8 Operational Land Imager (OLI) images. A set of 121 scenes from 2013 was processed using the image segmentation technique. The segments were exported in the shapefile format and interpreted visually in a geographical information system software using RGB/564 color composites. The following LULC classes were considered: annual croplands, perennial croplands, cultivated pasturelands, reforestation, mosaic of occupation, urban areas, mining areas, bare soil, forestlands, non-forestlands, water bodies, and non-identified (clouds and burned areas). The overall accuracy was estimated by an independent scientist with large experience in CerradoÂŽs image interpretation. The results showed that 43.4% of the study area (88.5 million hectares) were already converted into agricultural, urban and mining areas, 54.6% (111 million hectares) were still natural areas, and 1.9% (3.9 million hectares) was classified as non-identified. Cultivated pasturelands were the most representative land-use type (29.5%), followed by annual croplands (8.5%) and perennial croplands (3.1%).
