    Policy Responsiveness and Political Accountability in City Politics

    Peculiar patterns have emerged in municipal politics in Canada. Unlike at federal and provincial levels of government, party politics is weak or absent in cities. But looking at the entire history of municipal elections of three Western Canadian cities — Calgary, Edmonton and Vancouver — also finds that, again unlike politicians at higher levels, municipal representatives are increasingly more likely to win repeated re-elections, facing a much lower threat from any competitive challenge. In addition, the careers of municipal politicians are growing steadily longer, leading to relatively stable, almost static, city governments that tend to see change most commonly when councillors choose to step down, rather than being forced out.Such patterns, of course, run counter to the general presumption in lively democracies, including this one, that the responsiveness of government leaders (that is, following the wishes of their constituents) and their accountability for the actions they take are best served by frequent turnovers in government: The need to throw politicians out every now and again to let new ones try and do things better. Whether the remarkable levels of stability and incumbency on city councils actually do serve the best interests of voters is unclear. More clear is that the lack of a party system at the city level seems to have contributed to this peculiar dynamic, and that city politicians have an interest in keeping it that way.While party affiliations provide a candidate some benefits in the form of campaign co-ordination, they also provide voters with increased clarity about what each candidate stands for policy-wise. That might be helpful to voters, but city politicians might find it more useful to blur their positions, leaving voters uncertain of exactly how to define a specific councillor’s stand, overall. The amount of information required to root through a councillor’s voting record, and the relatively light media coverage of daily council business, leaves most voters inclined to rely on something other than political signals when they decide who to support. Inevitably, a candidate’s personal character and length of experience take on a larger priority in the voting decision. So, the longer a councillor serves, the longer he or she might be likely to keep serving.The natural outcome of this phenomenon is that city councils are less likely to become polarized as councillors have an incentive to seek consensus by limiting obvious policy distinctions between themselves and their fellow representatives, contributing to a dynamic where city council works more like a corporate body and less like a partisan legislature. As long as citizens remain largely unperturbed by the overall actions of their city council, they might judge the risk of replacing them at election time as unnecessarily high compared to sticking with the status quo.This behaviour challenges much of the prevailing theory about how political processes improve political responsiveness and accountability. This is occurring at a time when cities are emerging as important and increasingly powerful nodes in the modern global economy. That the very nature of how they are governed is diverging so markedly from the norm that Canadians have come to expect from other levels of government is not something to be considered lightly

    The Alberta Tory dynasty begun by Peter Lougheed is now 40 years old. With only four leaders across four decades, the party has managed to maintain its hold on the political imagination of Albertans. It has weathered a number of storms, from minor party assaults during the tumultuous 1980s to the Liberal threat of 1993 and the stresses associated with the global financial crisis. Now it confronts a new challenge in the form of the Wildrose Alliance led by Danielle Smith.Just as the Tories stole the centre ground from beneath Social Credit in the 1970s, the Wildrose leadership team hopes to take what was a fringe right wing party and turn it into a broad coalition capable of appealing to a large number of Albertans. What challenges do they face in repositioning the party? And how will the Tories protect their home turf?In brief, the Wildrose Alliance must modify its policies and present them in such a manner as to be able to plausibly claim that it now reflects the core values of Albertans better than the current government.For its part, the government must select a new leader capable of successfully painting Wildrose as outsiders who cannot be trusted to cleave to the values that Albertans hold dear. What are these values? Strong support for individualism, a populist view of government – including wariness of the federal government – combined with a deep commitment to a role for government in providing core programs in areas such as health care, the environment, and social welfare

    Party politics in Alberta can seem dull. Election after election the Conservative party is returned to power with a comfortable majority and campaigns are marked by little in the way of suspense (see Bell et al, 2007 and Stewart and Archer, 2000). The last time government changed hands in Alberta was 1971 when the Peter Lougheed led Progressive Conservatives eked out a narrow win over the Social Credit dynasty. Lougheed and the Conservatives positioned them- selves as safe, conservative change. This was also the case when the Progressive Conservatives faced their most serious challenge to date, in 1993. The Liberals, led by former Edmonton mayor Laurence Decore, launched a fiscal attack on the Conservatives, presenting themselves as the safe, conservative alternative (Stewart, 1995). The Ralph Klein Conservatives beat back that challenge and the party has easily carried each subsequent election

    SWCam: the short wavelength camera for the CCAT Observatory

    We describe the Short Wavelength Camera (SWCam) for the CCAT observatory including the primary science drivers, the coupling of the science drivers to the instrument requirements, the resulting implementation of the design, and its performance expectations at first light. CCAT is a 25 m submillimeter telescope planned to operate at 5600 meters, near the summit of Cerro Chajnantor in the Atacama Desert in northern Chile. CCAT is designed to give a total wave front error of 12.5 μm rms, so that combined with its high and exceptionally dry site, the facility will provide unsurpassed point source sensitivity deep into the short submillimeter bands to wavelengths as short as the 200 μm telluric window. The SWCam system consists of 7 sub-cameras that address 4 different telluric windows: 4 subcameras at 350 μm, 1 at 450 μm, 1 at 850 μm, and 1 at 2 mm wavelength. Each sub-camera has a 6’ diameter field of view, so that the total instantaneous field of view for SWCam is equivalent to a 16’ diameter circle. Each focal plane is populated with near unit filling factor arrays of Lumped Element Kinetic Inductance Detectors (LEKIDs) with pixels scaled to subtend an solid angle of (λ/D)2 on the sky. The total pixel count is 57,160. We expect background limited performance at each wavelength, and to be able to map < 35(°)2 of sky to 5 σ on the confusion noise at each wavelength per year with this first light instrument. Our primary science goal is to resolve the Cosmic Far-IR Background (CIRB) in our four colors so that we may explore the star and galaxy formation history of the Universe extending to within 500 million years of the Big Bang. CCAT's large and high-accuracy aperture, its fast slewing speed, use of instruments with large format arrays, and being located at a superb site enables mapping speeds of up to three orders of magnitude larger than contemporary or near future facilities and makes it uniquely sensitive, especially in the short submm bands

    Genomic, Proteomic and Physiological Characterization of a T5-like Bacteriophage for Control of Shiga Toxin-Producing Escherichia coli O157:H7

    Despite multiple control measures, Escherichia coli O157:H7 (STEC O157:H7) continues to be responsible for many food borne outbreaks in North America and elsewhere. Bacteriophage therapy may prove useful for controlling this pathogen in the host, their environment and food. Bacteriophage vB_EcoS_AKFV33 (AKFV33), a T5-like phage of Siphoviridae lysed common phage types of STEC O157:H7 and not non-O157 E. coli. Moreover, STEC O157:H7 isolated from the same feedlot pen from which the phage was obtained, were highly susceptible to AKFV33. Adsorption rate constant and burst size were estimated to be 9.31×10−9 ml/min and 350 PFU/infected cell, respectively. The genome of AKVF33 was 108,853 bp (38.95% G+C), containing 160 open reading frames (ORFs), 22 tRNA genes and 32 strong promoters recognized by host RNA polymerase. Of 12 ORFs without homologues to T5-like phages, 7 predicted novel proteins while others exhibited low identity (<60%) to proteins in the National Centre for Biotechnology Information database. AKVF33 also lacked the L-shaped tail fiber protein typical of T5, but was predicted to have tail fibers comprised of 2 novel proteins with low identity (37–41%) to tail fibers of E. coli phage phiEco32 of Podoviridae, a putative side tail fiber protein of a prophage from E. coli IAI39 and a conserved domain protein of E. coli MS196-1. The receptor-binding tail protein (pb5) shared an overall identify of 29–72% to that of other T5-like phages, with no region coding for more than 6 amino acids in common. Proteomic analysis identified 4 structural proteins corresponding to the capsid, major tail, tail fiber and pore-forming tail tip (pb2). The genome of AKFV33 lacked regions coding for known virulence factors, integration-related proteins or antibiotic resistance determinants. Phage AKFV33 is a unique, highly lytic STEC O157:H7-specific T5-like phage that may have considerable potential as a pre- and post-harvest biocontrol agent

    Cyclophosphamide Chemotherapy Sensitizes Tumor Cells to TRAIL-Dependent CD8 T Cell-Mediated Immune Attack Resulting in Suppression of Tumor Growth

    Background: Anti-cancer chemotherapy can be simultaneously lymphodepleting and immunostimulatory. Pre-clinical models clearly demonstrate that chemotherapy can synergize with immunotherapy, raising the question how the immune system can be mobilized to generate anti-tumor immune responses in the context of chemotherapy. Methods and Findings: We used a mouse model of malignant mesothelioma, AB1-HA, to investigate T cell-dependent tumor resolution after chemotherapy. Established AB1-HA tumors were cured by a single dose of cyclophosphamide in a CD8 T cell- and NK cell-dependent manner. This treatment was associated with an IFN-α/β response and a profound negative impact on the anti-tumor and total CD8 T cell responses. Despite this negative effect, CD8 T cells were essential for curative responses. The important effector molecules used by the anti-tumor immune response included IFN-γ and TRAIL. The importance of TRAIL was supported by experiments in nude mice where the lack of functional T cells could be compensated by agonistic anti-TRAIL-receptor (DR5) antibodies. Conclusion: The data support a model in which chemotherapy sensitizes tumor cells for T cell-, and possibly NK cell-, mediated apoptosis. A key role of tumor cell sensitization to immune attack is supported by the role of TRAIL in tumor resolution and explains the paradox of successful CD8 T cell-dependent anti-tumor responses in the absence of CD8 T cell expansion

    Abdominal aortic aneurysm is associated with a variant in low-density lipoprotein receptor-related protein 1

    Abdominal aortic aneurysm (AAA) is a common cause of morbidity and mortality and has a significant heritability. We carried out a genome-wide association discovery study of 1866 patients with AAA and 5435 controls and replication of promising signals (lead SNP with a p value &lt; 1 × 10-5) in 2871 additional cases and 32,687 controls and performed further follow-up in 1491 AAA and 11,060 controls. In the discovery study, nine loci demonstrated association with AAA (p &lt; 1 × 10-5). In the replication sample, the lead SNP at one of these loci, rs1466535, located within intron 1 of low-density-lipoprotein receptor-related protein 1 (LRP1) demonstrated significant association (p = 0.0042). We confirmed the association of rs1466535 and AAA in our follow-up study (p = 0.035). In a combined analysis (6228 AAA and 49182 controls), rs1466535 had a consistent effect size and direction in all sample sets (combined p = 4.52 × 10-10, odds ratio 1.15 [1.10-1.21]). No associations were seen for either rs1466535 or the 12q13.3 locus in independent association studies of coronary artery disease, blood pressure, diabetes, or hyperlipidaemia, suggesting that this locus is specific to AAA. Gene-expression studies demonstrated a trend toward increased LRP1 expression for the rs1466535 CC genotype in arterial tissues; there was a significant (p = 0.029) 1.19-fold (1.04-1.36) increase in LRP1 expression in CC homozygotes compared to TT homozygotes in aortic adventitia. Functional studies demonstrated that rs1466535 might alter a SREBP-1 binding site and influence enhancer activity at the locus. In conclusion, this study has identified a biologically plausible genetic variant associated specifically with AAA, and we suggest that this variant has a possible functional role in LRP1 expression

    Variations in T Cell Transcription Factor Sequence and Expression Associated with Resistance to the Sheep Nematode Teladorsagia circumcincta

    This study used selected lambs that varied in their resistance to the gastrointestinal parasite Teladorsagia circumcincta. Infection over 12 weeks identified susceptible (high adult worm count, AWC; high fecal egg count, FEC; low body weight, BW; low IgA) and resistant sheep (no/low AWC and FEC, high BW and high IgA). Resistance is mediated largely by a Th2 response and IgA and IgE antibodies, and is a heritable characteristic. The polarization of T cells and the development of appropriate immune responses is controlled by the master regulators, T-bet (TBX21), GATA-3 (GATA3), RORγt (RORC2) and RORα (RORA); and several inflammatory diseases of humans and mice are associated with allelic or transcript variants of these transcription factors. This study tested the hypothesis that resistance of sheep to T. circumcincta is associated with variations in the structure, sequence or expression levels of individual master regulator transcripts. We have identified and sequenced one variant of sheep TBX21, two variants of GATA3 and RORC2 and five variants of RORA from lymph node mRNA. Relative RT-qPCR analysis showed that TBX21, GATA3 and RORC2 were not significantly differentially-expressed between the nine most resistant (AWC, 0; FEC, 0) and the nine most susceptible sheep (AWC, mean 6078; FEC, mean 350). Absolute RT-qPCR on 29 all 45 animals identified RORAv5 as being significantly differentially-expressed (p = 0.038) 30 between resistant, intermediate and susceptible groups; RORAv2 was not differentially- 31 expressed (p = 0.77). Spearman’s rank analysis showed that RORAv5 transcript copy number 32 was significantly negatively correlated with parameters of susceptibility, AWC and FEC; and 33 was positively correlated with BW. RORAv2 was not correlated with AWC, FEC or BW but 34 was significantly negatively correlated with IgA antibody levels [corrected]. This study identifies the full length RORA variant (RORAv5) as important in controlling the protective immune response to T. circumcincta infection in sheep
