    Structure of the regulatory domain of the LysR family regulator NMB2055 (MetR-like protein) from Neisseria meningitidis

    Copyright @ 2012 International Union of CrystallographyThe crystal structure of the regulatory domain of NMB2055, a putative MetR regulator from Neisseria meningitidis, is reported at 2.5 Å resolution. The structure revealed that there is a disulfide bond inside the predicted effector-binding pocket of the regulatory domain. Mutation of the cysteines (Cys103 and Cys106) that form the disulfide bond to serines resulted in significant changes to the structure of the effector pocket. Taken together with the high degree of conservation of these cysteine residues within MetR-related transcription factors, it is suggested that the Cys103 and Cys106 residues play an important role in the function of MetR regulators.This study is funded by the Medical Research Council, with additional finance from the Biotechnology and Biological Science Research Council

    Evaluation of Sova's Lincolnshire Offender Mentoring scheme

    In January 2014 a team of researchers from the University of Lincoln undertook an evaluation of Sova’s Lincolnshire Offender Mentoring Programme (LOM). LOM is run in partnership with Lincolnshire Probation Trust. LOM works to address the multiple barriers faced by disadvantaged offenders. The project offers community mentoring and a range of intervention activities that aim to enable its participants to realise their full potential and reduce re-offending. The researchers interviewed 12 participants, which included 6 LPT staff, 2 Sova staff, 2 mentors and 2 offenders. Despite LOM being in its infancy, the research found that Sova and LPT had developed an effective inter-agency partnership with excellent levels of communication. The research also found the LOM is valued by both LPT staff and offenders and that it helped improve the service offered to offenders. Whilst the sample size of the research is relatively small there was evidence to suggest that LPT staff and offenders thought LOM had a positive impact on offenders’ behaviour. The research concludes that LOM should continue to be supported and further extended to other suitable offenders who would clearly benefit from the programme

    Evaluation of the angiotensin II receptor blocker azilsartan medoxomil in African-American patients with hypertension

    The efficacy and safety of azilsartan medoxomil (AZL-M) were evaluated in African-American patients with hypertension in a 6-week, double-blind, randomized, placebo-controlled trial, for which the primary end point was change from baseline in 24-hour mean systolic blood pressure (BP). There were 413 patients, with a mean age of 52years, 57% women, and baseline 24-hour BP of 146/91mmHg. Treatment differences in 24-hour systolic BP between AZL-M 40mg and placebo (-5.0mmHg; 95% confidence interval, -8.0 to -2.0) and AZL-M 80mg and placebo (-7.8mmHg; 95% confidence interval, -10.7 to -4.9) were significant (P.001 vs placebo for both comparisons). Changes in the clinic BPs were similar to the ambulatory BP results. Incidence rates of adverse events were comparable among the treatment groups, including those of a serious nature. In African-American patients with hypertension, AZL-M significantly reduced ambulatory and clinic BPs in a dose-dependent manner and was well tolerated

    The utility of MAS5 expression summary and detection call algorithms

    <p>Abstract</p> <p>Background</p> <p>Used alone, the MAS5.0 algorithm for generating expression summaries has been criticized for high False Positive rates resulting from exaggerated variance at low intensities.</p> <p>Results</p> <p>Here we show, with replicated cell line data, that, when used alongside detection calls, MAS5 can be both selective and sensitive. A set of differentially expressed transcripts were identified that were found to be changing by MAS5, but unchanging by RMA and GCRMA. Subsequent analysis by real time PCR confirmed these changes. In addition, with the Latin square datasets often used to assess expression summary algorithms, filtered MAS5.0 was found to have performance approaching that of its peers.</p> <p>Conclusion</p> <p>When used alongside detection calls, MAS5 is a sensitive and selective algorithm for identifying differentially expressed genes.</p
