    Antidepressant‐Like Effect of Ferulic Acid via Promotion of Energy Metabolism Activity

    Scope Ferulic acid (FA), a natural phenolic phytochemical abundantly present in whole grains, herbs, and dried fruits, exhibits anti-inflammatory, antioxidant, and neuroprotective effects. In the present study, the antidepressant-like effects of FA in male ICR mice using tail suspension test (TST) are investigated and its molecular mechanisms are explored. Methods and Results Oral administration of FA at a dose of 5 mg kg(-1) for 7 days significantly reduces immobility of mice compared to vehicle-administered control group. Microarray and real-time PCR analyses reveal that FA upregulates the expression of several genes associated with cell survival and proliferation, energy metabolism, and dopamine synthesis in mice limbic system of brain. Interestingly, it is found that FA, unlike antidepressant drug bupropion, strongly promotes energy metabolism. Additionally, FA increases catecholamine (dopamine and noradrenaline), brain-derived neurotrophic factor, and ATP levels, and decreases glycogen levels in the limbic system of the mice brain. Conclusion The research provides the first evidence that FA enhances energy production, which can be the underlying mechanism of the antidepressant-like effects of FA observed in this study

    Conceptual design of a wide-field near UV transient survey in a 6U CubeSat

    A conceptual design of a wide-field near UV transient survey in a 6U CubeSat is presented. Ultraviolet is one of the frontier in the transient astronomy. To open up the discovery region, we are developing a 6U CubeSat for transient exploration. The possible targets will be supernova shock-breakouts, tidal disruption events, and the blue emission from NS-NS mergers in very early phase. If we only focused on nearby/bright sources, the required detection limit is around 20 mag (AB). To avoid the background and optical light, we chose a waveband of 230-280 nm. As an imaging detector, we employ a delta-doped back-illuminated CMOS. In addition to delta doping, the multi-layer coating directly deposited on the detector enables both a high in-band UV QE and the ultra-low optical rejection ratio. Taking into account these specifications, even an 8 cm telescope can achieve the detection limit of 20 magAB. The expected FoV is larger than 60 deg^2

    女子大学を卒業した若手教員の意識と研修課題に関する調査研究―新卒3年目までの小学校教員を対象に―

     本研究では,女子大学を卒業した新卒3 年目までの正規採用の小学校教員を対象に,若手教員の意識や研修課題及び卒業生支援に対する要望を調べ,それらに対する方策について検討を行った。調査の結果,卒業生の若手教員の多くが教職にやりがいを感じながらも,採用前の予想を上回る学校現場の厳しい困難な現実に直面し,それぞれに研修課題を抱いている実状が明らかになった。例えば,研修課題においては,日々,適切な指導が求められる「教科等の指導」,「生徒指導」や新しい教育課題である「特別の教科 道徳」などのニーズが高いことなどが明らかになった。こうした結果を踏まえ,本研究報告は,在学中の早い段階から教職の魅力を伝えたり,実践的指導力の向上を意識した授業内容を設定したりすることなどの教職課程の改善や,卒業後もニーズに応じた研修を提供するなどの卒業生支援の拡充についての方策を提言している

    A Patient with Type 3 Autoimmune Polyglandular Syndrome who Developed Systemic Lupus Erythematosus 8 years after the Diagnosis of Autoimmune Hepatitis

    Eight years prior to her present admission, a 61-year-old Japanese woman was diagnosed with autoimmune hepatitis, slowly progressive insulin-dependent diabetes mellitus, and chronic thyroiditis; she had been treated with oral prednisolone (PSL). After she suddenly discontinued PSL, she newly developed systemic lupus erythematosus. A combination therapy of oral PSL and intravenous cyclophosphamide resulted in remission. She was finally diagnosed with autoimmune polyglandular syndrome (APS) type 3 (3A ,3B, 3D), complicated with four different autoimmune diseases. Since patients with type 3 APS may present many manifestations over a long period of time, they should be carefully monitored

    Spinocerebellar ataxia type 6 (SCA6) is an autosomal dominant neurodegenerative disease caused by a small polyglutamine (polyQ) expansion (control: 4–20Q; SCA6: 20–33Q) in the carboxyl(C)-terminal cytoplasmic domain of the α1A voltage-dependent calcium channel (Cav2.1). Although a 75–85-kDa Cav2.1 C-terminal fragment (CTF) is toxic in cultured cells, its existence in human brains and its role in SCA6 pathogenesis remains unknown. Here, we investigated whether the small polyQ expansion alters the expression pattern and intracellular distribution of Cav2.1 in human SCA6 brains. New antibodies against the Cav2.1 C-terminus were used in immunoblotting and immunohistochemistry. In the cerebella of six control individuals, the CTF was detected in sucrose- and SDS-soluble cytosolic fractions; in the cerebella of two SCA6 patients, it was additionally detected in SDS-insoluble cytosolic and sucrose-soluble nuclear fractions. In contrast, however, the CTF was not detected either in the nuclear fraction or in the SDS-insoluble cytosolic fraction of SCA6 extracerebellar tissues, indicating that the CTF being insoluble in the cytoplasm or mislocalized to the nucleus only in the SCA6 cerebellum. Immunohistochemistry revealed abundant aggregates in cell bodies and dendrites of SCA6 Purkinje cells (seven patients) but not in controls (n = 6). Recombinant CTF with a small polyQ expansion (rCTF-Q28) aggregated in cultured PC12 cells, but neither rCTF-Q13 (normal-length polyQ) nor full-length Cav2.1 with Q28 did. We conclude that SCA6 pathogenesis may be associated with the CTF, normally found in the cytoplasm, being aggregated in the cytoplasm and additionally distributed in the nucleus

In particular, studies of gene expression dynamics and regulation landscape in COVID-19 infected individuals are limited. Here, we report on a thorough analysis of whole blood RNA-seq data from 465 genotyped samples from the Japan COVID-19 Task Force, including 359 severe and 106 non-severe COVID-19 cases. We discover 1169 putative causal expression quantitative trait loci (eQTLs) including 34 possible colocalizations with biobank fine-mapping results of hematopoietic traits in a Japanese population, 1549 putative causal splice QTLs (sQTLs; e.g. two independent sQTLs at TOR1AIP1), as well as biologically interpretable trans-eQTL examples (e.g., REST and STING1), all fine-mapped at single variant resolution. We perform differential gene expression analysis to elucidate 198 genes with increased expression in severe COVID-19 cases and enriched for innate immune-related functions. Finally, we evaluate the limited but non-zero effect of COVID-19 phenotype on eQTL discovery, and highlight the presence of COVID-19 severity-interaction eQTLs (ieQTLs; e.g., CLEC4C and MYBL2). Our study provides a comprehensive catalog of whole blood regulatory variants in Japanese, as well as a reference for transcriptional landscapes in response to COVID-19 infection
