A new simple, sensitive, precise, economic and “green” spectrofluorimetric method for the determination of quinine both as a bulk drug and in tablet formulations was developed and validated using water as solvent. At a predetermined excitation wavelength (330 nm) and emission wavelength (380 nm), it was proved linear in the concentration range of 50-500 ng/mL, exhibited good correlation coefficient (R2= 0.999) and excellent mean recovery (97.5-103%). The results of the recovery studies showed that the method was not affected by the presence of common excipients. The method was applied for the analysis of the drug in the pure, tablet and injectable forms. The method was validated for precision, accuracy and recovery studies. Limit of Detection and Limit of Quantification for quinine were found to be 16.6 ng/mL and 19.8 ng/mL respectively. The method has been successfully applied for the analysis of marketed formulations available in Senegal. © 2013 International Formulae Group. Background African populations are considered to be particularly vulnerable to fever illnesses, including malaria, and acute respiratory disease, owing to limited resources and overcrowding. However, the overall burden of influenza in this context is poorly defined and incidence data for African countries are scarce. We therefore studied the fever syndrome incidence and more specifically influenza incidence in a cohort of inhabitants of Dielmo and Ndiop in Sokone district, Senegal. Methods Daily febrile-illness data were prospectively obtained from January 2012 to December 2013 from the cohort of the villages of Dielmo and Ndiop, initially dedicated to the study of malaria. Nasopharyngeal swabs were collected from, and malaria diagnosis tests (thick blood smears) carried out on, every febrile individual during clinical visits; reverse transcriptase- polymerase chain reaction was used to identify influenza viruses in the samples. Binomial negative regression analysis was used to study the relationship between the monthly incidence rate and various covariates. Results In Dielmo and Ndiop, the incidence ofmalaria has decreased, but fever syndromes remain frequent. Among the 1036 inhabitants included in the cohort, a total of 1,129 episodes of fever were reported. Influenza was present all year round with peaks in October-December 2012 and August 2013. The fever, ILI and influenza incidence density rates differed significantly between age groups. At both sites, the adjusted incidence relative risks for fever syndromes and ILI were significantly higher in the [6-24 months) than other age groups: 7.3 (95% CI: [5.7-9.3]) and 16.1 (95% CI: [11.1-23.3]) respectively. The adjusted incidence relative risk for influenza was significantly higher for the [0-6 months) than other age groups: 9.9 (95% CI: [2.9-33.6]). At both sites, incidence density rates were lowest among adults > = 50 years. Conclusions In this rural setting in Senegal, influenza was most frequent among the youngest children. Preventive strategies targeting this population should be implemented

Introduction: Halitosis is a common complain among dental patients, chronic among 50% of the adult population, and of oral origin in 90% of the cases. The objective of this study was to evaluate the prevalence and related factors of oral halitosis among a senegalese population. Material and method: This was a transversal descriptive study of 396 Senegalese adults using a 24-point self-assessment questionnaire on halitosis. All subjects were examined to estimate the organoleptic means and to measure the Sulfur Volatile Compounds (SVC), using a halimeter. Halitosis was diagnosed when the level of sulfur volatile Compound was ≥125 ppb and the organoleptic score ≥ 2. Data were statistically analyzed using the chi2 test. Results: The global prevalence of halitosis was 32.3 %. The difference was not statistically significant between male and female. Among the 396 patients suffering from bad breath, 128 had a permanent halitosis and 52 had a physiological halitosis. The means of the organoleptic scores were 3.12 and that of the (SVC) were 273.9 ppb. Conclusion: The results of the study showed a relation between the organoleptic tests and the level of sulfur volatile compounds (SVC). Acetabular fractures: epidemiological, lesional, therapeutic and evolutionary aspects

    Get PDF
Background: Fractures of the acetabulum are serious because they affect a deep, load-bearing joint that is difficult to repair surgically at first. They occur in a context of polytrauma. The aim of our work is to study the epidemiological, lesional, therapeutic and evolutionary aspects of acetabulum fractures. Methods: This is a 5-year retrospective study from January 2012 to December 2016 including 45 patients treated for acetabulum fracture. Results: 45 patients including 38 men and 7 women with a mean age of 36 years (extremes 18 and 74). The circumstances of occurrence were dominated by road traffic accidents with 42 cases (93.3%).According to the Judet-Letournel classification, we observed 28 elementary fractures (16 posterior wall fractures, 11 posterior column fractures, 1 anterior column fracture) and 17 complex fractures (7 posterior column and posterior wall fractures, 6 transverse and posterior wall fractures, 3 posterior and anterior column fractures, 1 anterior column and posterior hemi-transverse fracture). The association of a dislocation with an acetabulum fracture was found in 32 cases. Associated lesions (fractures) were observed in 23 patients. One case of associated TCE was noted. Treatment was orthopedic in 18 patients, surgical in 18 patients and functional in 9 patients. According to the Matta criteria we obtained after treatment 46.7% good reduction, 51.1% satisfactory and 2.2% unsatisfactory reduction. According to the Postel-Merle D'Aubigné rating, 16 patients obtained excellent results with overall 93.3% satisfactory results. The complications found were coxarthrosis (15 cases), para-articular ossification (2 cases) and vicious callus (1 case). Conclusions: The treatment of acetabulum fractures depends on the type of fracture, the age and activities of the patients. Orthopedic, surgical as well as functional treatment can give good acetabular reconstructions and functional results

Inhibitory humoral responses to the Plasmodium falciparum vaccine candidate EBA-175 are independent of the erythrocyte invasion pathway

    Get PDF
Plasmodium falciparum utilizes multiple ligand-receptor interactions for invasion. The invasion ligand EBA-175 is being developed as a major blood-stage vaccine candidate. EBA-175 mediates parasite invasion of host erythrocytes in a sialic acid-dependent manner through its binding to the erythrocyte receptor glycophorin A. In this study, we addressed the ability of naturally acquired human antibodies against the EBA-175 RII erythrocyte-binding domain to inhibit parasite invasion of ex vivo isolates, in relationship to the sialic acid dependence of these parasites. We have determined the presence of antibodies to the EBA-175 RII domain by enzyme-linked immunosorbent assay (ELISA) in individuals from areas of Senegal where malaria is endemic with high and low transmission. Using affinity-purified human antibodies to the EBA-175 RII domain from pooled patient plasma, we have measured the invasion pathway as well as the invasion inhibition of clinical isolates from Senegalese patients in ex vivo assays. Our results suggest that naturally acquired anti-EBA-175 RII antibodies significantly inhibit invasion of Senegalese parasites and that these responses can be significantly enhanced through limiting other ligand-receptor interactions. However, the extent of this functional inhibition by EBA-175 antibodies is not associated with the sialic acid dependence of the parasite strain, suggesting that erythrocyte invasion pathway usage by parasite strains is not driven by antibodies targeting the EBA-175/glycophorin A interaction. This work has implications for vaccine design based on the RII domain of EBA-175 in the context of alternative invasion pathways
