    Effects of BRCA2 deficiency on telomere recombination in non-ALT and ALT cells

Interestingly, anti cancer treatments that involve BRCA2 and telomerase individually are currently being explored. In the light of the above recent studies their combinatorial targeting may be justified in the development of future treatments. In order to investigate effects of BRCA2 that can be explored for this combinatorial targeting we focused on the analysis of recombination rates at telomeres by monitoring T-SCEs (Telomere Sister Chromatid Exchanges). Results: We observed a significant increase in T-SCE frequencies in four BRCA2 defective human cell lines thus suggesting that BRCA2 suppresses recombination at telomeres. To test this hypothesis further we analyzed T-SCE frequencies in a set of Chinese hamster cell lines with or without functional BRCA2. Our results indicate that introduction of functional BRCA2 normalizes frequencies of T-SCEs thus supporting the notion that BRCA2 suppresses recombination at telomeres. Given that ALT (Alternative Lengthening of Telomeres) positive cells maintain telomeres by recombination we investigated the effect of BRCA2 depletion in these cells. Our results show that this depletion causes a dramatic reduction in T-SCE frequencies in ALT positive cells, but not in non-ALT cells. Conclusion: BRCA2 suppresses recombination at telomeres in cells that maintain them by conventional mechanisms. Furthermore, BRCA2 depletion in ALT positive cells reduces high levels of T-SCEs normally found in these cells.     Analysis of airplane boarding via space-time geometry and random matrix theory

    We show that airplane boarding can be asymptotically modeled by 2-dimensional Lorentzian geometry. Boarding time is given by the maximal proper time among curves in the model. Discrepancies between the model and simulation results are closely related to random matrix theory. We then show how such models can be used to explain why some commonly practiced airline boarding policies are ineffective and even detrimental.Comment: 4 page

    What is the function of parent–child argumentation? This chapter intends to answer the main research question that has guided the study presented in this volume and open a discussion for future research on this topic. In the first part, the chapter provides a detailed overview of the main findings of the analysis of parent–child argumentative discussions during mealtime. The role played by parents and children in the inception and development of argumentation, and the types of conclusions of their argumentative discussions are described. Subsequently, two educational targets achieved by parents and children through their argumentative interactions are presented and critically discussed. In the last part, new open questions that should guide future investigation to expand our knowledge of the role and function of argumentation between parents and children are proposed

    Casting Light Upon The Great Endarkenment

    While the Enlightenment promoted thinking for oneself independent of religious authority, the ‘Endarkenment’ (Millgram 2015) concerns deference to a new authority: the specialist, a hyperspecializer. Non-specialists need to defer to such authorities as they are unable to understand their reasoning. Millgram describes how humans are capable of being serial hyperspecializers, able to move from one specialism to another. We support the basic thrust of Millgram’s position, and seek to articulate how the core idea is deployed in very different ways in relation to extremely different philosophical areas. We attend to the issue of the degree of isolation of different specialists and we urge greater emphasis on parallel hyperspecialization, which describes how different specialisms can be embodied in one person at one time
