Oxidative Stress in Neurodegeneration
It has been demonstrated that oxidative stress has a ubiquitous role in neurodegenerative diseases. Major source of oxidative stress due to reactive oxygen species (ROS) is related to mitochondria as an endogenous source. Although there is ample evidence from tissues of patients with neurodegenerative disorders of morphological, biochemical, and molecular abnormalities in mitochondria, it is still not very clear whether the oxidative stress itself contributes to the onset of neurodegeneration or it is part of the neurodegenerative process as secondary manifestation. This paper begins with an overview of how oxidative stress occurs, discussing various oxidants and antioxidants, and role of oxidative stress in diseases in general. It highlights the role of oxidative stress in neurodegenerative diseases like Alzheimer's, Parkinson's, and Huntington's diseases and amyotrophic lateral sclerosis. The last part of the paper describes the role of oxidative stress causing deregulation of cyclin-dependent kinase 5 (Cdk5) hyperactivity associated with neurodegeneration
Awareness of COVID-19 infection and mental health screening among medical students
Background: Novel corona virus disease (COVID-19) has spread throughout the world causing massive morbidity and mortality. The infection has drastically changed the way of living in the current situation. This change has led to higher levels of fear, anxiety and confusion in the minds of the people. The behavior of people influenced by their knowledge and mental status has a huge bearing on the course of COVID-19 pandemic. This study evaluates the awareness of COVID-19 infection and screens the mental health status of medical students of a tertiary care teaching hospital.Methods: This was a cross-sectional, observational survey conducted on an online platform. A predesigned questionnaire was created using google forms and sent to 912 medical students. Ten questions were used to assess knowledge and awareness on COVID-19 and a fear of COVID-19 scale rated on a 5-point Likert was used to screen mental status. Results: 641 students responded to the survey. Almost 95.5% of students lived with their family during the lockdown period. The mean score was 8.15±1.05 for awareness and knowledge. The overall awareness and knowledge of COVID-19 was satisfactory with 81.49% reporting correct answers. Almost 40% of participants were most afraid of COVID-19, 34% became nervous or anxious, watching news and nearly 20% were afraid of losing life because of COVID-19.Conclusions: Currently in the peak of infection, our focus is on reducing transmission and preventing mortality. Managing psychological issue of people, especially healthcare workers is equally important during this pandemic. Enhanced awareness to disease and providing psychological support will allay COVID-19 fears among the people.
Emission Line Metallicities From The Faint Infrared Grism Survey and VLT/MUSE
We derive direct measurement gas-phase metallicities of for 14 low-mass Emission Line Galaxies (ELGs) at
identified in the Faint Infrared Grism Survey (FIGS). We use deep slitless G102
grism spectroscopy of the Hubble Ultra Deep Field (HUDF), dispersing light from
all objects in the field at wavelengths between 0.85 and 1.15 microns. We run
an automatic search routine on these spectra to robustly identify 71 emission
line sources, using archival data from VLT/MUSE to measure additional lines and
confirm redshifts. We identify 14 objects with with measurable
O[III]4363 \AA\ emission lines in matching VLT/MUSE spectra. For these
galaxies, we derive direct electron-temperature gas-phase metallicities with a
range of . With matching stellar masses in the
range of , we construct a
mass-metallicity (MZ) relation and find that the relation is offset to lower
metallicities compared to metallicities derived from alternative methods
(e.g.,, O3N2, N2O2) and continuum selected samples. Using star
formation rates (SFR) derived from the emission line, we calculate
our galaxies' position on the Fundamental Metallicity Relation (FMR), where we
also find an offset toward lower metallicities. This demonstrates that this
emission-line-selected sample probes objects of low stellar masses but even
lower metallicities than many comparable surveys. We detect a trend suggesting
galaxies with higher Specific Star Formation (SSFR) are more likely to have
lower metallicity. This could be due to cold accretion of metal-poor gas that
drives star formation, or could be because outflows of metal-rich stellar winds

Current biological and pharmacological updates on wogonin
Wogonin (5, 7-Dihydroxy-8-methoxyflavone) is a traditional naturally occurring flavonoid derived from the root extract of Chinese medicine, named Scutellaria baicalensis Georgi. Wogonin contains various biological properties which include allergic diseases, anti-cancertherapy and anti-inflammatory activities. Wogonin also shows the effects of removing toxins and cleansing heat. Wogonin glycosides are known as wogonosides. Oroxindin, a wogonin glucuronide isolated from Oroxylum indicum, is one of the example of wogonoside. It is also used in Japanese herbal supplement named Sho-Saiko-To, as an important active ingredient. One of the major active constituents of wogonin, Scutellaria baicalensis, shows potent anticancer activities both in vivo and in vitro studies. The anticancer therapeutic activity of wogonin has been shown by regulation of different cell signaling pathways , which includes protein kinase B pathway (serine-threonine kinase) and AMP-activated protein kinase pathways. Wogonin also shows positive therapeutic anticancer effects in breast cancer by inhibiting the 5‑LO/BLT2/ERK/IL‑8/MMP‑9 signaling cascade and established a major pharmacological anticancer activity
Identification of potential serum biomarkers of glioblastoma: serum osteopontin levels correlate with poor prognosis
Background: The aim of this study is to identify serum biomarkers with classification and prognosis utility for astrocytoma, in particular glioblastoma (GBM). Methods: Our previous glioma microarray database was mined to identify genes that encode secreted or membrane-localized proteins. Subsequent analysis was done using significant analysis of microarrays, followed by reverse transcription-quantitative PCR (RT-qPCR) and immunohistochemical validation in tumor tissues, ELISA and Western blot validation in sera, and correlation with survival of GBM patients. Results: Significant analysis of microarrays identified 31 upregulated and 3 downregulated genes specifically in GBMs. RT-qPCR validation on an independent set of samples confirmed the GBM-specific differential expression of several genes, including three upregulated (CALU, CXCL9, and TIMP1) and two downregulated (GPX3 and TIMP3) novel genes. With respect to osteopontin (OPN), we show the GBM-specific upregulation by RT-qPCR and immunohistochemical staining of tumor tissues. Elevated serum OPN levels in GBM patients were also shown by ELISA and Western blot. GBM patients with high serum OPN levels had poorer survival than those with low serum OPN levels (median survival 9 versus 22 months respectively; P = 0.0001). Further, we also show high serum TIMP1 levels in GBM patients compared with grade II/III patients by ELISA and downregulation of serum GPX3 and TIMP3 proteins in GBMs compared with normal control by Western blot analysis. Conclusions: Several novel potential serum biomarkers of GBM are identified and validated. High serum OPN level is found as a poor prognostic indicator in GBMs. Impact: Identified serum biomarkers may have potential utility in astrocytoma classification and GBM prognosis
Appraising the therapeutical potentials of Alchornea laxiflora (Benth.) Pax & K. Hoffm., an underexplored medicinal herb: A systematic review
Ethnopharmacological relevance:Alchornea laxiflora (Benth.) Pax & K. Hoffm. (Euphorbiaceae) is an important traditional medicinal plant grown in tropical Africa. The stem, leaves, and root have been widely used in the folk medicine systems in Nigeria, Cameroon, South Africa, and Ghana to treat various ailments, including inflammatory, infectious, and central nervous system disorders, such as anxiety and epilepsy.Material and methods: The scientific name of the plant was validated using the “The Plant List,” “Kew Royal Botanic Gardens,” and Tropicos Nomenclatural databases. The literature search on A. laxiflora was performed using electronic search engines and databases such as Google scholar, ScienceDirect, PubMed, AJOL, Scopus, and Mendeley.Results: To the best of our knowledge, no specific and detailed review has been reported on A. laxiflora. Consequently, this review provides an up-to-date systematic presentation on ethnobotany, phytoconstituents, pharmacological activities, and toxicity profiles of A. laxiflora. Phytochemical investigations disclosed the presence of important compounds, such as alkaloids, flavonoids, phenolics, terpenoids, and fatty acids. Furthermore, various pharmacological activities and traditional uses reported for this botanical drug were discussed comprehensively.Conclusion: This systemic review presents the current status and perspectives of A. laxiflora as a potential therapeutic modality that would assist future researchers in exploring this African botanical drug as a source of novel drug candidates for varied diseases
