The ROTSE-III Robotic Telescope System
The observation of a prompt optical flash from GRB990123 convincingly
demonstrated the value of autonomous robotic telescope systems. Pursuing a
program of rapid follow-up observations of gamma-ray bursts, the Robotic
Optical Transient Search Experiment (ROTSE) has developed a next-generation
instrument, ROTSE-III, that will continue the search for fast optical
transients. The entire system was designed as an economical robotic facility to
be installed at remote sites throughout the world. There are seven major system
components: optics, optical tube assembly, CCD camera, telescope mount,
enclosure, environmental sensing & protection and data acquisition. Each is
described in turn in the hope that the techniques developed here will be useful
in similar contexts elsewhere.Comment: 19 pages, including 4 figures. To be published in PASP in January,

An engineered Tetrahymena tRNA(Gln) for in vivo incorporation of unnatural amino acids into proteins by nonsense suppression
A new tRNA, THG73, has been designed and evaluated as a vehicle for incorporating unnatural amino acids site-specifically into proteins expressed in vivo using the stop codon suppression technique. The construct is a modification of tRNAGln(CUA) from Tetrahymena thermophila, which naturally recognizes the stop codon UAG. Using electrophysiological studies of mutations at several sites of the nicotinic acetylcholine receptor, it is established that THG73 represents a major improvement over previous nonsense suppressors both in terms of efficiency and fidelity of unnatural amino acid incorporation. Compared with a previous tRNA used for in vivo suppression, THG73 is as much as 100-fold less likely to be acylated by endogenous synthetases of the Xenopus oocyte. This effectively eliminates a major concern of the in vivo suppression methodology, the undesirable incorporation of natural amino acids at the suppression site. In addition, THG73 is 4-10-fold more efficient at incorporating unnatural amino acids in the oocyte system. Taken together, these two advances should greatly expand the range of applicability of the in vivo nonsense suppression methodology
Electron impact excitation cross sections for allowed transitions in atoms
We present a semiempirical Gaunt factor for widely used Van Regemorter
formula [Astrophys. J. 136, 906 (1962)] for the case of allowed transitions in
atoms with the LS coupling scheme. Cross sections calculated using this Gaunt
factor agree with measured cross sections to within the experimental error.Comment: RevTeX, 3 pages, 10 PS figures, 2 PS tables, submitted to Phys. Rev.
Refractory Materials for Flame Deflector Protection
Fondu Fyre (FF) is currently the only refractory material qualified for use in the flame trench at KSC's Shuttle Launch Pads 39A and 3913. However, the material is not used as it was qualified and has undergone increasingly frequent and severe degradation due to the launch blasts. This degradation is costly as well as dangerous for launch infrastructure, crew and vehicle. The launch environment at KSC is unique. The refractory material is subject to the normal seacoast environment, is completely saturated with water before launch, and is subjected to vibrations and aggressive heat/blast conditions during launch. This report presents results comparing two alternate materials, Ultra-Tek FS gun mix and Kruzite GR Plus, with Fondu Fyre. The materials were subjected to bulk density, porosity, compression strength, modulus of rupture and thermal shock tests. In addition, test specimens were exposed to conditions meant to simulate the launch environment at KSC to help better understand how the materials will perform once installed
Improved Imputation of Common and Uncommon Single Nucleotide Polymorphisms (SNPs) with a New Reference Set
Statistical imputation of genotype data is an important technique for analysis of genome-wide association studies (GWAS). We have built a reference dataset to improve imputation accuracy for studies of individuals of primarily European descent using genotype data from the Hap1, Omni1, and Omni2.5 human SNP arrays (Illumina). Our dataset contains 2.5-3.1 million variants for 930 European, 157 Asian, and 162 African/African-American individuals. Imputation accuracy of European data from Hap660 or OmniExpress array content, measured by the proportion of variants imputed with R^2^>0.8, improved by 34%, 23% and 12% for variants with MAF of 3%, 5% and 10%, respectively, compared to imputation using publicly available data from 1,000 Genomes and International HapMap projects. The improved accuracy with the use of the new dataset could increase the power for GWAS by as much as 8% relative to genotyping all variants. This reference dataset is available to the scientific community through the NCBI dbGaP portal. Future versions will include additional genotype data as well as non-European populations
Stark Broadening of the B III 2s-2p Lines
We present a quantum-mechanical calculation of Stark line widths from
electron-ion collisions for the , lambda = 2066 and 2067
A, resonance transitions in B III. The results confirm the previous
quantum-mechanical R-matrix calculations but contradict recent measurements and
semi-classical and some semi-empirical calculations. The differences between
the calculations can be attributed to the dominance of small L partial waves in
the electron-atom scattering, while the large Stark widths inferred from the
measurements would be substantially reduced if allowance is made for
hydrodynamic turbulence from high Reynolds number flows and the associated
Doppler broadening.Comment: 21 pages, 4 figures; to be published in Phys. Rev.
