Through this cryptography technique the confidential data can be secured and authenticated

    Effect of growth regulators and Physiological Gradients on the High frequency plant regeneration from the long-term callus cultures of different germplasms of Rice (Oryza sativa L.)

    Callus cultures of rice were initiated from mature embryos of different cultivars on LS medium containing 2 mg/L 2,4-D. Increasing concentrations of 2,4-D and 2,4-5T also increased the frequency of callus initiation in all the cultivars tested. Of different cultivars, Tellahamsa was found to be superior for callus initiation. Genotypic differences for plant regeneration were also observed. Cultivar Tellahamsa showed the highest (65-75%) frequency of plant differentiation followed by DGWG, Yerragaluvadlu, Surekha, Basmati-370, Bala, Chakko amubi, Jaya and IR-8. Callus cultures of rice cultivar Bala grown on a shoot-forming medium (LS + 1 mg/L IAA + 4 mg/L KN + 2% sucrose) were exposed to gibberellic acid and abscisic acid for varying lengths of time and at different periods during culture. Gibberellic acid totally suppressed the organogenesis in callus cultures of rice. The results suggest that if the tissue accumulated sufficient gibberellic acid prior to the initiation of meristemoids and shoot primordia, repression of shoot formation occurred. This repression was not reversed by increasing the levels of IAA and KN in the medium, but abscisic acid could partially overcome the gibberellic acid repression of shoot formation in rice callus. It has been observed in rice that shoots usually emerge from the basal portions of callus. This observation suggested that perhaps physiological gradients of materials were operative during the organ initiation process. To test this hypothesis, starch content and the enzyme activity of malate dehydrogenase in upper and lower portions of shoot-forming and non-shoot-forming callus were determined. Starch began to accumulate in both upper and lower portions of the shoot forming tissues within 4 days of culture. The rate of accumulation however, was faster and more in the lower portion of the callus leading to a peak of accumulation on day 8 in culture, i.e., prior to shoot formation. Non-shoot-forming callus cultures accumulated little starch during the same period of culture. Malate dehydrogenase (MDH) activity was examined in order to know the overall rate of respiration. In the upper segment of shoot-forming callus, the activity of MDH was very high by day 4 but declined continuously thereafter. The rate of activity of the enzyme was significantly higher beyond four days in culture in case of the lower portion of the shoot forming callus. Enzyme activity was lower in the non-shoot-forming portions (both upper and lower) of the callus. The higher rate of enzyme activity for the upper portion of the tissue could be attributed to increased oxygen availability. Thus, the evidence for the idea that concentrations of gradients or physiological gradients of substances into the callus tissue may be the operative factors promoting organ initiation in vitro is presented

    Arthroscopic reattachment of tibial avulsion fractures of the posterior cruciate ligament using ABS button and tightrope (Mohandas Jagsun technique)

    Background: Avulsion fractures of tibial insertion of the posterior cruciate ligament are rare among the knee injuries. Purpose of this study is to determine the functional outcome of arthroscopic reattachment of tibial avulsion of the Posterior cruciate ligament by ABS button (Arthrex) and tight rope.Methods: 15 Patients with PCL avulsion fracture were included. The Inclusion criteria were: 1) Displaced avulsion fractures (type-2 and type-3). They were followed at regular intervals using IKDC score, Lysholm score and subjective questionnaire.Results: All cases showed complete osseous union during follow up. All knees are stable on examination by posterior sag sign and posterior drawer test. Two patients had loss of about 100 flexion. According to the IKDC form assessment 13 patients classified as normal and 2 were classified as near normal.Conclusions: Arthroscopic reattachment of tibial avulsion fractures of the posterior cruciate ligament using ABS button and tightrope gives fairly good clinical outcomes.

    Kinetics of Methylation by EcoP1I DNA Methyltransferase

    EcoP1I DNA MTase (M.EcoP1I), an N6-adenine MTase from bacteriophage P1, is a part of the EcoP1I restriction-modification (R-M) system which belongs to the Type III R-M system. It recognizes the sequence 5′-AGACC-3′ and methylates the internal adenine. M.EcoP1I requires Mg2+ for the transfer of methyl groups to DNA. M.EcoP1I is shown to exist as dimer in solution, and even at high salt concentrations (0.5 M) the dimeric M.EcoP1I does not dissociate into monomers suggesting a strong interaction between the monomer subunits. Preincubation and isotope partitioning studies with M.EcoP1I indicate a kinetic mechanism where the duplex DNA binds first followed by AdoMet. Interestingly, M.EcoP1I methylates DNA substrates in the presence of Mn2+ and Ca2+ other than Mg2+ with varying affinities. Amino acid analysis and methylation assays in the presence of metal ions suggest that M.EcoP1I has indeed two metal ion-binding sites [358ID(x)n … ExK401 and 600DxDxD604 motif]. EcoP1I DNA MTase catalyzes the transfer of methyl groups using a distributive mode of methylation on DNA containing more than one recognition site. A chemical modification of EcoP1I DNA MTase using N-ethylmaleimide resulted in an irreversible inactivation of enzyme activity suggesting the possible role of cysteine residues in catalysis

    Effects of Chest Compression Fraction on Return of Spontaneous Circulation in Patients with Cardiac Arrest; a Brief Report

    Introduction: The association between chest compression fraction (CCF) and return of spontaneous circulation (ROSC) has been a controversial issue in literature; and both positive and negative correlations have been reported between CCF and survival rate. Objective: The present study was conducted to determine the relationship between the rate and outcomes of chest compression and between CCF and ROSC in patients with cardiac arrest. Method: The present prospective observational study was conducted during 2018 on patients with cardiac arrest aged 18-80 years. Participants with end-stage renal diseases, malignancies and grade IV heart failure were excluded. A stop watch was set upon the occurrence of a code blue in the emergency department, and time was recorded by the observer upon the arrival of the code blue team leader (a maximum permissible duration of 10 minutes). The interruptions in chest compressions were recorded using a stopwatch, and CCF was calculated by dividing the duration of chest compression by the total duration of cardiac arrest observed. Results: Totally, 45 participants were enrolled. Most of the patients had non-shockable rhythms and underwent CPR based on related algorithm. Hypoxia and hypovolemia were the two probable etiology of cardiac arrest; and coronary artery disease was the most prevalent underlying disease. All patients with ROSC had CCF more than 70%. A CCF below 70% was observed in 21 cases (46.7%), and a fraction of at least 70% in 24 cases. All patients with ROSC had CCF more than 70%. A significantly higher duration and fraction of chest compression was observed in the participants who attained ROSC (P<0.001). Conclusion: Based on the findings of current study, it seems that significantly higher chest compression durations and fractions were found to be associated with ROSC, which was achieved in the majority of the participants with a CCF of at least 80%
