Physiological indicators of the interaction between water deficit and soil acidity in sugarcane
O objetivo deste trabalho foi avaliar os indicadores fisiológicos da interação entre deficit hídrico e acidez do solo em plantas jovens de cana-de-açúcar. As plantas foram submetidas a três tratamentos de disponibilidade hídrica, medidos em percentagem de capacidade de campo (CC) – sem estresse (70% CC), estresse moderado (55% CC) e estresse severo (40% CC); e três tratamentos de acidez no solo, medidos em termos de saturação por bases (V) – baixa acidez (V = 55%), média acidez (V = 33%) e alta acidez (V = 23%). O experimento foi realizado em casa de vegetação a 29,7±4,3oC e 75±10% UR. O delineamento experimental utilizado foi o de blocos ao acaso, em esquema fatorial 3x3, com quatro repetições. Após 60 dias, foram determinados os teores de solutos compatíveis – trealose, glicina betaína e prolina – na folha diagnóstico e o crescimento inicial da parte aérea. Os solutos compatíveis trealose, glicina betaína e prolina são indicadores do efeito da interação dos estresses hídrico e ácido no solo. O acúmulo dos solutos compatíveis nos tecidos foliares das plantas não é capaz de impedir a redução na produção de matéria seca da cana-de-açúcar, resultante do agravamento nas condições de disponibilidade hídrica e de acidez no solo.The aim of this work was to assess the physiological indicators of the interaction between water deficit and soil acidity, in sugarcane. The plants were submitted to three treatments of water availability – no stress (70% of field capacity, FC), moderate stress (55% FC), and extreme stress (40% FC); and three acidity treatments – no acidity [base saturation (V) = 55%], average acidity (V = 33%), and high acidity (V = 23%). The experiment was carried out in greenhouse, with 29.7±4.3oC and 75±10% RH. The experimental design was in randomized blocks, in 3x3 factorial arrangement, with four replicates. After 60 days, the contents of compatible solutes – trehalose, glycine betaine and proline – in the diagnostic leaf and the initial growth of shoots were determined. The compatible solutes trehalose, glycine betaine, and proline are indicators of the interaction of water and acidity stresses in the soil. The accumulation of compatible solutes in plant foliar tissues can not prevent sugarcane losses in dry matter production, caused by increasing water deficit and soil acidity
Harvest types and times of gliphosate application in the eradication of sugarcane ratoons
O objetivo deste trabalho foi avaliar a sensibilidade de quatro genótipos de cana-de-açúcar, ao glifosato aplicado para a erradicação de soqueiras de duas idades, em dois sistemas de colheita, em um Latossolo Vermelho eutroférrico. O produto foi aplicado na dose de 2.400 g ha-1 de i.a., aos 40 e 55 dias de idade da touceira, depois do terceiro corte. Para quantificar a ação do glifosato, foram contados os perfilhos vivos e mortos aos 30, 45 e 60 dias depois da aplicação do produto. Em relação à porcentagem de morte dos perfilhos, ocorrem diferenças entre os genótipos quanto à suscetibilidade ao glifosato. A dose aplicada não foi suficiente para erradicar 100% dos perfilhos, mas para os genótipos IAC87-3184, RB835486 e SP87-344 o controle foi superior a 80%, tendo sido considerados sensíveis/intermediários, enquanto o IAC91-5155 foi considerado tolerante. O sistema de colheita da cana-de-açúcar não interferiu na eficiência do glifosato na eliminação da soqueira, entretanto, a aplicação feita aos 40 dias da touceira apresentou a maior porcentagem de perfilhos mortos.The objective of this work was to evaluate sensibility of four sugarcane genotypes to gliphosate application for the eradication of ratoons of two ages, and in two crop systems, in a Rhodic Eutrudox. The product was applied in the dose (a.i.) of 2,400 g ha-1 to the 40 and 55 day-old shoots of the third cut harvest. In order to quantify the action of the glyphosate, alive and dead tillerings were counted in the 30th, 45th and 60th day after its application. In relation to the percentage of tillerings death, there were differences among the genotypes as for the susceptibility to the glyphosate. The applied dose was not enough to eradicate 100% of the tillerings, but for the genotypes IAC87-3184, RB835486 and SP87-344 the control was superior to 80%, being considered sensitive/intermediate, while the IAC91-5155 was considered tolerant. Crop system did not interfere in the glyphosate efficiency for ratoon elimination, however, the application made to 40 day-old shoots presented the largest died tillerings percentage
Biochemical and physiological responses of sugarcane cultivars to soil water deficiencies
For sugarcane (Saccharum spp.) crops, the effects of an environmental stress, especially water deficiency, may cause severe productivity reduction, inferring negatively in the sugarcane industry. The tolerance of two sugarcane cultivars to a lack of water was made by analyzing the levels of the osmoprotectors, trehalose and free proline, and the biometrical variables of their initial growth. Biochemical and physiological responses of the cultivars, when subjected to water stress, were assayed to determine how these plants tolerate drought. The study was conducted in an acclimatized greenhouse (29.7 ± 4.3ºC and 75.0 ± 10.1% relative humidity) during 100 days and was divided into random blocks using a factorial 2 × 3 × 2 design (sugarcane cultivars × water availability × time periods) with four replicates. Forty days after germination, sugarcane was planted in pots (12 dm³) containing topsoil material taken from a medium textured Rhodic Ferralsol, submitted to three levels of water availability (WAS): 55% (control), 40% (moderate stress) and 25% (severe stress), for 60 days. The effect of the WAS on the accumulation of trehalose and free proline was detected in both cultivars, although it was found to be more distinctive for the cv. IAC91-5155. Trehalose and free proline are biochemical and physiological indicators of water deficiency. The cv. IAC91-5155 had altered growth and allocation of biomass when subjected to severe water stress conditions. The univariate and the multivariate analysis of the biochemical and physiological responses, presented by the IAC91-5155 cultivar, indicate relative tolerance to drought conditions
Biomass Accumulation and Cell Wall Structure of Rice Plants Overexpressing a Dirigent-Jacalin of Sugarcane (ShDJ) Under Varying Conditions of Water Availability
A sugarcane gene encoding a dirigent-jacalin, ShDJ, was induced under drought stress. To elucidate its biological function, we integrated a ShDJ-overexpression construction into the rice Nipponbare genome via Agrobacterium-mediated transformation. Two transgenic lines with a single copy gene in T0 were selected and evaluated in both the T1 and T4 generations. Transgenic lines had drastically improved survival rate under water deficit conditions, at rates close to 100%, while WT did not survive. Besides, transgenic lines had improved biomass production and higher tillering under water deficit conditions compared with WT plants. Reduced pectin and hemicellulose contents were observed in transgenic lines compared with wild-type plants under both well-watered and water deficit conditions, whereas cellulose content was unchanged in line #17 and reduced in line #29 under conditions of low water availability. Changes in lignin content under water deficit were only observed in line #17. However, improvements in saccharification were found in both transgenic lines along with changes in the expression of OsNTS1/2 and OsMYB58/63 secondary cell wall biosynthesis genes. ShDJ-overexpression up-regulated the expression of the OsbZIP23, OsGRAS23, OsP5CS, and OsLea3 genes in rice stems under well-watered conditions. Taken together, our data suggest that ShDJ has the potential for improving drought tolerance, plant biomass accumulation, and saccharification efficiency
Mecanismos fisiológicos de cana-de-açúcar sob efeito da interação dos estresses hídrico e ácido no solo
No presente trabalho foi verificado o ajuste osmótico no crescimento inicial de cana-de-açúcar, cv. IAC91-5155, sob efeito da interação dos estresses hídrico e ácido no solo. Para tanto, foram quantificados os teores de três solutos compatíveis, prolina, glicina betaína e trealose. Também foi estudada a atividade da nitrato redutase e os teores de clorofilas. O estudo de todas estas variáveis teve por objetivo propor a utilização destes compostos como indicadores fisiológicos de aclimatação da cana-deaçúcar sob efeito da interação de ambos os estresses. O experimento foi realizado em casa de vegetação com 29,7±4,3oC e 75±10 UR%. Foram utilizados três tratamentos de disponibilidade hídrica (capacidade de campo, CC): controle (70%), estresse moderado (55%) e estresse severo (40%) e três tratamentos de acidez no solo (saturação por bases, V%): controle (55%), média acidez (33%) e alta acidez (23%). O delineamento experimental utilizado foi o de blocos casualizados em esquema fatorial 3x3, com quatro repetições. Após 60 dias sob estresses foram determinados os teores dos solutos compatíveis, a atividade da nitrato redutase e os teores das clorofilas. Os resultados mostram que houve acúmulo dos três solutos compatíveis evidenciando que ocorreu ajuste osmótico. Prolina e glicina betaína são indicadores fisiológicos do efeito da interação de ambos os estresses enquanto trealose é indicadora apenas para o estresse hídrico. Também, a atividade da nitrato redutase pode ser utilizada como indicador do estresse hídrico e os teores das clorofilas a, b e total para os estresses hídrico ou ácido. A massa de matéria seca da parte aérea, o número de folhas e a área foliar das plantas jovens de cana-de-açúcar, ajustadas osmoticamente, são reduzidos pelos estresses. Os efeitos interativos de ambos...In this present work was verified the osmotic adjustment in the initial plant growth sugarcane cv. IAC91-5155 under interaction effect of water and acid stress in the soil. For this, were quantified three compatible solutes content, as follows: proline, glycine betaine and trehalose. It was verified also the nitrate reductase activity as well the chlorophyll content. The study of all parameters aimed to propose the use components as physiological indicators to acclimation of sugarcane plants under interaction the both stress. The experiment was carried out in a greenhouse under 29.7±4.3oC and 75±10%. Three water availability treatments (Field Capacity – FC%): control (70%), moderate stress (55%), and extreme stress (40%) and three soil acidification treatment (Base Saturation – BS%): control (55%), average acidity (33%), and high acidity (23%) were used. The experimental design used was the randomized blocks in a 3x3 factorial scheme and four replicates. After sixty days under stress were determined compatible solutes, nitrate reductase and chlorophyll content. Our results showed that there was the accumulation of three compatible solutes exhibiting osmotic adjustment. Glycine betaine and proline are both physiologic indicators of stresses interaction effect whereas trehalose indicates only water stress. The nitrate reductase activity can be used as water stress indicator and chlorophylls a, b and total are indicators to water or acid stress. The dry mass of shoot part, leaves number and leaf area are significantly reduced by stress in young sugarcane plants adjusted osmotically. Atividade da enzima redutase do nitrato e teores de prolina livre em raízes de cana-de-açúcar sob os estresses hídrico e ácido no solo
The aim of this work was evaluate the physiological indicators of association between water deficiency and soil acidity, by determining the nitrate reductase activity, the levels of free proline and initial growth of the root system of seedlings of sugarcane cv. IAC91-5155. After 30 days, the seedlings were transferred to plastic pots with 12 dm3 of dystrophic alic Red Latosol (Oxisol) and submitted to association of three treatments of water availability: no stress (70%), moderate stress (55%) and severe stress (40%), in according with field capacity and three acidity treatments: no stress (55%), moderate stress (33%) and severe stress (23%), considering the base saturation. The experimental design was that of random blocks under factorial scheme of 3x3, with four replicates. After 60 days under the stress association, the levels of free proline, the nitrate reductase activity and the growth of the sugarcane roots system were evaluated in seedlings of sugarcane. The nitrate reductase enzyme activity can be considered a physiological indicator of the effect of the association of acid and water stress in moderate conditions in soil, while the free proline can be considered physiological indicator to both stress in severe conditions.
