Construction of human induced pluripotent stem cell-derived oriented bone matrix microstructure by using in vitro engineered anisotropic culture model
Bone tissue has anisotropic microstructure based on collagen/biological apatite orientation, which plays essential roles in the mechanical and biological functions of bone. However, obtaining an appropriate anisotropic microstructure during the bone regeneration process remains a great challenging. A powerful strategy for the control of both differentiation and structural development of newly-formed bone is required in bone tissue engineering, in order to realize functional bone tissue regeneration. In this study, we developed a novel anisotropic culture model by combining human induced pluripotent stem cells (hiPSCs) and artificially-controlled oriented collagen scaffold. The oriented collagen scaffold allowed hiPSCs-derived osteoblast alignment and further construction of anisotropic bone matrix which mimics the bone tissue microstructure. To the best of our knowledge, this is the first report showing the construction of bone mimetic anisotropic bone matrix microstructure from hiPSCs. Moreover, we demonstrated for the first time that the hiPSCs-derived osteoblasts possess a high level of intact functionality to regulate cell alignment. © 2017 The Authors Journal of Biomedical Materials Research Part A Published by Wiley Periodicals, Inc. Hinode Calibration for Precise Image Co-alignment between SOT and XRT (November 2006 -- April 2007)
To understand the physical mechanisms for activity and heating in the solar
atmosphere, the magnetic coupling from the photosphere to the corona is an
important piece of information from the Hinode observations, and therefore
precise positional alignment is required among the data acquired by different
telescopes. The Hinode spacecraft and its onboard telescopes were developed to
allow us to investigate magnetic coupling with co-alignment accuracy better
than 1 arcsec. Using the Mercury transit observed on 8 November 2006 and
co-alignment measurements regularly performed on a weekly basis, we have
determined the information necessary for precise image co-alignment and have
confirmed that co-alignment better than 1 arcsec can be realized between Solar
Optical Telescope (SOT) and X-Ray Telescope (XRT) with our baseline
co-alignment method. This paper presents results from the calibration for

Allosteric regulation by cooperative conformational changes of actin filaments drives mutually exclusive binding with cofilin and myosin
Heavy meromyosin (HMM) of myosin II and cofilin each binds to actin filaments cooperatively and forms clusters along the filaments, but it is unknown whether the two cooperative bindings are correlated and what physiological roles they have. Fluorescence microscopy demonstrated that HMM-GFP and cofilin-mCherry each bound cooperatively to different parts of actin filaments when they were added simultaneously in 0.2 μM ATP, indicating that the two cooperative bindings are mutually exclusive. In 0.1 mM ATP, the motor domain of myosin (S1) strongly inhibited the formation of cofilin clusters along actin filaments. Under this condition, most actin protomers were unoccupied by S1 at any given moment, suggesting that transiently bound S1 alters the structure of actin filaments cooperatively and/or persistently to inhibit cofilin binding. Consistently, cosedimentation experiments using copolymers of actin and actin-S1 fusion protein demonstrated that the fusion protein affects the neighboring actin protomers, reducing their affinity for cofilin. In reciprocal experiments, cofilin-actin fusion protein reduced the affinity of neighboring actin protomers for S1. Thus, allosteric regulation by cooperative conformational changes of actin filaments contributes to mutually exclusive cooperative binding of myosin II and cofilin to actin filaments, and presumably to the differential localization of both proteins in cells
Solar Lyman-Alpha Polarization Observation of the Chromosphere and Transition Region by the Sounding Rocket Experiment CLASP
We are planning an international rocket experiment Chromospheric Lyman-Alpha Spectro-Polarimeter (CLASP) is (2015 planned) that Lyman line (Ly(alpha) line) polarization spectroscopic observations from the sun. The purpose of this experiment, detected with high accuracy of the linear polarization of the Ly(alpha) lines to 0.1% by using a Hanle effect is to measure the magnetic field of the chromosphere-transition layer directly. For polarization photometric accuracy achieved that approx. 0.1% required for CLASP, it is necessary to realize the monitoring device with a high throughput. On the other hand, Ly(alpha) line (vacuum ultraviolet rays) have a sensitive characteristics that is absorbed by the material. We therefore set the optical system of the reflection system (transmission only the wavelength plate), each of the mirrors, subjected to high efficiency of the multilayer coating in accordance with the role. Primary mirror diameter of CLASP is about 30 cm, the amount of heat about 30,000 J is about 5 minutes of observation time is coming mainly in the visible light to the telescope. Oriented Collagen Scaffolds for Tissue Engineering
Oriented collagen scaffolds were developed in the form of sheet, mesh and tube by arraying flow-oriented collagen string gels and dehydrating the arrayed gels. The developed collagen scaffolds can be any practical size with any direction of orientation for tissue engineering applications. The birefringence of the collagen scaffolds was quantitatively analyzed by parallel Nicols method. Since native collagen in the human body has orientations such as bone, cartilage, tendon and cornea, and the orientation has a special role for the function of human organs, the developed various types of three-dimensional oriented collagen scaffolds are expected to be useful biomaterials for tissue engineering and regenerative medicines
