Regulation of antioxidant enzyme activities in female rat brain by ovarian steroids
The sex steroids produce their effects by acting on numerous target tissues and organs, such as the reproductive organs, bone tissue and cartilage, peripheral blood vessels as well as the central nervous system (CNS). In our studies we have monitored the change of enzyme activity of the antioxidant (AO) system in the brain of female rats depending on the ovarian steroids. We have chosen it as a new parameter that might represent an important indicator of the changes within the CNS, bearing in mind the biological importance of the enzymes of the AO system. The experimental results of our study indicate that the enzyme activity of the AO system in the brain tissue of female rats shows a certain dependence on the concentration of ovarian hormones, progesterone and estradiol, in the organism. Study of the activity of the enzymes of the AO system in the brain of female rats depending on the influence of ovarian hormones can answer whether the action of ovarian steroids on the CNS includes maintenance of a dynamic equilibrium of free radicals in the neurons
Activity of antioxidative defense enzymes in the blood of patients with liver echinococcosis
Background/Aim. Chronic echinococcocal disease is the parasite human disease caused by the penetration of larval (asexual) stages of the canine tapeworm (Echinococcus granulosus) in the liver of humans. After the penetration of the parasite, the host organism react by activating complement- depending immune response. The aim of this study was to elucidate the influence of larval form of Echinococcus granulosus in the liver on the activity of antioxidative defense enzymes in the blood of patients before and after the surgical intervention. Methods. We investigated the activity of antioxidative defense enzymes: copper/zinc containing superoxide dismutase (CuZn SOD), catalase (CAT), glutathione peroxidase (GSH-Px), glutathione reductase (GR) and glutathione-S-transferase (GST) in the blood of patients before and after the surgical intervention in respect to the controls, clinically healthy persons. Results. Our results showed that the activity of the GSH-Px was significantly decreased in the plasma of the patients with echinocococal disease before the surgery in respect to the controls. The activity of GST was significantly higher in the blood of the patients after the surgery in comparison to the controls. Conclusion. Chronic liver echinoccocal disease caused significant changes of some antioxidative defense enzymes, first of all Se-dependent enzyme GSH-Px, which could be a suitabile biomarker in the biochemical evaluation of the disease. This work represents a first comprehensive study of the activity of antioxidative defense enzymes in cronic liver echinococcocosis in the patients before and after the surgical intervention in respect to the clinically healthy persons
Time course of hematological parameters in bleeding-induced anemia
In order to investigate daily changes of hematological parameters in bleeding-induced anemia, we treated Wistar albino male rats by daily bleeding (1.5-2 mL of blood from the tail vein for eight days). Blood samples were taken before (on day zero) and on the first to eighth days of bleeding. The values of hematocrit, hemoglobin, and erythrocyte count decreased significantly after the second, sixth, and second days of bleeding, respectively. The number of leukoĀcytes and platelets, as well as Heinz body levels, increased significantly after the third and second days of treatment. The percentage of reticulocytes increased significantly from the second day and attained the maximum level (32.55 Ā± 0.96%) on the eighth day

The results suggest that CP intoxication induces oxidative stress and alters the glutathione redox status: reduced glutathione (GSH), oxidized glutathione (GSSG) and the GSH/GSSG ratio (GSH RI), resulting in increased lipid peroxidation (LPO) in rat liver. The pretreatment with selenium prior to CP treatment showed a protective effect against the toxic influence of CP on peroxidation of the membrane lipids and an altering of the glutathione redox status in the liver of rats. From our results we conclude that selenium functions as a potent antioxidant and suggest that it can control CP-induced hepatotoxicity in rats

Mužjaci Wistar albino pacova su inicirani jednom dozom cisplatina (7.5 mg CP/kg t.m., i.p.) i selena (6 mg Se/kg t.m, kao Na2SeO3, i.p.) pojedinaÄno ili u kombinaciji. Rezultati pokazuju da intoksikacija SR uzrokuje oksidacioni stres i promenu glutation redoks statusa: redukovanog (GSH), oksidovanog (GSSG) i GSH/GSSG indeksa (GSH RI), kao i poveÄanje lipidne peroksidacije (LPO) u jetri pacova. Tretman Se koji je prethodio tretmanu SR pokazao je zaÅ”titne efekte protiv toksiÄnog delovanja SR na peroksidaciju lipida membrane i promenu glutation redoks statusa u jetri pacova. Na osnovu naÅ”ih rezultata zakljuÄujemo da Se, deluje kao snažan antioksidans i da može imati ulogu u kontroli SR indukovane hepatotoksiÄnosti kod pacova.Projekat ministarstva br. 143035
Aktivnost superoksid-dismutaze tokom maturacije kulture Äelija b-16 melanoma miÅ”a
The C3 clone of B-16 mouse melanoma was cultured for 1, 6 and 9 days and analyzed. The changes which are not directly linked to melanogenesis in the B-16 / C3 cultures during their maturation were characterized. Early (1 day) confluent (6 days) and old (9 days) cell cultures are distinguished by their leucine aminopeptidase (LAP) and Ī±-naphthyl acetate esterase (ANAE) isoenzyme patterns. Both quantitative and qualitative changes in LAP and ANAE isoenzyme can be observed during culture maturation. There is an increase in the activity of the enzyme copper, zinc-containing superoxide-dismutase (CuZn SOD). The increaase in the CuZn SOD enzyme activity might be related to B-16/C3 cell melanogenesis and / or to differentiation.C3 klonovi B-16 melanoma miÅ”a su gajeni u kulturi 1, 6 i 9 dana i potom analizirani. IzvrÅ”ena je karakterizacija promena B-16/C3 Äelijskih kultura tokom njihove maturacije koje nisu direktno povezane sa melanogenezom. Pokazano je da se kulture u poÄetnoj fazi (1 dan), konfluentnoj fazi (6 dana) i kasnoj fazi (9 dana) Äelijskog rasta razlikuju na osnovu izoenzimskih spektara leucin-aminopeptidaze i alfa-naftil-acetat-esteraze. Tokom razvitka Äelijskih kultura B-16/C3 zapažene su kvantitativne i kvalitativne promene leucin-aminopeptidaze i alfa-naftil-acetat-esteraze u njihovim izoenzimskim spektrima. UtvrÄeno je da postoji porast aktivnosti bakar-cink superoksid-dismutaze tokom starenja kulture. Cilj ovog rada bio je da se utvrdi da li razvoj larvenog oblika pseÄe pantljiÄare (Echinococcus granulosus) u jetri ima uticaja na aktivnost enzima sistema zaÅ”tite od oksidacionih oÅ”teÄenja u krvi obolelih pre i posle operativnog zahvata. Metode. Ispitivane su promene aktivnosti enzima sistema zaÅ”tite od oksidacionih oÅ”teÄenja: bakar/cink sadržavajuÄe superoksid-dismutaze (CuZn SOD), katalaze (CAT), glutation-peroksidaze (GSH-Px), glutation-reduktaze (GR) i glutation-S-transferaze (GST) u krvi obolelih od ehinokokusne bolesti jetre pre i posle operacije u odnosu na kontrolne, kliniÄki zdrave osobe. Rezultati. Ustanovljen je statistiÄki znaÄajan pad aktivnosti enzima GSH-Px u plazmi bolesnika obolelih od ehinokokusne bolesti jetre pre operacije u odnosu na kontrolu. Aktivnost enzima GST bila je statistiÄki znaÄajno poveÄana u krvi obolelih od hroniÄne ehinokokusne bolesti jetre posle operacije u odnosu na kontrolne vrednosti. ZakljuÄak. HroniÄna ehinokokusna bolest jetre izaziva promene aktivnosti nekih enzima sistema zaÅ”tite od oksidacionih oÅ”teÄenja, pre svega Se-zavisnog enzima GSH-Px, Å”to može predstavljati dobar biomarker u daljoj biohemijskoj evaluaciji ove bolesti. Biochemical and ultrastructural changes in the liver of European perch (Perca fluviatilis L.) in response to cyanobacterial bloom in the Gruža reservoir
We investigated the biochemical and ultrastructural changes in the liver of the freshwater fish, European perch (Perca fluviatilis), in response to Aphanizomenon flos-aquae bloom in the Gruža Reservoir, Serbia. The activities of total manganese- and copper zinc-containing superoxide dismutase (Tot SOD, Mn-SOD, Cu/Zn-SOD), catalase (CAT), glutathione peroxidase (GSH-Px), glutathione reductase (GR) and biotransformation phase II enzyme glutathione-S-transferase (GST), as well as concentrations of total glutathione (GSH) and sulfhydryl (-SH) groups were examined before and during the bloom period. Mn-SOD activity was significantly higher, while the activities of Cu/Zn-SOD, CAT and GSH-Px and the concentration of the -SH groups were significantly lower during the bloom. The ultrastructure of the liver revealed necrotic and apoptotic damage to the hepatocytes during the bloom period. Our work represents the first study to report the influences of an Aphanizomenon flos-aquae bloom in the Gruža Reservoir on antioxidant biomarkers and on histopathological alterations in the liver of the freshwater fish European perch (Perca fluviatilis)
