    Estimation of the nutritive value of grape pomace for ruminant using gas production technique

    The aim of this study was to determine the chemical composition and estimation of nutritive value of white grape pomace (WGP) using in vitro gas production technique. Fermentation of WGP samples were carried out with rumen fluids obtained from three mature cannulated steers. The samples were collected from a factory in Urmia, Iran. The amount of gas production for WGP at 2, 4, 6, 8, 12, 24, 48, 72 and 96 h were measured. The results showed that the crude protein (CP), neutral detergent fiber (NDF), acid detergent fiber (ADF) and non-fibrous carbohydrate (NFC) contents were 17.27, 59.5, 52.5 and 13.5%, respectively. Gas production at 24 h and potential gas production (a + b) were 30.92 and 79.89 ml, respectively. The organic matter digestibility (OMD), metabolizable energy (ME) and short chain fatty acid (SCFA) contents were 50.50%, 7.4 MJ kg-1 DM and 0.69 mmol, respectively. The net energy for lactation (NEL) content was 3.31 MJ kg-1 DM. According to the results of this study, it seems that WGP could be used as a valuable food industrial by-product in ruminant nutrition.Key words: Nutritive value, gas production, grape pomace, short chain fatty acid, metabolizable energy

    Combined modalities of resistance in an oxaliplatin-resistant human gastric cancer cell line with enhanced sensitivity to 5-fluorouracil

    To identify mechanisms underlying oxaliplatin resistance, a subline of the human gastric adenocarcinoma TSGH cell line, S3, was made resistant to oxaliplatin by continuous selection against increasing drug concentrations. Compared with the parental TSGH cells, the S3 subline showed 58-fold resistance to oxaliplatin; it also displayed 11-, 2-, and 4.7-fold resistance to cis-diammine-dichloroplatinum (II) (CDDP), copper sulphate, and arsenic trioxide, respectively. Interestingly, S3 cells were fourfold more susceptible to 5-fluorouracil-induced cytotoxicity due to downregulation of thymidylate synthase. Despite elevated glutathione levels in S3 cells, there was no alteration of resistant phenotype to oxaliplatin or CDDP when cells were co-treated with glutathione-depleting agent, l-buthionine-(S,R)-sulphoximine. Cellular CDDP and oxaliplatin accumulation was decreased in S3 cells. In addition, amounts of oxaliplatin- and CDDP–DNA adducts in S3 cells were about 15 and 40% of those seen with TSGH cells, respectively. Western blot analysis showed increased the expression level of copper transporter ATP7A in S3 cells compared with TSGH cells. Partial reversal of the resistance of S3 cells to oxaliplatin and CDDP was observed by treating cell with ATP7A-targeted siRNA oligonucleotides or P-type ATPase-inhibitor sodium orthovanadate. Besides, host reactivation assay revealed enhanced repair of oxaliplatin- or CDDP-damaged DNA in S3 cells compared with TSGH cells. Together, our results show that the mechanism responsible for oxaliplatin and CDDP resistance in S3 cells is the combination of increased DNA repair and overexpression of ATP7A. Downregulation of thymidylate synthase in S3 cells renders them more susceptible to 5-fluorouracil-induced cytotoxicity. These findings could pave ways for future efforts to overcome oxaliplatin resistance

    Worldwide comparison of survival from childhood leukaemia for 1995–2009, by subtype, age, and sex (CONCORD-2): a population-based study of individual data for 89 828 children from 198 registries in 53 countries

    Background Global inequalities in access to health care are reflected in differences in cancer survival. The CONCORD programme was designed to assess worldwide differences and trends in population-based cancer survival. In this population-based study, we aimed to estimate survival inequalities globally for several subtypes of childhood leukaemia. Methods Cancer registries participating in CONCORD were asked to submit tumour registrations for all children aged 0-14 years who were diagnosed with leukaemia between Jan 1, 1995, and Dec 31, 2009, and followed up until Dec 31, 2009. Haematological malignancies were defined by morphology codes in the International Classification of Diseases for Oncology, third revision. We excluded data from registries from which the data were judged to be less reliable, or included only lymphomas, and data from countries in which data for fewer than ten children were available for analysis. We also excluded records because of a missing date of birth, diagnosis, or last known vital status. We estimated 5-year net survival (ie, the probability of surviving at least 5 years after diagnosis, after controlling for deaths from other causes [background mortality]) for children by calendar period of diagnosis (1995-99, 2000-04, and 2005-09), sex, and age at diagnosis (< 1, 1-4, 5-9, and 10-14 years, inclusive) using appropriate life tables. We estimated age-standardised net survival for international comparison of survival trends for precursor-cell acute lymphoblastic leukaemia (ALL) and acute myeloid leukaemia (AML). Findings We analysed data from 89 828 children from 198 registries in 53 countries. During 1995-99, 5-year agestandardised net survival for all lymphoid leukaemias combined ranged from 10.6% (95% CI 3.1-18.2) in the Chinese registries to 86.8% (81.6-92.0) in Austria. International differences in 5-year survival for childhood leukaemia were still large as recently as 2005-09, when age-standardised survival for lymphoid leukaemias ranged from 52.4% (95% CI 42.8-61.9) in Cali, Colombia, to 91.6% (89.5-93.6) in the German registries, and for AML ranged from 33.3% (18.9-47.7) in Bulgaria to 78.2% (72.0-84.3) in German registries. Survival from precursor-cell ALL was very close to that of all lymphoid leukaemias combined, with similar variation. In most countries, survival from AML improved more than survival from ALL between 2000-04 and 2005-09. Survival for each type of leukaemia varied markedly with age: survival was highest for children aged 1-4 and 5-9 years, and lowest for infants (younger than 1 year). There was no systematic difference in survival between boys and girls. Interpretation Global inequalities in survival from childhood leukaemia have narrowed with time but remain very wide for both ALL and AML. These results provide useful information for health policy makers on the effectiveness of health-care systems and for cancer policy makers to reduce inequalities in childhood survival

    Numerical investigation of the thermo‐hydraulic performance of water‐based nanofluids in a dimpled channel flow using Al 2

    In this study, the authors study the impact of spherical dimple surfaces and nanofluid coolants on heat transfer and pressure drop. The main objective of this paper is to evaluate the thermal performance of nanofluids with respect to different Reynolds numbers (Re) and nanoparticle compositions in dimpled channel flow. Water-based nanofluids with Al2O3, CuO, and Al2O3-CuO nanoparticles are considered for this investigation with 1%, 2%, and 4% volume fraction for each nanofluid. The simulations are conducted at low Reynolds numbers varying from 500 to 1250, assuming constant and uniform heat flux. The effective properties of nanofluids are estimated using models proposed in the literature and are combined with the computational fluid dynamics solver ANSYS Fluent for the analysis. The results are discussed in terms of heat transfer coefficient, temperature distributions, pressure drop, Nusselt number, friction factors, and performance criterion for all the cases. For all cases of different nanoparticle compositions, the heat transfer coefficient was seen as 35%-46% higher for the dimpled channel in comparison with the smooth channel. Besides, it was observed that with increasing volume fraction, the values of heat transfer and pressure drop were increased. With a maximum of 25.18% increase in the thermal performance, the 1% Al2O3/water was found to be the best performing nanofluid at Re = 500 in the dimpled channel flow
