    Current Status and Recent Achievements of the Sterile Insect Technique Program Against Dengue Vector, Aedes aegypti, in Indonesia

    Given the facts that Indonesia has suitable environment condition to survival, reproduction and development of the principal mosquito vector for dengue virus, Aedes aegypti, the transmission of dengue virus does routinely occur every year. The sterile insect technique (SIT) as one of the techniques in area-wide integrated pest management (AW-IPM) has been developed to control Aedes aegypti population since 2004. The first 5 years of the program (2004-2009) was focused on mass rearing facility establishment. The lab-work study for the entomological data and field trials were performed in 2010-2014. In 2014-2019, the program was addressed to enhance the mass rearing facility to increase sterile male production and strengthening the networking throughout the involvement in the regional and interregional program coordinated by International Atomic Energy Agency (IAEA). The main future challenge of the SIT program against Aedes aegypti is developing the capacity building and strong commitment of all stakeholders i.e. the Ministry of Health, the universities, the research institutes and the public. In the future work plan, the pilot project design to proof the concept of the SIT is strongly recommended to obtain a firm, unbiased and strong results as the consideration for the decision maker to create the Aedes control program in Indonesia

    The study aims to know the level of Information and Communication Technology (ICT) usage, the level of Extension Workers’ performance, the relation between the use of ICT and the performance of Extension Workers, and factors related to the use of ICT in Agricultural Extension Centers, Fisheries and Forestry (BP3K) Jati Agung, South Lampung District. This study was conducted at BP3K Jati Agung Regency, South Lampung District in 2017  with 17 respondents of Extension Workers. The research method used is a survey method. Methods of analysis used were descriptive analysis and Rank Spearman correlation. The results showed that Extension Workers have a high rate of ICT utilization and a high performance level.  The use of ICT has a significant relationship with the performance of Extension Workers and factors related to the use of ICT are age, education, income, and cosmopolitan characteristics. Key words: extension workers, ICT, performanc

    FUS/TLS Is a Co-Activator of Androgen Receptor in Prostate Cancer Cells

    Androgen receptor (AR) is a member of the nuclear receptor family of transcription factors. Upon binding to androgens, AR becomes transcriptionally active to regulate the expression of target genes that harbor androgen response elements (AREs) in their promoters and/or enhancers. AR is essential for the growth and survival of prostate cancer cells and is therefore a target for current and next-generation therapeutic modalities against prostate cancer. Pathophysiologically relevant protein-protein interaction networks involving AR are, however, poorly understood. In this study, we identified the protein FUsed/Translocated in LipoSarcoma (FUS/TLS) as an AR-interacting protein by co-immunoprecipitation of endogenous proteins in LNCaP human prostate cancer cells. The hormonal response of FUS expression in LNCaP cells was shown to resemble that of other AR co-activators. FUS displayed a strong intrinsic transactivation capacity in prostate cancer cells when tethered to basal promoters using the GAL4 system. Chromatin immunoprecipitation experiments showed that FUS was recruited to ARE III of the enhancer region of the PSA gene. Data from ectopic overexpression and “knock-down” approaches demonstrated that AR transcriptional activity was enhanced by FUS. Depletion of FUS reduced androgen-dependent proliferation of LNCaP cells. Thus, FUS is a novel co-activator of AR in prostate cancer cells

    Androgen receptor condensates as drug targets

    Transcription factors are among the most attractive therapeutic targets, but are considered largely undruggable. Here we provide evidence that small molecule-mediated partitioning of the androgen receptor, an oncogenic transcription factor, into phase-separated condensates has therapeutic effect in prostate cancer models. We show that the phase separation capacity of the androgen receptor is driven by aromatic residues and short unstable helices in its intrinsically disordered activation domain. Based on this knowledge, we developed tool compounds that covalently attach aromatic moieties to cysteines in the receptors’ activation domain. The compounds enhanced partitioning of the receptor into condensates, facilitated degradation of the receptor, inhibited androgen receptor-dependent transcriptional programs, and had antitumorigenic effect in models of prostate cancer and castration-resistant prostate cancer in vitro and in vivo. These results establish a generalizable framework to target the phase- separation capacity of intrinsically disordered regions in oncogenic transcription factors and other disease-associated proteins with therapeutic intent

    Pre-ART Levels of Inflammation and Coagulation Markers Are Strong Predictors of Death in a South African Cohort with Advanced HIV Disease

    BACKGROUND: Levels of high-sensitivity C-reactive protein (hsCRP), interleukin-6 (IL-6), and D-dimer predict mortality in HIV patients on antiretroviral therapy (ART) with relatively preserved CD4+ T cell counts. We hypothesized that elevated pre-ART levels of these markers among patients with advanced HIV would be associated with an increased risk of death following the initiation of ART. METHODS: Pre-ART plasma from patients with advanced HIV in South Africa was used to measure hsCRP, IL-6 and D-dimer. Using a nested case-control study design, the biomarkers were measured for 187 deaths and two controls matched on age, sex, clinical site, follow-up time and CD4+ cell counts. Odds ratios were estimated using conditional logistic regression. In addition, for a random sample of 100 patients, biomarkers were measured at baseline and 6 months following randomization to determine whether ART altered their levels. RESULTS: Median baseline biomarkers levels for cases and controls, respectively, were 11.25 vs. 3.6 mg/L for hsCRP, 1.41 vs. 0.98 mg/L for D-dimer, and 9.02 vs. 4.20 pg/mL for IL-6 (all p<0.0001). Adjusted odds ratios for the highest versus lowest quartile of baseline biomarker levels were 3.5 (95% CI: 1.9-6.7) for hsCRP, 2.6 (95%CI 1.4-4.9) for D-dimer, and 3.8 (95% CI: 1.8-7.8) for IL-6. These associations were stronger for deaths that occurred more proximal to the biomarker measurements. Levels of D-dimer and IL-6, but not hsCRP, were significantly lower at month 6 after commencing ART compared to baseline (p<0.0001). CONCLUSIONS: Among patients with advanced HIV disease, elevated pre-ART levels of hsCRP, IL-6 and D-dimer are strongly associated with early mortality after commencing ART. Elevated levels of inflammatory and coagulation biomarkers may identify patients who may benefit from aggressive clinical monitoring after commencing ART. Further investigation of strategies to reduce biomarkers of inflammation and coagulation in patients with advanced HIV disease is warranted.
