    Thermodynamic Forecasts of the Mediterranean Sea Acidification

    Anthropogenic CO2 is a major driver of the present ocean acidification. This latter is threatening the marine ecosystems and has been identified as a major environmental and economic menace. This study aims to forecast from the thermodynamic equations, the acidification variation (ΔpH) of the Mediterranean waters over the next few decades and beyond this century. In order to do so, we calculated and fitted the theoretical values based upon the initial conditions from data of the 2013 MedSeA cruise. These estimates have been performed both for the Western and for the Eastern basins based upon their respective physical (temperature and salinity) and chemical (total alkalinity and total inorganic carbon) properties. The results allow us to point out four tipping points, including one when the Mediterranean Sea waters would become acid (pH<7). In order to provide an associated time scale to the theoretical results, we used two of the IPCC (2007) atmospheric CO2 scenarios. Under the most optimistic scenario of the “Special Report: Emissions Scenarios” (SRES) of the IPCC (2007), the results indicate that in 2100, pH may decrease down to 0.245 in the Western basin and down to 0.242 in the Eastern basin (compared to the pre-industrial pH). Whereas for the most pessimistic SRES scenario of the IPCC (2007), the results for the year 2100, forecast a pH decrease down to 0.462 and 0.457, for the Western and for the Eastern basins, respectively. Acidification, which increased unprecedentedly in recent years, will rise almost similarly in both Mediterranean basins only well after the end of this century. These results further confirm that both basins may become undersaturated (< 1) with respect to calcite and aragonite (at the base of the mixed layer depth), only in the far future (in a few centuries)

    Tests of Landscape Influence: Nest Predation and Brood Parasitism in Fragmented Ecosystems

    The effects of landscape fragmentation oil nest predation and brood parasitism, the two primary Causes of avian reproductive failure, have been difficult to generalize across landscapes, yet few Studies have clearly considered the context and spatial scale of: fragmentation. Working in two river systems fragmented by agricultural and rural-housing development, we tracked nesting Success and brood parasitism in \u3e 2500 bird nests in 38 patches of deciduous riparian woodland. Patches oil both river Systems were embedded in one of two local contexts (buffered from agriculture by coniferous forest, or adjacent to agriculture), but the abundance of agriculture and human habitation within 1 km of each patch was highly variable. We examined evidence for three models of landscape effects oil nest predation based on (1) the relative importance of generalist agricultural nest predators, (2) predators associated with the natural habitats typically removed by agricultural development, or (3) an additive combination of: these two predator communities. We found strong support for all additive predation model in which landscape features affect nest predation differently at different spatial scales. Riparian habitat with forest buffers had higher nest predation rates than sites adjacent to agriculture, but nest predation also increased with increasing agriculture in the larger landscape Surrounding each site. These results suggest that predators living in remnant woodland buffers, as well as generalist nest predators associated with agriculture, affect nest predation rates, but they appear to respond at different spatial scales. Brood parasitism, in Contrast, Was unrelated to agricultural abundance oil the landscape, but showed a strong nonlinear relationship with farm and house density, indicating a critical point at which increased human habitat Causes increased brood parasitism. Accurate predictions regarding landscape effects oil nest predation and brood parasitism will require an increased appreciation of the multiple scales at which landscape components influence predator and parasite behavior

    Alternative antibody for the detection of CA19-9 antigen: a European multicenter study for the evaluation of the analytical and clinical performance of the Access (R) GI Monitor assay on the UniCel (R) Dxl 800 Immunoassay System

    Background: Gastrointestinal cancer antigen CA19-9 is known as a valuable marker for the management of patients with pancreatic cancer. Methods: The analytical and clinical performance of the Access(R) GI Monitor assay (Beckman Coulter) was evaluated on the UniCel(R) Dxl 800 Immunoassay System at five different European sites and compared with a reference method, defined as CA19-9 on the Elecsys System (Roche Diagnostics). Results: Total imprecision (%CV) of the GI Monitor ranged between 3.4% and 7.7%, and inter-laboratory reproducibility between 3.6% and 4.0%. Linearity upon dilution showed a mean recovery of 97.4% (SD+7.2%). Endogenous interferents had no influence on GI Monitor levels (mean recoveries: hemoglobin 103%, bilirubin 106%, triglycerides 106%). There was no high-dose hook effect up to 115,000 kU/L. Clinical performance investigated in sera from 1811 individuals showed a good correlation between the Access' GI Monitor and Elecsys CA19-9 (R = 0.959, slope = 1.004, intercept +0.17). GI Monitor serum levels were low in healthy individuals (n = 267, median = 6.0 kU/L, 95th percentile = 23.1 kU/L), higher in individuals with various benign diseases (n = 550, medians = 5.8-13.4 kU/L, 95th percentiles = 30.1-195.5 kU/L) and even higher in individuals suffering from various cancers (n = 995, medians = 8.4-233.8 kU/L, 95th percentiles = 53.7-13,902 kU/L). Optimal diagnostic accuracy for cancer detection against the relevant benign control group by the GI Monitor was found for pancreatic cancer {[}area under the curve (AUC) 0.83]. Results for the reference CA19-9 assay were comparable (AUC 0.85). Conclusions: The Access(R) GI Monitor provides very good methodological characteristics and demonstrates an excellent analytical and clinical correlation with the Elecsys CA19-9. The GI Monitor shows the best diagnostic accuracy in pancreatic cancer. Our results also suggest a clinical value of the GI Monitor in other cancers

    Sex-specific Gene Expression in Flupirtine-Treated Cln3Δex7/8 Mouse Brain

    Gene expression is a powerful tool to understand structure-function relationships in the nervous system. This study reports global gene expression changes induced by flupirtine in brain of male and female Cln3Δex7/8 mice, exposing potential flupirtine targets at the molecular level. Gene expression analysis of male and female Cln3Δex7/8 mouse brain was determined following oral administration of flupirtine for 14 weeks, using Mouse Genome 430 2.0 array Chips and an Affymetrix platform. Fifty-six genes in males and 79 in females were differentially expressed in flupirtine- versus vehicle-treated Cln3Δex7/8 mouse brain. Flupirtine altered several pathways in Cln3Δex7/8 mouse brain: apoptosis, the complement cascade, NF-kB, and p38α MAPK signaling pathways. Gene-gene network analysis highlighted networks and processes functionally pertinent to flupirtine treatment. These encompassed neurodegeneration, neuro-inflammation, and implicated neurological disorders such as Alzheimer and Parkinson disease. Flupirtine mediates its action in males and females through distinctive actionable targets in the same pathways. This work consolidates the groundwork for considering flupirtine as a treatment option in human CLN3 disease

    Exogenous Flupirtine as Potential Treatment for CLN3 Disease

    CLN3 disease is a fatal neurodegenerative disorder affecting children. Hallmarks include brain atrophy, accelerated neuronal apoptosis, and ceramide elevation. Treatment regimens are supportive, highlighting the importance of novel, disease-modifying drugs. Flupirtine and its new allyl carbamate derivative (compound 6) confer neuroprotective effects in CLN3-deficient cells. This study lays the groundwork for investigating beneficial effects in Cln3Δex7/8 mice. WT/Cln3Δex7/8 mice received flupirtine/compound 6/vehicle for 14 weeks. Short-term effect of flupirtine or compound 6 was tested using a battery of behavioral testing. For flupirtine, gene expression profiles, astrogliosis, and neuronal cell counts were determined. Flupirtine improved neurobehavioral parameters in open field, pole climbing, and Morris water maze tests in Cln3Δex7/8 mice. Several anti-apoptotic markers and ceramide synthesis/degradation enzymes expression was dysregulated in Cln3Δex7/8 mice. Flupirtine reduced astrogliosis in hippocampus and motor cortex of male and female Cln3Δex7/8 mice. Flupirtine increased neuronal cell counts in male mice. The newly synthesized compound 6 showed promising results in open field and pole climbing. In conclusion, flupirtine improved behavioral, neuropathological and biochemical parameters in Cln3Δex7/8 mice, paving the way for potential therapies for CLN3 disease

    Prognostic and therapeutic significance of carbohydrate antigen 19-9 as tumor marker in patients with pancreatic cancer

    In pancreatic cancer ( PC) accurate determination of treatment response by imaging often remains difficult. Various efforts have been undertaken to investigate new factors which may serve as more appropriate surrogate parameters of treatment efficacy. This review focuses on the role of carbohydrate antigen 19- 9 ( CA 19- 9) as a prognostic tumor marker in PC and summarizes its contribution to monitoring treatment efficacy. We undertook a Medline/ PubMed literature search to identify relevant trials that had analyzed the prognostic impact of CA 19- 9 in patients treated with surgery, chemoradiotherapy and chemotherapy for PC. Additionally, relevant abstract publications from scientific meetings were included. In advanced PC, pretreatment CA 19- 9 levels have a prognostic impact regarding overall survival. Also a CA 19- 9 decline under chemotherapy can provide prognostic information for median survival. A 20% reduction of CA 19- 9 baseline levels within the first 8 weeks of chemotherapy appears to be sufficient to define a prognostic relevant subgroup of patients ('CA 19- 9 responder'). It still remains to be defined whether the CA 19- 9 response is a more reliable method for evaluating treatment efficacy compared to conventional imaging. Copyright (c) 2006 S. Karger AG, Basel

    Low-threshold analysis of CDMS shallow-site data

    Data taken during the final shallow-site run of the first tower of the Cryogenic Dark Matter Search (CDMS II) detectors have been reanalyzed with improved sensitivity to small energy depositions. Four ~224 g germanium and two ~105 g silicon detectors were operated at the Stanford Underground Facility (SUF) between December 2001 and June 2002, yielding 118 live days of raw exposure. Three of the germanium and both silicon detectors were analyzed with a new low-threshold technique, making it possible to lower the germanium and silicon analysis thresholds down to the actual trigger thresholds of ~1 keV and ~2 keV, respectively. Limits on the spin-independent cross section for weakly interacting massive particles (WIMPs) to elastically scatter from nuclei based on these data exclude interesting parameter space for WIMPs with masses below 9 GeV/c^2. Under standard halo assumptions, these data partially exclude parameter space favored by interpretations of the DAMA/LIBRA and CoGeNT experiments' data as WIMP signals, and exclude new parameter space for WIMP masses between 3 GeV/c^2 and 4 GeV/c^2.Comment: 18 pages, 12 figures, 5 table

    Search for inelastic dark matter with the CDMS II experiment

    Results are presented from a reanalysis of the entire five-tower data set acquired with the Cryogenic Dark Matter Search (CDMS II) experiment at the Soudan Underground Laboratory, with an exposure of 969 kg-days. The analysis window was extended to a recoil energy of 150 keV, and an improved surface-event background-rejection cut was defined to increase the sensitivity of the experiment to the inelastic dark matter (iDM) model. Three dark matter candidates were found between 25 keV and 150 keV. The probability to observe three or more background events in this energy range is 11%. Because of the occurrence of these events the constraints on the iDM parameter space are slightly less stringent than those from our previous analysis, which used an energy window of 10-100 keV.Comment: 10 pages, 10 figures, minor changes to match published version, conclusion unchange

To the best of our knowledge, this is the first case report on Takayasu's arteritis and beta thalassemia presenting together.</p> <p>Case presentation</p> <p>This is a case report of a 23-year-old Asian woman of Pakistani descent who presented with a headache, blurred vision and dizziness.</p> <p>Conclusion</p> <p>The correct diagnosis of our patient was based on clinical suspicion, appropriate imaging studies, and deliberation of the differential diagnosis. The management of our patient depended on the correct diagnosis of both the diseases.</p
