    Conformity and controversies in the diagnosis, staging and follow-up evaluation of canine nodal lymphoma: a systematic review of the last 15 years of published literature

    Diagnostic methods used in the initial and post-treatment evaluation of canine lymphoma are heterogeneous and can vary within countries and institutions. Accurate reporting of clinical stage and response assessment is crucial in determining the treatment efficacy and predicting prognosis. This study comprises a systematic review of all available canine multicentric lymphoma studies published over 15 years. Data concerning diagnosis, clinical stage evaluation and response assessment procedures were extracted and compared. Sixty-three studies met the eligibility criteria. Fifty-five (87.3%) studies were non-randomized prospective or retrospective studies. The survey results also expose variations in diagnostic criteria and treatment response assessment in canine multicentric lymphoma. Variations in staging procedures performed and recorded led to an unquantifiable heterogeneity among patients in and between studies, making it difficult to compare treatment efficacies. Awareness of this inconsistency of procedure and reporting may help in the design of future clinical trials

    Levantamento florístico de um componente arbóreo de mata ciliar do Rio Paraná, Marechal Cândido Rondon, PR

    The objective this study was to conduct a floristic survey in the tree component of riparian forest area in the left bank of the Paraná river, Marechal Cândido Rondon, district of Iguiporã, PR. The study was conducted in an area of 600 m2, in six parcels of 20m x 5m, and were used to sample trees with DAP  ≥ 10 cm. Frequency, density parameters and Shannon-Weaver index were calculated for each species. The survey resulted in 54 individuals belonging to ten species, ten genera and eight families. The most frequent and abundant species was Parapiptadenia rigida. The family with greater frequency of species was the Leguminosae-Mimosoideae. The Shannon-Weaver index found was 2.05.Objetivou-se realizar um levantamento florístico no componente arbóreo de uma área de mata ciliar na margem esquerda do rio Paraná, município de Marechal Cândido Rondon, distrito de Iguiporã, PR. O estudo foi realizado em uma área de 600 m2, em seis parcelas amostrais de 5 x 20 m, tendo-se como critério de inclusão um diâmetro à altura do peito (DAP) mínimo de 10 cm. Para cada espécie amostrada foram estimados parâmetros relativos à frequência, densidade e índice de diversidade de Shannon-Weaver. O levantamento resultou em 54 indivíduos pertencentes a dez espécies, dez gêneros e oito famílias. A espécie mais abundante e frequente foi  Parapiptadenia rigida. A família com maior frequência de espécies foi a Leguminosae-Mimosoideae. O índice de diversidade de Shannon-Weaver obtido foi de 2,05

    NOS1AP polymorphisms reduce NOS1 activity and interact with prolonged repolarization in arrhythmogenesis

    Aims  NOS1AP single-nucleotide polymorphisms (SNPs) correlate with QT prolongation and cardiac sudden death in patients affected by long QT syndrome type 1 (LQT1). NOS1AP targets NOS1 to intracellular effectors. We hypothesize that NOS1AP SNPs cause NOS1 dysfunction and this may converge with prolonged action-potential duration (APD) to facilitate arrhythmias. Here we test (i) the effects of NOS1 inhibition and their interaction with prolonged APD in a guinea pig cardiomyocyte (GP-CMs) LQT1 model; (ii) whether pluripotent stem cell-derived cardiomyocytes (hiPSC-CMs) from LQT1 patients differing for NOS1AP variants and mutation penetrance display a phenotype compatible with NOS1 deficiency. Methods and results  In GP-CMs, NOS1 was inhibited by S-Methyl-L-thiocitrulline acetate (SMTC) or Vinyl-L-NIO hydrochloride (L-VNIO); LQT1 was mimicked by IKs blockade (JNJ303) and β-adrenergic stimulation (isoproterenol). hiPSC-CMs were obtained from symptomatic (S) and asymptomatic (AS) KCNQ1-A341V carriers, harbouring the minor and major alleles of NOS1AP SNPs (rs16847548 and rs4657139), respectively. In GP-CMs, NOS1 inhibition prolonged APD, enhanced ICaL and INaL, slowed Ca2+ decay, and induced delayed afterdepolarizations. Under action-potential clamp, switching to shorter APD suppressed ‘transient inward current’ events induced by NOS1 inhibition and reduced cytosolic Ca2+. In S (vs. AS) hiPSC-CMs, APD was longer and ICaL larger; NOS1AP and NOS1 expression and co-localization were decreased. Conclusion  The minor NOS1AP alleles are associated with NOS1 loss of function. The latter likely contributes to APD prolongation in LQT1 and converges with it to perturb Ca2+ handling. This establishes a mechanistic link between NOS1AP SNPs and aggravation of the arrhythmia phenotype in prolonged repolarization syndromes
