Observation of second-harmonic generation induced by pure spin currents
Extensive efforts are currently being devoted to developing a new electronic
technology, called spintronics, where the spin of electrons is explored to
carry information. [1,2] Several techniques have been developed to generate
pure spin currents in many materials and structures. [3-10] However, there is
still no method available that can be used to directly detect pure spin
currents, which carry no net charge current and no net magnetization.
Currently, studies of pure spin currents rely on measuring the induced spin
accumulation with optical techniques [5, 11-13] or spin-valve configurations.
[14-17] However, the spin accumulation does not directly reflect the spatial
distribution or temporal dynamics of the pure spin current, and therefore
cannot monitor the pure spin current in a real-time and real-space fashion.
This imposes severe constraints on research in this field. Here we demonstrate
a second-order nonlinear optical effect of the pure spin current. We show that
such a nonlinear optical effect, which has never been explored before, can be
used for the non-invasive, non-destructive, and real-time imaging of pure spin
currents. Since this detection scheme does not rely on optical resonances, it
can be generally applied in a wide range of materials with different electronic
bandstructures. Furthermore, the control of nonlinear optical properties of
materials with pure spin currents may have potential applications in photonics

Graphene dispersed cellulose microfibers composite for efficient immobilization of hemoglobin and selective biosensor for detection of hydrogen peroxide

In the present work, we have investigated the electrochemical behavior and electrocatalysis of hemoglobin (Hb) immobilized on a glassy carbon electrode (GCE) modified with a graphene-cellulose microfiber (GR–CMF) composite. The GR–CMF composite was characterized by scanning electron microscopy, elemental analysis, and Raman and Fourier transform infrared spectroscopy. Well-defined electrochemical redox characteristics of Hb were observed for Hb immobilized on a GR–CMF composite modified GCE, with a formal potential of −0.306V and a peak to peak separation of approximately 67 mV. Due to the high biocompatibility of the GR–CMF composite, the electrochemical behavior of the Hb heme redox couple (FeII/FeIII) was enhanced for Hb immobilized on the GR–CMF composite when compared to Hb immobilized on pristine GR. The heterogeneous electron transfer constant (ks) was calculated as 6.17 s−1, and is higher than previously reported for Hb immobilized GR supports. The Hb immobilized GR–CMF composite modified electrode was used for the quantification of H2O2 under optimal conditions, and shows a wider linear amperometric response ranging from 0.05 to 926 M. The limit of detection of the biosensor was 0.01 M with the sensitivity of 0.49 A M−1 cm−2. The biosensor also showed high selectivity in the presence of the range of interfering compounds and exhibits good operational stability and practicality in the detection of H2O2

Coupled discrete models abound in several areas of physics. Here we provide
an extensive set of exact quasiperiodic solutions of a number of coupled
discrete models in terms of Lame polynomials of order one and two. Some of the
models discussed are (i) coupled Salerno model, (ii) coupled Ablowitz-Ladik
model, (iii) coupled saturated discrete nonlinear Schrodinger equation, (iv)
coupled phi4 model, and (v) coupled phi6 model. Furthermore, we show that most
of these coupled models in fact also possess an even broader class of exact

α-Synuclein Suppression by Targeted Small Interfering RNA in the Primate Substantia Nigra
The protein α-synuclein is involved in the pathogenesis of Parkinson's disease and other neurodegenerative disorders. Its toxic potential appears to be enhanced by increased protein expression, providing a compelling rationale for therapeutic strategies aimed at reducing neuronal α-synuclein burden. Here, feasibility and safety of α-synuclein suppression were evaluated by treating monkeys with small interfering RNA (siRNA) directed against α-synuclein. The siRNA molecule was chemically modified to prevent degradation by exo- and endonucleases and directly infused into the left substantia nigra. Results compared levels of α-synuclein mRNA and protein in the infused (left) vs. untreated (right) hemisphere and revealed a significant 40–50% suppression of α-synuclein expression. These findings could not be attributable to non-specific effects of siRNA infusion since treatment of a separate set of animals with luciferase-targeting siRNA produced no changes in α-synuclein. Infusion with α-synuclein siRNA, while lowering α-synuclein expression, had no overt adverse consequences. In particular, it did not cause tissue inflammation and did not change (i) the number and phenotype of nigral dopaminergic neurons, and (ii) the concentrations of striatal dopamine and its metabolites. The data represent the first evidence of successful anti-α-synuclein intervention in the primate substantia nigra and support further development of RNA interference-based therapeutics
Methods to study splicing from high-throughput RNA Sequencing data
The development of novel high-throughput sequencing (HTS) methods for RNA
(RNA-Seq) has provided a very powerful mean to study splicing under multiple
conditions at unprecedented depth. However, the complexity of the information
to be analyzed has turned this into a challenging task. In the last few years,
a plethora of tools have been developed, allowing researchers to process
RNA-Seq data to study the expression of isoforms and splicing events, and their
relative changes under different conditions. We provide an overview of the
methods available to study splicing from short RNA-Seq data. We group the
methods according to the different questions they address: 1) Assignment of the
sequencing reads to their likely gene of origin. This is addressed by methods
that map reads to the genome and/or to the available gene annotations. 2)
Recovering the sequence of splicing events and isoforms. This is addressed by
transcript reconstruction and de novo assembly methods. 3) Quantification of
events and isoforms. Either after reconstructing transcripts or using an
annotation, many methods estimate the expression level or the relative usage of
isoforms and/or events. 4) Providing an isoform or event view of differential
splicing or expression. These include methods that compare relative
event/isoform abundance or isoform expression across two or more conditions. 5)
Visualizing splicing regulation. Various tools facilitate the visualization of
the RNA-Seq data in the context of alternative splicing. In this review, we do
not describe the specific mathematical models behind each method. Our aim is
rather to provide an overview that could serve as an entry point for users who
need to decide on a suitable tool for a specific analysis. We also attempt to
propose a classification of the tools according to the operations they do, to

The influence of methylphenidate on the power spectrum of ADHD children – an MEG study
BACKGROUND: The present study was dedicated to investigate the influence of Methylphenidate (MPH) on cortical processing of children who were diagnosed with different subtypes of Attention Deficit Hyperactivity Disorder (ADHD). As all of the previous studies investigating power differences in different frequency bands have been using EEG, mostly with a relatively small number of electrodes our aim was to obtain new aspects using high density magnetoencephalography (MEG). METHODS: 35 children (6 female, 29 male) participated in this study. Mean age was 11.7 years (± 1.92 years). 17 children were diagnosed of having an Attention-Deficit/Hyperactivity Disorder of the combined type (ADHDcom, DSM IV code 314.01); the other 18 were diagnosed for ADHD of the predominantly inattentive type (ADHDin, DSM IV code 314.0). We measured the MEG during a 5 minute resting period with a 148-channel magnetometer system (MAGNES™ 2500 WH, 4D Neuroimaging, San Diego, USA). Power values were averaged for 5 bands: Delta (D, 1.5–3.5 Hz), Theta (T, 3.5–7.5 Hz), Alpha (A, 7.5–12.5 Hz), Beta (B, 12.5–25 Hz) and Global (GL, 1.5–25 Hz).). Additionally, attention was measured behaviourally using the D2 test of attention with and without medication. RESULTS: The global power of the frequency band from 1.5 to 25 Hz increased with MPH. Relative Theta was found to be higher in the left hemisphere after administration of MPH than before. A positive correlation was found between D2 test improvement and MPH-induced power changes in the Theta band over the left frontal region. A linear regression was computed and confirmed that the larger the improvement in D2 test performance, the larger the increase in Theta after MPH application. CONCLUSION: Main effects induced by medication were found in frontal regions. Theta band activity increased over the left hemisphere after MPH application. This finding contradicts EEG results of several groups who found lower levels of Theta power after MPH application. As relative Theta correlates with D2 test improvement we conclude that MEG provide complementary and therefore important new insights to ADHD
Mediator of DNA Damage Checkpoint 1 (MDC1) Contributes to High NaCl-Induced Activation of the Osmoprotective Transcription Factor TonEBP/OREBP
Background: Hypertonicity, such as induced by high NaCl, increases the activity of the transcription factor TonEBP/OREBP whose target genes increase osmoprotective organic osmolytes and heat shock proteins. Methodology: We used mass spectrometry to analyze proteins that coimmunoprecipitate with TonEBP/OREBP in order to identify ones that might contribute to its high NaCl-induced activation. Principal Findings: We identified 20 unique peptides from Mediator of DNA Damage Checkpoint 1 (MDC1) with high probability. The identification was confirmed by Western analysis. We used small interfering RNA knockdown of MDC1 to characterize its osmotic function. Knocking down MDC1 reduces high NaCl-induced increases in TonEBP/OREBP transcriptional and transactivating activity, but has no significant effect on its nuclear localization. We confirm six previously known phosphorylation sites in MDC1, but do not find evidence that high NaCl increases phosphorylation of MDC1. It is suggestive that MDC1 acts as a DNA damage response protein since hypertonicity reversibly increases DNA breaks, and other DNA damage response proteins, like ATM, also associate with TonEBP/OREBP and contribute to its activation by hypertonicity. National Outbreak of Salmonella Serotype Saintpaul Infections: Importance of Texas Restaurant Investigations in Implicating Jalapeño Peppers
BACKGROUND: In May 2008, PulseNet detected a multistate outbreak of Salmonella enterica serotype Saintpaul infections. Initial investigations identified an epidemiologic association between illness and consumption of raw tomatoes, yet cases continued. In mid-June, we investigated two clusters of outbreak strain infections in Texas among patrons of Restaurant A and two establishments of Restaurant Chain B to determine the outbreak's source. METHODOLOGY/PRINCIPAL FINDINGS: We conducted independent case-control studies of Restaurant A and B patrons. Patients were matched to well controls by meal date. We conducted restaurant environmental investigations and traced the origin of implicated products. Forty-seven case-patients and 40 controls were enrolled in the Restaurant A study. Thirty case-patients and 31 controls were enrolled in the Restaurant Chain B study. In both studies, illness was independently associated with only one menu item, fresh salsa (Restaurant A: matched odds ratio [mOR], 37; 95% confidence interval [CI], 7.2-386; Restaurant B: mOR, 13; 95% CI 1.3-infinity). The only ingredient in common between the two salsas was raw jalapeño peppers. Cultures of jalapeño peppers collected from an importer that supplied Restaurant Chain B and serrano peppers and irrigation water from a Mexican farm that supplied that importer with jalapeño and serrano peppers grew the outbreak strain. CONCLUSIONS/SIGNIFICANCE: Jalapeño peppers, contaminated before arrival at the restaurants and served in uncooked fresh salsas, were the source of these infections. Our investigations, critical in understanding the broader multistate outbreak, exemplify an effective approach to investigating large foodborne outbreaks.
