Chemo- and Thermosensory Responsiveness of Grueneberg Ganglion Neurons Relies on Cyclic Guanosine Monophosphate Signaling Elements
Neurons of the Grueneberg ganglion (GG) in the anterior nasal region of mouse pups respond to cool temperatures and to a small set of odorants. While the thermosensory reactivity appears to be mediated by elements of a cyclic guanosine monophosphate (cGMP) cascade, the molecular mechanisms underlying the odor-induced responses are unclear. Since odor-responsive GG cells are endowed with elements of a cGMP pathway, specifically the transmembrane guanylyl cyclase subtype GC-G and the cyclic nucleotide-gated ion channel CNGA3, the possibility was explored whether these cGMP signaling elements may also be involved in chemosensory GG responses. Experiments with transgenic mice deficient for GC-G or CNGA3 revealed that GG responsiveness to given odorants was significantly diminished in these knockout animals. These findings suggest that a cGMP cascade may be important for both olfactory and thermosensory signaling in the GG. However, in contrast to the thermosensory reactivity, which did not decline over time, the chemosensory response underwent adaptation upon extended stimulation, suggesting that the two transduction processes only partially overlap. Experimental measurement-based quantum computing beyond the cluster-state model
The paradigm of measurement-based quantum computation opens new experimental
avenues to realize a quantum computer and deepens our understanding of quantum
physics. Measurement-based quantum computation starts from a highly entangled
universal resource state. For years, clusters states have been the only known
universal resources. Surprisingly, a novel framework namely quantum computation
in correlation space has opened new routes to implement measurement-based
quantum computation based on quantum states possessing entanglement properties
different from cluster states. Here we report an experimental demonstration of
every building block of such a model. With a four-qubit and a six-qubit state
as distinct from cluster states, we have realized a universal set of
single-qubit rotations, two-qubit entangling gates and further Deutsch's
algorithm. Besides being of fundamental interest, our experiment proves
in-principle the feasibility of universal measurement-based quantum computation
without using cluster states, which represents a new approach towards the

On-demand semiconductor single-photon source with near-unity indistinguishability
Single photon sources based on semiconductor quantum dots offer distinct
advantages for quantum information, including a scalable solid-state platform,
ultrabrightness, and interconnectivity with matter qubits. A key prerequisite
for their use in optical quantum computing and solid-state networks is a high
level of efficiency and indistinguishability. Pulsed resonance fluorescence
(RF) has been anticipated as the optimum condition for the deterministic
generation of high-quality photons with vanishing effects of dephasing. Here,
we generate pulsed RF single photons on demand from a single,
microcavity-embedded quantum dot under s-shell excitation with 3-ps laser
pulses. The pi-pulse excited RF photons have less than 0.3% background
contributions and a vanishing two-photon emission probability.
Non-postselective Hong-Ou-Mandel interference between two successively emitted
photons is observed with a visibility of 0.97(2), comparable to trapped atoms
and ions. Two single photons are further used to implement a high-fidelity

An Analysis by Synthesis Approach for Automatic Vertebral Shape Identification in Clinical QCT
Quantitative computed tomography (QCT) is a widely used tool for osteoporosis
diagnosis and monitoring. The assessment of cortical markers like cortical bone
mineral density (BMD) and thickness is a demanding task, mainly because of the
limited spatial resolution of QCT. We propose a direct model based method to
automatically identify the surface through the center of the cortex of human
vertebra. We develop a statistical bone model and analyze its probability
distribution after the imaging process. Using an as-rigid-as-possible
deformation we find the cortical surface that maximizes the likelihood of our
model given the input volume. Using the European Spine Phantom (ESP) and a high
resolution \mu CT scan of a cadaveric vertebra, we show that the proposed
method is able to accurately identify the real center of cortex ex-vivo. To
demonstrate the in-vivo applicability of our method we use manually obtained

<p>Abstract</p> <p>Introduction</p> <p>Fractures of the neck of the femoral component have been reported in uncemented total hip replacements, however, to our knowledge, no fractures of the neck of a cementless forged titanium alloy femoral stem coated in the proximal third with hydroxy-apatite have been reported in the medical literature.</p> <p>Case presentation</p> <p>This case report describes a fracture of the neck of a cementless forged titanium alloy stem coated in the proximal third with hydroxy-apatite.</p> <p>Conclusion</p> <p>The neck of the femoral stem failed from fatigue probably because of a combination of factors described analytically below.</p

Evaluation of volumetric modulated arc therapy (VMAT) with Oncentra MasterPlan® for the treatment of head and neck cancer
Background
Several comparison studies have shown the capability of VMAT to achieve similar or better plan quality as IMRT, while reducing the treatment time. The experience of VMAT in a multi vendor environment is limited. We compared the plan quality and performance of VMAT to IMRT and we investigate the effects of varying various user-selectable parameters.
Methods
IMRT, single arc VMAT and dual arc VMAT were compared for four different head-and-neck tumors. For VMAT, the effect of varying gantry angle spacing and treatment time on the plan quality was investigated. A comparison of monitor units and treatment time was performed.
Results
IMRT and dual arc VMAT achieved a similar plan quality, while single arc could not provide an acceptable plan quality. Increasing the number of control points does not improve the plan quality. Dual arc VMAT delivery time is about 30% of IMRT delivery time.
Conclusions
Dual arc VMAT is a fast and accurate technique for the treatment of head and neck cancer. It applies similar number of MUs as IMRT, but the treatment time is strongly reduced, maintaining similar or better dose conformity to the PTV and OAR sparing
NGF and proNGF Regulate Functionally Distinct mRNAs in PC12 Cells: An Early Gene Expression Profiling
The biological activities of NGF and of its precursor proNGF are quite distinct, due to different receptor binding profiles, but little is known about how proNGF regulates gene expression. Whether proNGF is a purely pro-apoptotic molecule and/or simply a “less potent NGF” is still a matter of debate. We performed experiments to address this question, by verifying whether a proNGF specific transcriptional signature, distinct from that of NGF, could be identified. To this aim, we studied gene expression regulation by proNGF and NGF in PC12 cells incubated for 1 and 4 hours with recombinant NGF and proNGF, in its wild-type or in a furin-cleavage resistant form. mRNA expression profiles were analyzed by whole genome microarrays at early time points, in order to identify specific profiles of NGF and proNGF. Clear differences between the mRNA profiles modulated by the three neurotrophin forms were identified. NGF and proNGF modulate remarkably distinct mRNA expression patterns, with the gene expression profile regulated by NGF being significantly more complex than that by proNGF, both in terms of the total number of differentially expressed mRNAs and of the gene families involved. Moreover, while the total number of genes modulated by NGF increases dramatically with time, that by proNGFs is unchanged or reduced. We identified a subset of regulated genes that could be ascribed to a “pure proNGF” signalling, distinct from the “pure NGF” one. We also conclude that the composition of mixed NGF and proNGF samples, when the two proteins coexist, influences the profile of gene expression. Based on this comparison of the gene expression profiles regulated by NGF and its proNGF precursor, we conclude that the two proteins activate largely distinct transcriptional programs and that the ratio of NGF to proNGF in vivo can profoundly influence the pattern of regulated mRNAs
