    Evaluation of Tre Gai bamboo (Bambusa spinosa) bending strength perpendicular-to-fibre along the culm length

    The continued need to decarbonize buildings has led to a deeper exploration of the use of bamboo culms in building systems. For the design and use of full culm bamboo structures, the bending strength perpendicular-to-fibre has received little comprehensive study along the length of commercially produced bamboo poles. We intensively measured the bending strength perpendicular-to-the-fibre for both node and internode sections along the length of commercially purchased Tre Gai (Bambusa spinosa) culms. Eight culms, each four meters long, were sectioned to measure the bending strength terms including tension and compression strengths in both the N-S (top/bottom) and E/W (left/right) directions using ISO 22157 procedures. Digital scanning was used to measure the inner and outer diameter of each section. Node strength was approximately 2.64 times greater than the internode strength. The performances of perpendicular-to-fibre strength terms were related to the primary tissue strength rather than the bamboo fibre strength within the culm. Based upon observations and measurements, the fmT,NS and fmC,EW strengths had greater values with lower variability, and were less sensitive to changes in length.     Mouse Paternal RNAs Initiate a Pattern of Metabolic Disorders in a Line-Dependent Manner.

    A wide range of diseases result from environmental effects, and the levels of many native transcripts are altered. The alteration of non-coding RNAs (ncRNAs) and transmission of the variation to the next generation is increasingly recognized as a marker of disease. However, the determining signals and mechanisms of RNA-induced heritability remain unclear. We performed functional tests with four different genotypes of mice maintained on a high-fat diet to trace the transfer of the obesity/diabetes phenotype to the next generation in order to detect common signals. Two founders of four mouse lines (B6/D2 hybrid and Dnmt2(-/-C57BL/6)) resist and do not change their phenotype while their sperm RNAs after microinjection into fertilized mouse eggs transfer the newly acquired phenotypes in a susceptible inbred line (C57BL/6 or Balb/c). Unexpectedly, in the same line of experiments, sperm RNA from animals raised on a normal diet when mixed with the sperm RNA from animals raised on a diet high in fat or synthetic miR-19b (inducer of obesity) affects or prevents the development of obesity and diabetes. However, it remains unclear what happens to ncRNA signaling under diet. With a comprehensive new analysis of the transcripts maintained as an RNA/DNA hybrid in sperm, we suggest that a fraction of the RNAs are stably attached to the genome. Thus, we propose that changes in the dynamics of ncRNA retention on DNA by factors such as transcriptional variations or lack of adequate methylation could serve as molecular markers to trace these epigenetics events

    First report on the molecular detection, phylogeny, virological and pathological investigations of Avibacterium paragallinarum in chickens in Turkey

    Avibacterium paragallinarum is an important pathogen affecting the respiratory tract of chickens. There is a paucity of information on the molecular characteristics and pathology of A. paragallinarum in Turkish poultry flocks. In the present study, broiler and layer flocks (n = 2) suspected of viral infections with serious respiratory signs and significant mortality were visited. Chickens showed various disease signs and necropsy lesions, including purulent nasal discharge, respiratory distress, facial edema, sticky eyes, mucoid tracheitis, hemorrhagic inflammation of the infraorbital sinuses along with fibrinous mass and conjunctivitis. Histopathological lesions included loss of cilia along with necrosis and exfoliation of the superficial mucosal epithelium of the trachea, facial cellulitis, dermatitis, fibrinous plasmatic edema and infiltration of inflammatory cells, especially heterophils. A. paragallinarum was detected in tissue samples by species-specific polymerase chain reaction (PCR). The sequencing and phylogenetic analysis of the core region of the hemagglutinin (HA) gene revealed that Turkish strains detected here belonged to serotype A (serovar A1). They were related to strains reported from India (VRDC), the United States (0083), and Japan (0221), which are representatives of serovar A1. A homology of 88-90% was found between Indian strains and the Turkish strains detected in this study. Surprisingly, only vaccine strains of infectious bronchitis virus (IBV) were detected as a co-infecting agent in all samples positive for A. paragallinarum. Our findings suggest that A. paragallinarum may be an emerging pathogen in Turkish poultry flocks, and direct PCR may facilitate rapid diagnosis of infectious coryza. These results will also help to develop control strategies for A. paragallinarum
